Triacylglycerol lowering properties of the adenosine analog GR 79236 during 5 days of dosing large, old rats: a pilot study

We assessed the antilipolytic action of N-(1S, trans)-2-hydroxycyclopentyl adenosine (GR 79236) in orally-treated large, old rats. The rats were gavaged with either GR 79236 (1 mg/kg) or water (vehicle) and their plasma/serum biochemistry determined. GR 79236 significantly reduced plasma triacylglycerol and non-esterified fatty acid concentrations over a 5-day period (between group and within group comparison). There was no significant change in median blood glucose concentration when the GR 79236 and water-treated groups were compared. However, there was a significantly (p< 0.03) lower frequency of serum glucose values <10 mmol/l (the lowest glucose value in the water-treated group) in the GR 79236-treated group. These findings suggest that GR 79236 may be potentially useful in the treatment of insulin resistance such as seen in "syndrome X."
