Biocontrol con Trichoderma spp. de Fusarium oxysporum causal del “mal de almácigos” en pre y post emergencia en cebolla

En Argentina, la “podredumbre basal” y el “mal de almácigos” causados por el complejo Fusarium spp. constituyen uno de los problemas más serios en cebolla. En este trabajo se estudió el uso de especies del género Trichoderma como agentes de control biológico (ACB) de F. oxysporum, causal del “mal de almácigos” en cebolla. Se identificaron y caracterizaron morfológica, cultural y fisiológicamente a 10 aislamientos de Trichoderma spp. del cepario de Fitopatología. Por secuenciación del gen TEF-1α se identificaron las especies de Trichoderma harzianum s.l. (7), T. atroviride (1), T. koningiopsis (1) y T. longibrachiatum (1). Los aislamientos nativos se compararon con una cepa de un biofungicida comercial (PHCTM T22, Root Shield/Plant Shield) en su capacidad antagonista de aislamientos de F. oxysporum. Se estudió in vitro el potencial antagónico en cultivos duales y enfrentados. Además, se determinó la velocidad de crecimiento en placa y la tolerancia a metabolitos volátiles producidos por Brassica oleracea. Aislamientos nativos seleccionados de Trichoderma y el comercial se evaluaron en ensayos de eficacia de biocontrol in vivo: (i) en almácigo, aplicados sobre la semilla y (ii) en el trasplante, aplicados en raíces de plantines. Trichoderma koningiopsis (T13) y T. atroviride inhibieron en 50% al patógeno en cultivos duales. Sólo el crecimiento de Trichoderma sp. (T14) y del aislamiento comercial no fue afectado por los volátiles producidos por Brassica oleracea. En el bioensayo de almácigo, el tratamiento de T. koningiopsis (T14) sobre las semillas y como riego en suelo, consiguió el valor más alto de germinación de plántulas de cebolla (61%) y la mayor colonización de las raíces de cebolla. Este mismo tratamiento aplicado en las raíces sobre plantines de 120 días, produjo menor reducción de raíces afectadas de cebolla que con el aislamiento comercial.

In Argentina, the "basal rot” and the "damping off" caused by the complex Fusarium spp., are one of the most serious problems in onion. In this work, we studied the use of species of the genus Trichoderma as agents of biological control (ACB) against Fusarium oxysporum, causal "damping off" in onion. Were identified and characterized morphological, cultural and physiologically to 10 native isolates of Trichoderma spp. of the Collection of Phytopatology. Identification of species was performed by sequencing of the TEF-1α gene: T. harzianum s.l. (7), T. atroviride (1), T. koningiopsis (1) and T. longibrachiatum (1). The native isolates were compared with an isolate of a commercial biofungicide (PHCTM T22, Root Shield/Plant Shield) in its antagonistic ability “in vitro” against isolates of F. oxysporum in dual and opposite cultures. In addition, was determined the speed of growth in plate and tolerance to volatile metabolites produced by Brassica oleracea. Commercial and selected native isolates of Trichoderma were evaluated in efficacy of biocontrol in vivo: (i) in nursery, applied on the seed, and (ii) applied in roots of seedlings. Trichoderma koningiopsis (T13) and T. atroviride inhibited the growth of F. oxysporum at the rate of 50% in dual cultures. Only the growth of Trichoderma sp. (T14) and of the commercial isolate not was affected by volatiles produced by Brassica oleracea. In the bioassay, the treatment of T. koningiopsis (T14) on seeds and irrigation into soil got the highest value of germination of onion seedlings (61%) and the greater colonization of roots. This same treatment applied to the roots on seedlings of 120 days, resulted in lesser reduction of affected roots of onion that the treatment by commercial isolate.
