Properties of Cucumber Bulgarian latent virus - a new species in the genus Tombusvirus [Cucumis sativus L.]

This paper reports the properties of a tombusvirus isolated by mechanical inoculation from severely stunted and mottled cucumber plants, infected also by Watermelon mosaic virus (WMV), grown in glasshouses in the Plovdiv area (Bulgaria). After separation from WMV, this virus was characterized biologically (host range reaction), physico-chemically, molecularly and ultrastructurally. The virus invaded systemically with difficulty different cucumber cultivars, producing latent infections. It was not serologically related to 24 different viruses in the genera Tombusvirus, Carmovirus, Necrovirus and Sobemovirus, but it had physico-chemical and ultrastructural properties typical of tombusviruses. The genome, which was totally sequenced, is a single-stranded positive-sense RNA of 4,576 nucleotides. The genome structure was identical to that of tombusviruses, but the sequence homology of the protein cistrons was not higher than 41% when compared to that of all sequenced members of the genus Tombusvirus. Distinct differences were also found in the N-terminal region of the ORF 1 sequence. The cucumber virus was therefore identified as a new putative tombusvirus species provisionally named Cucumber Bulgarian latent virus (CBLV)

[Questo lavoro riferisce sulle proprieta' di un tombusvirus isolato mediante inoculazione meccanica da piante di cetriolo gravemente affette da nanismo e da maculature, infettate anche dal Virus del mosaico del cocomero (WMV), coltivate in serra nell'area di Plovdiv (Bulgaria). Dopo la separazione da WMV, questo virus e' stato caratterizzato dai punti di vista biologico (reazione concernente il tipo di ospite), fisico-chimico, molecolare e ultrastrutturale. Il virus invadeva sistematicamente con difficolta' diverse cultivar di cetriolo, producendo infezioni latenti. Non era in relazione, dal punto di vista sierologico, con 24 virus diversi dei generi Tomburvirus, Carmovirus, Necrovirus e Sobemovirus, ma possedeva proprieta' fisico-chimiche e ultrastrutturali tipiche dei tombusvirus. Il genoma, che e' stato sequenziato completamente, e' un RNA a elica semplice in senso positivo di 4.576 nucleotidi. La struttura del genoma era identica a quella dei tombusvirus, ma l'omologia della sequenza dei cistroni della proteina non era superiore al 41% rispetto a quella di tutti i membri sequenziali del genere Tombusvirus. Sono state pure riscontrate chiare differenze nella regione terminale N della sequenza ORF 1. Il virus del cetriolo e' stato quindi identificato come una nuova specie putativa di tombusvirus denominata provvisoriamente Virus latente bulgaro del cetriolo]
