Funktionelle Analyse von PLCζ mit Bezug zur Aktivierung boviner Eizellen

Bei den Eizellen der Säugetiere aktiviert das fertilisierende Spermium die Entwicklung der Eizelle, indem es eine langanhaltende Folge von Oszillationen der cytosolischen Ca²⁺-Konzentration auslöst. Ca²⁺-Oszillationen sind für die Initiation der Eizellaktivierung und der Embryonalentwicklung unerlässlich. Der genaue Mechanismus, der die intrazellulären Calciumoszillationen auslöst, ist immer noch ungeklärt. Es existieren jedoch vermehrt Studien, die auf einen Botenstoff, den Spermienfaktor (SF) hinweisen. Dieser Spermienfaktor wird vom fertilisierenden Spermium zum Zeitpunkt der Gametenfusion in das Ooplasma der Eizelle entlassen. Eine Theorie der Signalübermittlung bei der Fertilisation ist, dass der SF auf diese Weise die Ca²⁺-Oszillationen auslöst. Kürzlich wurde von Saunders et al. (2002) diese spermien-spezifische Protein identifiziert als eine neue Isoform der Phospholipase C (PLCζ). Es wurde gezeigt, dass PLCζ in der Lage ist, in unfertilisierten Mäuseeizellen Ca²⁺-Oszillationen hervorzurufen.<br /> Wir untersuchten zunächst die Ca²⁺-Oszillationsmuster in bovinen IVM Eizellen nach Mikroinjektion eines Spermienextraktes, der den SF enthielt. Es wurde beobachtet, dass die Ca²⁺-Freisetzung in einer Dosis-abhängigen Form erfolgt. Wir wiesen weiterhin nach, dass in vivo-gereifte Eizellen sensitiver auf die Anwesenheit eines Eizell-aktivierenden Faktors reagieren als IVM Eizellen. Sie exprimieren ein gleichmäßigeres Oszillationsmuster. Ferner berichten wir, dass die Injektion einer komplementären RNA der bovinen PLCζ in verschiedenen Konzentrationen nicht in der Lage ist, in bovinen IVM Eizellen Ca²⁺-Oszillationen auszulösen. Dieses Ergebnis widerspricht den Resultaten anderer Arbeitsgruppen (Malcuit et al. 2005). Andererseits konnten wir in 2 von 5 injizierten in vivo-maturierten Eizellen sowohl eine transiente Ca²⁺-Freisetzung, als auch eine Oszillation beobachten.<br /> In dieser Arbeit können wir die Wirkung eines Spermienextraktes auf die Ca²⁺-Ausschüttung in bovinen Eizellen aus dem in vitro-System und dem in vivo-System. bestätigen. Die Detektion von Ca²⁺-Oszillationen in bovinen Eizellen unterschiedlichen Ursprungs betreffend könnte darauf hinweisen, dass PLCζ auch beim Rind der Eizell-aktivierende Faktor sein könnte. Es bedarf jedoch noch weiterer Forschung, um die Ca²⁺-freisetzende Wirkung von PLCζ in bovinen Eizellen zu bestätigen.

<strong>Functional analysis of PLCζ regarding activation of bovine oocytes</strong><br /> In mammalian eggs, the fertilizing sperm activates development of the oocyte by inducing a prolonged series of oscillations in cytosolic free Ca²⁺ concentration. Ca²⁺ oscillations are essential for initiation of egg activation and embryonic development. Although the exact mechanism leading to initiation of intracellular calcium oscillations still remains unclear, accumulating studies suggest that a substance, termed sperm factor (SF), is delivered from the fertilizing sperm into the ooplasm after gamete membrane fusion. One theory of signal transduction at fertilization is that this SF causes the Ca²⁺ oscillations. Recently, this sperm-specific protein was identified as a novel isoform of a phospholipase C (PLCζ) (Saunders et al. 2002). It was shown that PLCζ triggers Ca²⁺ oscillations in unfertilized mouse oocytes.<br /> We examined the patterns of Ca²⁺ signalling in bovine IVM oocytes upon microinjection of crude sperm extracts of different concentrations which contain the SF. It is observed that Ca²⁺ release as a response to a soluble activating SF are dose-dependent. We present evidence that in vivo matured bovine oocytes are more sensible towards the presence of an oocyte-activating factor than IVM oocytes, and elicit a more consistent oscillation pattern.<br /> Further we report that microinjection of the complementary RNA for bovine PLCζ in different concentrations does not cause Ca²⁺ oscillations in bovine IVM oocytes. This finding is in contradiction to earlier results (Malcuit et al. 2005). On the contratry, we could detect a transient Ca²⁺ release in 2 from 5 injected in vivo matured bovine oocytes.<br /> In this thesis, we could confirm the effect of a sperm extract on the Ca²⁺ releasing ability of bovine IVM and in vivo oocytes. Regarding the detection of Ca²⁺ oscillations in bovine oocytes of different origins upon the injection of a cRNA of PLCζ, we can say that PLCζ could be the oocyte-activating factor also in the bovine. Yet, further work has to be done to confirm the Ca²⁺-releasing ability of PLCζ in oocytes.
