Comparative efficacy of certain tests in the diagnosis of experimental sub clinical paratuberculosis in goats

The present experiment was carried out to assess the efficacy of certain diagnostic tests in experimentally produced sub clinical caprine paratuberculosis. A total of 12 goats of 8–12 weeks old were infected with 4.23 × 10⁹M. a. paratuberculosis on 8 occasions. Seven goats were kept in-contact with infected group and 4 goats as uninfected control. Six out of 12 infected goats showed gross and histological lesions, and were positive in different diagnostic tests. Characteristic lesions of paratuberculosis were found from 6 months onwards. Intradermal johnin test detected 6 infected goats, out of which 3 goats were add-fast positive and had histological lesions of paratuberculosis. Agar gel immuno-diffusion (AGID) test detected 2 infected goats positive at 12 MPI. Absorbed ELISA detected 5 goats positive, among which two goats were acid-fast bacilli positive. Faecal smear was positive in 2 infected goats, whereas faecal culture was positive in 3 goats. Similarly tissue smear and tissue PCR detected two infected goats positive. One infected goat was positive in bacterial culture of tissue. It was concluded that johnin test was most sensitive followed by absorbed ELISA.
