Genetic diversity of Lansium parasiticum (Osbeck) K. C. Sahni & Bennet accessions based on vegetative morphological characters and simple sequence repeat markers

Morphological characters and SSR (Simple Sequence Repeat) molecular marker information were combined to study the genetic diversity of Lansium domesticum Corrêa. The research aimed to study the genetic diversity of accessions of L. parasiticum (duku, langsat, kokosan) accessions based on morphological characters and SSR markers. The field research was conducted at the Bogor Botanical Gardens, Mekarsari Fruit Park, and Condet Fruit Reserve. Morphological characterization was carried out by direct observation. SSR was characterized by using the Type-it Microsatellite PCR kit protocol. Data were analyzed using Cervus 3.07, POPGENE 3.2, and PBSTAT-CL 2.1. Accessions formed four groups at a cophenetic distance of 0.45. Duku condet is in group one. Group two was identified as kokosan-langsat, while the other groups’ names could not be identified. Analysis based on SSR markers yielded two main groups at a cophenetic distance of 0.35. Almost group one consisted of accessions with local names duku and langsat, while group two consisted of accessions with local names of kokosan. Primers P1, P4, P5, and P8 are the best primers in distinguishing these two groups. The dendrogram resulted from the combined data analysis formed two groups at a cophenetic distance of 0.35. The observed genetic variability among accessions was high based on morphological and molecular characters. The combination of morphological and SSR markers can distinguish accession with the vernacular name langsat and duku but cannot distinguish accessions with the vernacular name duku since langsat and kokosan are separate groups while duku is in both groups.
