Comparison of ELISA [enzyme-linked immunosorbent assay] and RPT-PCR [Reverse transcription polymerase chain reaction] assays for the detection of banana bract mosaic virus causing the banana bract mosaic disease in the Philippines

Banana bract mosaic virus (BBrMV), a member of the genus Potyvirus, family Potyviridae, is the causal agent of banana bract mosaic disease (BBRMD). The disease is considered to be a serious threat to the Philippines' banana industry, where it was recorded to cause about 40% yield reduction. Diagnosis of Banana bract mosaic virus in banana can be accomplished by either enzyme linked immunosorbent assay (ELISA) or reverse transcription polymerase chain reaction (RT-PCR); however, there is currently no existing comparison regarding the success of the two detection assays in evaluating samples from field-grown plants. Autors collected banana leaf samples from field grown Musa balbisiana with varying levels of disease severity and tested these samples by both double antibody sandwich (DAS)- ELISA and RT-PCR assays for the presence of BBrMV. Of the 45 samples analyzed seven tested positive for the presence of the virus using ELISA, while 17 tested positive using RT-PCR. These results suggest that ELISA is able to detect the virus from field collected samples, but molecular detection via RT-PCR presents a highly specific and sensitive approach for the detection of BBrMV. The fast and cost efficient ELISA combined with RT-PCR can be a practical approach for future epidemiological studies of BBrMV.
