The optimum conditions and mechanism for increasing exo-polysaccharide production of Truffles melanosporum by Dioscorea saponins

Polysaccharides have multiple biomedical functions. In the present study, we explored the optimal culture conditions and mechanism to increase exo-polysaccharide (EPS) production (DSEAEPS) of Truffles melanosporum by Dioscorea saponins (DS). DS was screened by comparing effect of herbs on EPS production of T. melanosporum, and DSEAEPS was assessed by two-dimensional gel electrophoresis. Effects of culture conditions and synergetic action between DS and medium components on DSEAEPS were studied by single-factor test and orthogonal design, respectively. The optimal medium contained (g/L): sucrose 30, frucose 40, yeast extract 7, corn flour 11, KNO3 5, peptone 9, KH2PO4 1.8, MnSO4 2.4, and DS 5. The maximal EPS production of 9.853 g/L was achieved under the optimal conditions as follows: inoculum ratio 7 mL/100 mL medium, culture temperature 20 °C and rotary speed 139 rpm. Collectively, our current investigation provided high EPS production of T. melanosporum by submerged culture for its potential application in food industry as well as in pharmaceutical and cosmetic manufacturing.
