Dissomia uniparental em híbrido interespecífico de Passiflora edulis x Passiflora mucronata, seu potencial como porta-enxerto e resistência a fusariose | Uniparental dissymia in interspecific hybrid of Passiflora edulis x Passiflora mucronate, its potential as rootstock and resistance to fusariosis

O Brasil é o maior produtor de maracujá do mundo, sendo seus pomares formados principalmente por sementes, o que propicia grande variabilidade genética e desuniformidade. Visando maior uniformidade, assim como a manutenção do material e uso de espécies silvestres e híbridos como fonte de resistência e/ou rusticidade, o uso de propagação vegetativa como estaquia e enxertia pode ser constantemente visto na literatura em diferentes estudos. As espécies de Passiflora e seus híbridos são atraentes por suas características exóticas, adaptabilidade natural, potencial de resistência de algumas espécies silvestres e variabilidade genética inter e intraespecífica; existindo espécies de Passiflora que se demonstram resistentes a doenças que ocorrem no maracujazeiro tornando-se necessário estudos sobre o uso da enxertia nessas espécies para o uso de tais características. Assim, este trabalho teve como objetivo: realizar a confirmação de híbridos interespecíficos de Passiflora edulis x Passiflora mucronata; selecionar os genótipos de maracujazeiro quanto à resistência a Fusariose; avaliar a influência do número de folhas e da aplicação de AIB (Ácido Indolbutírico) na estaquia caulinar dos híbridos; e analisar o potencial uso do maracujazeiro e do híbrido interespecífico como porta- enxerto. O presente trabalho visou avaliar as progênies oriundas de cruzamentos interespecíficos entre P. edulis x P. mucronata, bem como seu potencial uso como porta- enxerto resistente a Fusariose. O trabalho é constituído de quatro capítulos, com o primeiro abordando a caracterização morfológica, mensuração do conteúdo de DNA nuclear, confirmação da hibridação por meio de marcadores microssatélites (SSR) e confirmação da alopoliploidia por meio da hibridização genômica in situ (GISH). O segundo capítulo abrange a avaliação quanto à resistência a Fusariose. O terceiro capítulo trata dos testes de enraizamento de estacas caulinares dos híbridos com estacas com uma e duas folhas reduzidas à metade, ambas submetidas à aplicação de diferentes concentrações de ácido indolbutírico (0, 500, 1000, 1500 e 2000 mg L -1 AIB). E o quarto capítulo versa sobre a viabilidade da enxertia e o desenvolvimento inicial de maracujazeiro-azedo (P. edulis) enxertado sobre espécie silvestre P. mucronata, híbridos e P. edulis UFV-M7. Os resultados obtidos demonstram que os cromossomos dos possíveis híbridos apresentam apenas material paterno. O que é confirmado por meio dos marcadores SSR. Por meio destes, é possível inferir que não houve duplicação do material genético dos pais, pois em alguns primers os híbridos se apresentam em heterozigose. As plantas avaliadas não sofreram alterações em sua ploidia, segundo a análise do conteúdo de DNA. Por fim, morfologicamente, os híbridos apresentam as mesmas características dos genitores paternos (P. mucronata) e apresentam esterilidade. Apresentando dissomia uniparental com eliminação seletiva dos cromossomos maternos. As plantas inoculadas com F. oxysporum f. sp. passiflorae apresentaram um mecanismo de defesa dos genótipos P. mucronata e M7-UFV, por meio da formação de géis e novas raízes adventícias que podem auxiliar em sua sobrevivência. Além disso, constatou-se que os possíveis híbridos foram suscetíveis, fato esse dado pela baixa produção de raízes das estacas. Os resultados obtidos pelas inoculações de F. solani não permitiram identificar fatores de resistência ou suscetibilidade, pois os tratamentos inoculados não diferiram das testemunhas. O uso de AIB foi significativo apenas nas variáveis número de raízes e percentagem de estacas vivas sem raiz. As estacas com uma folha e com a aplicação de AIB na concentração de 2000 mg L -1 promoveram a maior percentagem de enraizamento. Assim, nas condições estudadas, o uso de AIB foi indispensável para produção de estacas. A percentagem de pegamento da enxertia foi de 100% para as combinações P. edulis/UFV-M7 e P. edulis/BRS Sol do Cerrado, já as combinações P. edulis/P. mucronata e P. edulis/Híbrido apresentaram menor pegamento. O maior diâmetro do enxerto foi apresentado por P. edulis/UFV-M7 e P. edulis/BRS Sol do Cerrado, indicando maior compatibilidade de enxertia dentro da mesma espécie.

