Bolusgabe von Weißem und Grünem Tee bei Gesunden : Plasmakinetik von Flavan-3-olen und Einfluss auf oxidative Biomarker

<strong>Einleitung:</strong> Es wird angenommen, dass der Konsum von Weißem Tee und Grünem Tee aufgrund des hohen Gehalts an antioxidativ wirkenden Flavan-3-olen das pro-/antioxidative Gleichgewicht beeinflusst. Somit gilt der Anstieg der Plasmaspiegel an Flavan-3-olen als Voraussetzung für die gesundheitsfördernden Effekte unfermentierter Tees. Ziel dieser offenen, randomisierten, kontrollierten Studie war es, den Einfluss einer Bolusgabe von Weißem Tee und Grünem Tee auf die Plasmakinetik von Flavan 3 olen (Catechin, Epicatechin, Epigallocatechingallat, Epicatechingallat) bei gesunden Nichtrauchern im Vergleich zu einer Kontrollgruppe (Mineralwasser) zu untersuchen. Zusätzlich wurde die Wirkung der Bolusgabe von 600 mL dieser Getränke auf die antioxidative Kapazität im Plasma sowie oxidative Biomarker untersucht.<br /><strong>Methoden:</strong> 68 gesunde Nichtraucher (21 ♂, 47 ♀; 24±4 Jahre; BMI 22,0±2,2 kg/m²) erhielten nach polyphenolarmer Kost am Vortag morgens nüchtern 600 mL Weißen Tee (EC: 28 µmol/L; ECGC: 46 µmol/L; ECG: 11 µmol/L), Grünen Tee (EC: 88 µmol/L; EGCG: 201 µmol/L; ECG: 25 µmol/L) oder Wasser. Blutentnahmen erfolgten vor sowie 1, 2, 3, 5 und 8 h nach Konsum für die Plasmakinetik und vor sowie 2 und 5 h nach Konsum für die weiteren Parameter. Die Konzentration von C, EC, ECG und EGCG wurde mittels HPLC-EC bei 0,6 V im Plasma als Summe von freien Flavan-3-olen und glucuronidierten sowie sulfatierten Metaboliten bestimmt und daraus AUC_0-8h, Cmax, Tmax und T_1/2 ermittelt. Der Gesamtphenolgehalt sowie die Harnsäure wurden photometrisch und Vitamin C mittels HPLC bestimmt. Im Plasma wurden weiterhin die antioxidative Kapazität als TEAC und 8 Isoprostaglandin F_2α mittels ELISA erfasst. DNA-Einzelstrangbrüche in Leukozyten wurden <em>in vivo </em>und<em> ex vivo </em>(H₂O₂ Behandlung) mit Comet Assay gemessen. Die statistische Auswertung erfolgte mit Mann-Whitney-U und Wilcoxon-Test.<br /><strong>Ergebnisse:</strong> Zur Baseline lagen die Konzentrationen der Flavan-3-ole unter den jeweiligen Nachweisgrenzen und waren mit Ausnahme von ECG (WT vs. Kontrolle, p=0,02) zwischen den Gruppen nicht verschieden. Tmax von EC, ECG und EGCG wurde 2 h nach GT-Konsum erreicht, bei WT nach 2 h (ECG), 3 h (EGCG) bzw. 5 h (EC). Die AUC_0-8h von EC war bei GT im Vergleich zu größer zu WT und Kontrolle größer (beide: p<0,01). Die AUC_0-8h von EGCG (WT, GT: p<0,01) und ECG (WT, GT: p<0,001) war größer nach Konsum von WT und GT im Vergleich zur Kontrolle, aber nicht untereinander verschieden. Die weiteren Baselinewerte waren zwischen den Gruppen, mit Ausnahme von Harnsäure, nicht verschieden. TEAC wurde durch Teekonsum nicht verändert, nahm aber in der Kontrollgruppe ab. Vitamin C stieg 2 h nach GT- und Wasserkonsum und fiel nach 5 h auf das Baselineniveau ab. In allen Gruppen wurde nach 2 h ein temporärer Anstieg der DNA-Schäden <em>in vivo </em>beobachtet, während kein Effekt auf DNA-Schäden<em> ex vivo</em> und F₂ Isoprostane gemessen wurde.<br /><strong>Schlussfolgerungen:</strong> Unterschiede in der Plasmakinetik sind wahrscheinlich durch den unterschiedlichen Flavan-3-ol-Gehalt der Tees sowie durch ihr Flavan-3-ol-Muster zu erklären. Die Effekte von Grünem Tee sind trotz der höheren Konzentration an Flavan-3-olen im Teeaufguss nicht verschieden von denen von Weißem Tee, da die Absorption einzelner Flavan-3-ole aus Weißem Tee höher zu sein scheint. Das Ausbleiben von Effekten auf Biomarker für oxidativen Stress wie F₂ Isoprostane und DNA-Einzelstrangbrüche<em> ex vivo</em> könnte durch den hohen antioxidativen Schutz bei gesunden Nichtrauchern erklärbar sein. Die Bolusgabe unfermentierter Tees führt zu einem Anstieg der Flavan-3-ole im Plasma, aber beeinflusst nicht die antioxidative Kapazität sowie oxidative Biomarker.

