Elucidating the Genetics of Fruit Quality Traits in a Pepper Core Collection

The Horizon 2020 G2PSOL project aimed at characterizing the genetic diversity in genebank accessions of 4 Solanaceous crop species, tomato, pepper, eggplant and potato. Based on a low density GBS analysis of 10.301 pepper genebank accessions, a core collection of 400 C. annuum accessions, representing the genetic variation in this Capsicum species, was selected for high density genotypic analysis and broad phenotypic characterisation in trials at different locations (McLeod et al, 2023). Fruit metabolite content was determined in two trials, conducted in France and Italy, and included specialized metabolites, such as flavonoids, carotenoids, casaicinoids and flavour-related volatiles. GWAS analysis revealed the presence of a strong major QTL for flavonoids on chromosome 5. The most significant SNP was located in the CaMYB12-like gene, which was previously demonstrated as the causal gene underlying flavonoid variation in mapping populations segregating for flavonoid content, by VIGS analysis and by complementation studies in tomato (Wu et al. 2023). In addition, several other mQTLs were identified for carotenoids, capsaicinoids and flavour volatiles. The pepper core collection was also characterized for variation in postharvest water loss, an important determinant of shelf life in pepper fruit. GWAS analysis revealed several significant QTLs associated with water loss characteristics and these are currently being validated in segregating populations.