Brazil is the largest producer of passion fruit in the world, with its orchards mainly composed of seeds, which provides great genetic variability and unevenness. Aiming for greater uniformity, as well as the maintenance of the material and use of wild and hybrid species as a source of resistance and/or rusticity, the use of vegetative propagation such as cutting and grafting can be constantly seen in the literature in different studies. Passiflora species and their hybrids are attractive because of their exotic characteristics, natural adaptability, resistance potential of some wild species and inter and intraspecific genetic variability; there are Passiflora species that are resistant to diseases that occur in passion fruit, making it necessary to study the use of grafting in these species for the use of such characteristics. Thus, this work had as objective: to carry out the confirmation of interspecific hybrids of Passiflora edulis x Passiflora mucronata; to select the passion fruit genotypes for resistance to Fusarium; to evaluate the influence of the number of leaves and the application of IBA (Indolbutyric Acid) on the stem cutting of the hybrids; and to analyze the potential use of passion fruit and interspecific hybrid as rootstock. The present work aimed to evaluate the progenies from interspecific crosses between P. edulis and P. mucronata, as well as their potential use as rootstock resistant to Fusariosis. The work consists of four chapters, the first one dealing with morphological characterization, measurement of nuclear DNA content, confirmation of hybridization by microsatellite markers (SSR) and confirmation of allopolyploidy by genomic in situ hybridization (GISH). The second chapter covers the evaluation of Fusarium resistance. The third chapter deals with the tests of rooting stem cuttings of the hybrids with cuttings with one and two leaves reduced by half, both submitted to the application of different concentrations of indolebutyric acid (0, 500, 1000, 1500 and 2000 mg L-1 AIB). And the fourth chapter deals with the viability of grafting and the initial development of passion fruit sour (P. edulis) grafted on wild species P. mucronata, hybrids and P. edulis UFV-M7. The results obtained show that the chromosomes of the possible hybrids present only paternal material. This is confirmed through the SSR markers. By means of these, it is possible to infer that there was no duplication of the genetic material of the parents, because in some primers hybrids present in heterozygosis. The evaluated plants did not undergo changes in their ploidy, according to the analysis of the DNA content. Finally, morphologically, the hybrids have the same characteristics of the paternal parents (P. mucronata) and present sterility. Presenting uniparental disomy with selective elimination of the maternal chromosomes. Plants inoculated with F. oxysporum f. sp. passiflorae presented a mechanism of defense of the genotypes P. mucronata and M7-UFV, through the formation of gels and new adventitious roots that can aid in its survival. In addition, it was verified that the possible hybrids were susceptible, fact given by the low production of roots of the cuttings. The results obtained by the inoculations of F. solani did not allow the identification of resistance or susceptibility factors, since the treatments inoculated did not differ from the controls. The use of AIB was significant only in the number of roots and percentage of live cuttings without roots. The cuttings with a leaf and with the application of AIB in the concentration of 2000 mg L-1 promoted the greater percentage of rooting. Thus, under the conditions studied, the use of AIB was indispensable for the production of cuttings. The percentage of grafting glue was 100% for the combinations P. edulis/UFV-M7 and P. edulis/BRS Sol do Cerrado, already the combinations P. edulis/P. mucronata and P. edulis/Hybrid presented smaller glue. The largest diameter of the graft was presented by P. edulis/UFV-M7 and P. edulis/BRS Sol do Cerrado, indicating greater compatibility of grafting within the same species.
