إنجليزي. Grapevines can be infected by five different viroids, of which Australian grapevine viroid (AGVd) is the least studied. Here we report the use of RT-PCR for the detection of this viroid from infected grapevines in Tunisia. RT-PCR results were confirmed by cDNA sequencing. This is the first report of AGVd from the Mediterranean region. The data presented indicate a wider geographical distribution of this viroid than previously reported and record its occurrence in Africa. The molecular variability of AGVd from two different cultivars, Carignan and Syrah, was studied by restriction analyses and by cDNA sequencing. Sequence variability was not clustered in any specific domain or region of the genome
اظهر المزيد [+] اقل [-]إيطالي. [La vite puo' essere infettata da cinque viroidi diversi, fra i quali il Viroide australiano della vite (AGVd) e' quello meno studiato. In questo contributo riferiamo sull'impiego della RT-PCR per l'identificazione di questo viroide nelle piante infette in Tunisia. I risultati della RT-PCR sono stati confermati dal sequenziamento del cDNA. Questa e' la prima segnalazione di AGVd nella regione mediterranea. I dati presentati mettono in evidenza una distribuzione piu' ampia di questo viroide rispetto a quanto segnalato in precedenza e fanno registrarne la presenza in Africa. Mediante analisi di restrizione e sequenziamento del cDNA e' stata studiata la variabilita' molecolare di AGVd derivante da due cultivar diverse, Carignan e Syrah. La variabilita' della sequenza non e' risultata raggruppata in alcun specifico dominio o regione del genoma]