<strong>Bolus ingestion of White and Green Tea in healthy volunteers - Plasma kinetics of flavan 3 ols and influence on oxidative biomarkers</strong><br /><strong>Introduction:</strong> It is well accepted that the ingestion of White tea and Green tea affects the pro/antioxidative balance due to their high content of antioxidative flavan-3-ols. Thus the increase of flavan-3-ols in plasma is assumed as a requirement for the health-promoting effects of non-fermented teas. The aim of this open, randomized, controlled trial was to determine the influence of a bolus consumption of White tea and Green tea on plasma kinetics of flavan-3-ols (catechin, epicatechin, epigallocatechin gallate, epicatechin gallate) in healthy nonsmokers compared with a control group (mineral water). In addition the effects of the bolus intake of 600 mL of these beverages were examined on the antioxidant capacity in plasma as well as on oxidative biomarkers.<br /><strong>Methods:</strong> 68 healthy nonsmokers (21 ♂, 47 ♀; 24±4 years; BMI 22,0±2,2 kg/m²) consumed after a polyphenol-free diet on the day before treatment 600 mL White tea (EC: 28 µmol/L; ECGC: 46 µmol/L; ECG: 11 µmol/L), Green tea (EC: 88 µmol/L; EGCG: 201 µmol/L; ECG: 25 µmol/L) or water. Blood was withdrawn before as well as 1, 2, 3, 5 and 8 h after consumption for the plasma kinetics and before as well as 2 and 5 h after consumption for the further parameters. The concentrations of C, EC, ECG and EGCG were measured by HPLC-EC with 0,6 V as the sum of free flavan-3-ols as well as glucuronidated and sulfated conjugates in the plasma and from this AUC_0-8h, Cmax, Tmax and T_1/2 were determined. The total phenol content as well as uric acid were determined photometrically and vitamin C was measured by HPLC. In addition the antioxidative capacity as TEAC and 8-isoprostaglandine-F_2α were measured with ELISA. DNA single strand breaks in leukocytes were measured <em>in vivo </em>and<em> ex vivo</em> (H₂O₂ treatment) with the Comet Assay. The statistic evaluation was performed with Mann-Whitney-U and Wilcoxon test.<br /><strong>Results:</strong> The concentrations of the flavan-3-ols were under the respective detection limits at baseline and were not different between the groups with exception of ECG (WT vs. control, p=0,02). Tmax of EC, ECG and EGCG was reached 2 h after GT consumption and with WT intake after 2 h (ECG), 3 h (EGCG) and/or 5 h (EC). The AUC_0-8h of EC was higher with GT compared with WT and controls (both: p<0.01). The AUC_0-8h of EGCG (WT, GT: p<0.01) and ECG (WT, GT: p<0.001) was higher after consumption of WT and GT in the comparison controls, but not between themselves. The further baseline values were not different between the groups, with exception of uric acid. TEAC was not changed by tea consumption, however decreased in the control group. Vitamin C increased 2 h after GT and water consumption and decreased after 5 h to baseline value. A temporary increase of the DNA damage was observed <em>in vivo</em> in all groups after 2 h, while no effect was measured on DNA damage <em>ex vivo</em> as wells as F₂-isoprostanes.<br /><strong>Conclusions:</strong> Differences in plasma kinetics are probably to be explained by the different content of flavan-3-ols of the teas as well as by their flavan-3-ol composition. The effects of Green tea are not different to White tea despite the higher concentration of flavan-3-ols, since absorption of individual flavan-3-ols from White tea seems to be higher. The lacking effect on biomarkers for oxidative stress like F₂-isoprostane and DNA single strand breaks <em>ex vivo</em> could be explained by the high anti-oxidative protection in healthy nonsmokers. The bolus consumption of non-fermented teas leads to an increase of flavan-3-ols, but has no effect on antioxidative capacity as well as oxidative biomarkers.
