Evaluation of inhibitory activity of natural plant polyphenols on Soybean lipoxygenase by UFLC-mass spectrometry

The present study was carried out to elucidate the inhibitory effects of natural plant polyphenols on Soybean lipoxygenase (Soy LOX). The IC50 values of the first two compounds were calculated as 61.27 and 95.57 μmol/L, respectively. From the anti-oxidant assays, caffeic acid and chlorogenic acid possessed the highest DPPH scavenging activities and reducing power. In this study, a method based on ultra filtration liquid chromatography–mass spectrometry (UF-LC–MS) was used to investigate the binding constant of the interaction between the six polyphenols and the Soy LOX. The UF-LC–MS results showed that these compounds can bind to Soy LOX. The highest binding rate for chlorogenic acid was observed with the binding constant value of 11.21 followed by caffeic acid, sinapic acid, ferulic acid, naringenin and chrysin. It was evident from the in vitro activity and the results of the combined use of the ultra-filtrate, that the stronger binding of these compounds to the Soy LOX led to the highest inhibition of Soy LOX. The present study revealed that these natural compounds possess an appealing therapeutic profile combining with high antioxidant and potent Soy LOX inhibition activity.
