The present experiment was undertaken to optimize the rooting responses in micropropagated shoots of sugarcane varieties CoS 96268 and CoS 95255. The results showed that half strength MS medium supplemented with NAA (5.0 mg/1) alongwith sucrose (50 g/1) proved to be the best combination for rooting in both the varieties. On this medium, maximum 75.4 ±7.1 % rooting in CoS 96268 and 81.6 ±7.6 % in CoS 95255 were recorded. Morphological observation of rooted shoots revealed that at higher concentration of NAA (7.0 mg/1) and sucrose (70 g/1) shoots became stunted, bore curved leaves and the roots became thin and brownish in colour whereas on medium containing NAA (5.0 mg/l) + sucrose (50 g/l) most of the roots appeared healthy, white with normal root hair.
