Genetic diversity of 34 Chinese vegetable mustard landraces was tested using 69 microsatellite loci. A total of 452 alleles with 92.7% polymorphism were detected. At each locus, the number of alleles varied from 3 to12, with an average of 6.6. On the basis of the genetic similarity coefficients, clustering analysis separated the landraces into seven clusters. 480 individuals from 16 varieties were analyzed to estimate the mean number of alleles ‘A’, the effective allelic number ‘Aₑ’, the observed heterozygosity ‘Hₒ’, and expected heterozygosity ‘Hₑ’ were 7.2, 3.9, 0.68, and 0.36, respectively. An obvious genetic deviation from Hardy–Weinberg expectation was observed both among and within landraces and a considerable genetic variation was revealed within rather than among landraces. This information will aid in the application of molecular markers to identify patterns of variation which can be used to design further breeding programs and protect Brassica juncea genetic resources.
