Vers une meilleure compréhension de l'axe hypothalamo-hypophyso-thyroïdien des bovins : épreuve technique et physiologique d'un kit ELISA pour le dosage de la b-TSH | Towards a better understanding of the hypothalamic-pituitary-thyroid axis of cattle: technical and physiological testing of an ELISA kit for the determination of b-TSH

إنجليزي. Thyroid metabolism plays an important role in the health and production of cattle. In rural medicine, this metabolism remains poorly understood compared to other animal species and diagnostic tools to assess it are insufficient. This study tested an ELISA kit for the determination of b-TSH (bovine thyrotropin) via a TRH (thyrotropin releasing hormone) stimulation test and a b-TSH inhibition test performed on 4 Holstein calves less than one month old. Following these tests, experimentally induced hyperthyroidism is described by comparing clinical examinations, blood parameters and thyroid ultrasound of the four calves before and after induction. The ELISA kit has high intra-assay coefficients (>10%) and does not respond consistently to the inhibition test (differences between extreme values: p>0.1). Nevertheless, this kit is sensitive to the stimulation test and b-TSH concentrations significantly different from the basal value are found at T+5, T+10 and T+100 minutes (p<0.05) after intravenous TRH injection. Calves showed symptoms of hyperthyroidism after the inhibition test (lacrimation, hyperthermia, tachycardia, exophthalmia). No significant differences were observed between ante-mortem and post-mortem calculated thyroid ultrasoud volumes or between measurements before and after induction of hyperthyroidism. Further studies are needed to determine whether this ELISA kit can be used routinely. Thyroid ultrasound, on the other hand, is a promising way to measure the thyroid gland in bovine medicine.

فرنسي. Le métabolisme thyroïdien joue un rôle important dans la santé et la production des bovins. En médecine rurale, ce métabolisme reste méconnu en comparaison avec d’autres espèces animales et les outils de diagnostic pour l’évaluer sont insuffisants. Cette étude met à l’épreuve un kit ELISA de dosage de la b-TSH (thyrotropine bovine) via un test de stimulation à la TRH (thyrotropin releasing hormone) et un test de freination de la b-TSH effectués sur 4 veaux Holstein pie-noires âgés de moins d’un mois. Suite à ces tests, l’hyperthyroïdie induite expérimentalement est décrite en comparant les examens cliniques, les paramètres sanguins et les échographies thyroïdiennes des quatre veaux avant et après l’induction. Le kit ELISA présente des coefficients intra-essais élevés (>10%) et ne répond pas de manière uniforme au test de freination (différences entre les valeurs extrêmes : p>0,1). Néanmoins, ce kit est sensible au test de stimulation et des concentrations en b-TSH significativement différentes de la valeur basale sont constatées à T+5, T+10 et T+100 minutes (p<0,05) après injection intraveineuse de TRH. Les veaux ont présenté des symptômes d’hyperthyroïdie après le test de freination (larmoiement, hyperthermie, tachycardie, exophtalmie). Aucune différence significative n’a été observée entre les volumes calculés à l’échographie thyroïdienne en ante-mortem et mesurés en post-mortem, ni entre les mesures avant et après l’induction de l’hyperthyroïdie. Des études supplémentaires sont nécessaires afin de déterminer si ce kit ELISA peut être utilisé en routine. L’échographie thyroïdienne, quant à elle, est un moyen prometteur de mesurer la glande thyroïde en médecine bovine.
