Genome-wide analysis of cyclophilin gene family in wheat and identification of heat stress responsive members

Cyclophilins are ubiquitous proteins and constitute a large family, with several members possessing peptidyl prolyl cis-trans isomerase activity that is essential for cis-trans isomerization of peptidyl-prolyl bond. However, information on the cyclophilin gene family in wheat is lacking. In silico analysis of the recently available wheat genome sequence from the International Wheat Genome Sequencing Consortium (IWGSC) resulted in identification of 83 different wheat cyclophilin genes that encode 85 different proteins ranging between 17 kDa to 92 kDa. While 58 TaCYPs are single domain proteins and consist of a single cyclophilin-like domain, 27 cyclophilins also contain additional domains including the F-box and PAN domains. The TaCYPs are predicted to be localized to cytosol (28), chloroplasts (22), nucleus (19) mitochondria (9), secretory system (4) and endoplasmic reticulum (3). Besides the open reading frame, the introns are also present in the 5′ and 3'untranslated regions of all but eleven cyclophilin genes. The distribution patterns of TaCYPs on the chromosomes strongly suggest that there are segmental, ectopic and tandem duplications in addition to whole genome duplications. This study also revealed that expression of cyclophilin genes in wheat is differentially modulated under heat stress, suggesting their role in thermotolerance.
