Envolvimento de enzimas NADP-desidrogenases na tolerância ao alumínio em plântulas de soja | Involvement of the NADP-dehydrogenases enzymes in the aluminum tolerance in soybean seedlings.

Um dos maiores limitantes das culturas são os solos ácidos com prevalência de íons biodisponíveis de alumínio (Al 3+ ), o qual causa danos ao crescimento e desenvolvimento das raízes. Glicose-6-fosfato desidrogenase (G6PDH), 6-fosfogluconato desidrogenase (6PGDH), malato desidrogenase (NADP-ME) e isocitrato desidrogenase (ICDH) são enzimas NADP- desidrogenases responsáveis pela produção de nicotinamida adenina dinucleotídeo fosfato (NADPH), molécula essencial para a modulação do status redox celular. Adicionalmente, o aumento na atividade dessas enzimas está relacionado a respostas a agentes estressores. A molécula de NADPH é utilizada pelo Ciclo Ascorbato-Glutationa (Ciclo AsA-GSH) na destoxificação celular do H 2 O 2 . No presente trabalho, avaliamos o desempenho de dois genótipos de soja NEO610 (Al-tolerante) e M5917I (Al-sensível) na ausência (0 mM) e presença do alumínio (1,5 mM) na fase de desenvolvimento VE/VC analisando seu sistema radicular. Neste estudo avaliamos a atividade e expressão das enzimas NADP-desidrogenases, produção de NADPH, atividade das enzimas do Ciclo Asc-GSH e seus metabólitos. Encontramos no genótipo de soja NEO610 (Al-tolerante) aumento na expressão e na atividade das enzimas NADP-desidrogenases, G6PDH, 6PGDH, ICHD, NADP-ME, bem como aumento no conteúdo de NADPH. O genótipo Al-tolerante aumentou a atividade da enzima MDHAR e manteve elevado a atividade da enzima GR, sendo acompanhado do aumento nos níveis de seus respectivos metabólitos antioxidantes, AsA e GSH. Além disso, o genótipo Al- tolerante manteve menores valores da peroxidação lipídica e do teor de H 2 O 2 e não apresentou redução no crescimento radicular frente ao estresse por Al. Dessa forma, concluímos que o aumento na expressão de genes e na atividade das enzimas NADP-desidrogenases contribuiu para a tolerância do genótipo de soja NEO610 ao Al, através do fornecimento de equivalentes redutores na forma de NAD(P)H para os componentes do Ciclo Ascorbato-Glutationa, resultando em menores danos oxidativos e melhor desenvolvimento desse genótipo frente a esse metal. One of the biggest limiting factors for crops are acidic soils with a prevalence of bioavailable aluminum ions (Al 3+ ), which damages the growth and development of roots. Glucose-6- phosphate dehydrogenase (G6PDH), 6-phosphogluconate dehydrogenase (6PGDH), malate dehydrogenase (NADP-ME) and isocitrate dehydrogenase (ICDH) are NADP-dehydrogenase enzymes responsible for the production of nicotinamide adenine dinucleotide phosphate (NADPH), an essential molecule for modulating cellular redox status. Additionally, the increase in the activity of these enzymes is related to responses to stressors. The NADPH molecule is used by the Ascorbate-Glutathione Cycle (AsA-GSH Cycle) in the cellular detoxification of H2O2. In the present work, we evaluated the performance of two soybean genotypes NEO610 (Al-tolerant) and M5917I (Al-sensitive) in the absence (0 mM) and presence of aluminum (1.5 mM) in the VE/VC development phase, analyzing their root system. In this study we evaluated the activity and expression of NADP-dehydrogenase enzymes, NADPH production, activity of Asc-GSH Cycle enzymes and their metabolites. We found in the soybean genotype NEO610 (Al-tolerant) an increase in the expression and activity of the enzymes NADP-dehydrogenases, G6PDH, 6PGDH, ICHD, NADP-ME, as well as an increase in the content of NADPH. The Al-tolerant genotype increased the activity of the MDHAR enzyme and maintained the elevated activity of the GR enzyme, being accompanied by an increase in the levels of their respective antioxidant metabolites, AsA and GSH. In addition, the Al-tolerant genotype maintained lower values of lipid peroxidation and H 2 O 2 content and showed no reduction in root growth in the face of Al stress. Thus, we conclude that the increase in the expression and activity of NADP-dehydrogenase enzymes contributed to the tolerance of the soybean NEO610 genotype to Al, through the supply of reducing equivalents in the form of NAD(P)H for the components of the Ascorbate- Glutathione, resulting in less oxidative damage and better development of this genotype against this metal.
