A computer system helps through the analysis of a polynucleotide context for the role it plays in the appearing of mutations

That the polynucleotide context of specified variants of the polynucleotide sequences is of profound importance as for the appearing of mutations, has become a fact by now. However, the molecular mechanisms by means of which the polynucleotide context comes to be an influence on the appearing of mutations are what still hangs in the balance. The efficacious methods of sequencing led to the fast piling-up of data on the mutation localization in the sequences. We have worked out a computer system which proved to promote analysing the features of a polynucleotyde context that influences the appearing of mutations. At present it consists of two substructures, namely The Mutational Events Data Bank and The Complex of Methods To Analyse The Correlation Between The Context Features And The Appearing Of Singular and Multiple Substitions. The Mutational Events Data Bank to a description of DNA-localized mutations, the DNA polynucleotide sequence known. The Complex of Analysing Methods comprises of a number of programmes with the following purposes: 1. Analysing various characteristics of the point mutations, including the mutation distribution as for the types of nucleotide positions, the mutational transitions etc. 2. Analysing the statistical non-randomness of the correlation between the mutation localization and various features of the context (direct repeats, complementary palindromes, tracts, consenous of various sorts etc.). 3. Constructing the consensus of "hot-spots" of mutations. On the base of the actual methods, we also study the correlation between the somatic mutations in the Ig genes and the features of the context. Some mechanisms of mutagenesis are shown to contribute to the appearing of the mutations. Firstly, a row of somatic mutations showed a statistically reliable correlation with the repeats, which is in line with the model of reparational correction of the heteroduplices forming up throughout the segments of direct repeats and complementary palindromes. Secondly, reliability was revealed for the correlation between a number of somatic mutations with the consensus RSY. Finally, a conservative signal sequence, containing the consensus TCAS, was detected at the gene conversion, the V lambda-gene of the chicken taken as a demo. Nowadays our complex forks in its development. Firstly, the number of context features (the energy of the B-form of DNA, Calladine-Dickerson's angles, polytracts, z-form or DNA and so forth) is neverstoppingly upping. Secondly, the work at a complex or methods for studying various mutational and recombinational events is in headway. On the base of merging The Mutational Events Data Bank and The Complex of Analysing Methods, we are also developping an intellectuallzed systems for analysing mutational and recombinational events. Besides, the system is supposed to include a Database, an interface with the user and a control program to run the analysis.
