e effects of aldehyde fixation of excised roots and subcellular fractions of Zea mays have been studied in relation to the preservation of ATP-ase and peroxidase activities. Total peroxidase was little affected by either formaldehyde or glutaraldehyde whereas ATP-ase showed considerable loss of activity, particularly with glutaraldehyde. The activity remaining after fixation was dependent on both the concentration of fixative and the pH of fixation. Subcellular fractions differed in their response to fixation with the cell wall fraction generally showing higher retention of activity than the mitochondrial or microsomal fractions.
