The first part of a two-part article defined terms used in discussing adipose tissue cellularity and describes in detail the most commonly used methods for measuring fat cell size and number. Two basic methods are: (1) measurement of the DNA content of the tissue and (2) direct observation and measurement of fat cells. Advantages and disadvantages of the methods are specified. Errors in measurement of the mass and distribution of body fat and differences in fat cell size between depots are factors affecting the estimation of fat cell number in practice.
