Regulation der Biosynthese von Sekundärmetaboliten und praktische Nutzung von Pflanzenextraktem am Beispiel von Streptomyces bottropensis und Plasmopara viticola | Biosynthesis Regulation of Secondary Metabolites and Practical Use of Plant Extracts on the Examples of <i>Streptomyces bottropensis and Plasmopara viticola</i>

Bakterielle sowie pflanzliche Sekundärmetaboliten bilden ein außerordentlich großes Reservoir an Stoffen, welche gegen Erkrankungen bei Säugetieren aber auch zum Schutz von Pflanzen gegenüber Phytopathogenen genutzt werden können. Während die Suche nach praktisch nutzbaren Naturstoffen seit Jahrzehnten intensiv betrieben wird, ist die biologische Funktion bei der Mehrheit dieser Stoffe völlig unbekannt. In der Phytopathologie verbinden sich die beiden Forschungsrichtungen, da Sekundärmetaboliten zum einen als Virulenz- bzw. Pathogenitätsfaktoren von Krankheitserregern wirken und Naturstoffe zum anderen als Leitsubstanzen für die Entwicklung von Pflanzenschutzmitteln dienen können. Neben direkten Wirkungen von Sekundärmetaboliten auf pflanzliche Schaderreger spielen viele bioaktive Stoffe auch bei der Regulation komplexer Sachverhalte eine wichtige Rolle. Ziel der hier präsentierten Untersuchungen ist es, den Einfluss des aus Streptomyces bottropensis isolierten bioaktiven Pyridons Iromycin A bei der Penetration von Kartoffeln näher zu untersuchen. Die Fähigkeit von Streptomyceten Wirtspflanzen befallen zu können, korreliert direkt mit dem Produktionsvermögen des Phytotoxins Thaxtomin A. Als ein essentieller Schritt der Thaxtomin A Biosynthese ist das Einwirken einer bakteriellen Stickstoffmonoxid-Synthase (NOS) anzusehen. Da Iromycine als bekannte NO Synthase-Inhibitoren fungieren, wurde zunächst anhand von Fütterungsexperimenten der Einfluss von Iromycin A auf die Thaxtomin A Produktion überprüft. Bei HPLC-DAD sowie HPLC-MS Auswertungen ergaben sich eine dosisabhängige Verringerung der Thaxtomin A Produktion bei Verwendung zweier unterschiedlicher phytopathogener Stämme von Streptomyces spp.. Es kann daher vermutet werden, dass Iromycin A einen direkten Einfluss auf die Thaxtomin A Biosynthese ausübt und somit auch ein Einfluss auf die Pathogenität eines Stammes einhergeht. Hierauf aufbauend wurden im Weiteren Untersuchungen zur Produktionsdynamik von Iromycinen und Thaxtomin in planta sowie der Einfluss der Einzelmetaboliten auf das pflanzliche Wachstum geprüft. Analog zu den Fütterungsexperimenten wurde neben der Detektion der Sekundärmetaboliten mittels HPLC-DAD und HPLC-MS auch die bakterielle Biomasse mittels quantitativer Real-Time PCR ermittelt. Neben der Regulation der Biosynthese von Sekundärmetaboliten wurden im Rahmen dieser Arbeit auch ergänzende Untersuchungen zur praktischen Nutzung von Pflanzenextrakten bei der Bekämpfung des Falschen Mehltaus der Rebe (Plasmopara viticola) durchgeführt. Hierbei standen neben einem bereits etablierten Bioassay die Optimierung der Extraktion von Pflanzeninhaltsstoffen als auch der Einfluss der Pflanzenextrakte auf das intrazellulare Wachstum des Oomyceten im Mittelpunkt der Arbeiten.

Bacterial and herbal secondary metabolites make up an enormous reservoir of substances which can benefit towards mammalian diseases and protect plants against phytopathogens. While the search for active natural products is practiced intensively for the last years, the biological function for most of these substances is completely unknown. In phytopathology both fields of research can be combined. On one hand secondary metabolites are acting as virulence- and pathogenicity factors; on the other hand many natural products can also serve as lead structures for plant protectants. Beside to these direct effects, many bioactive compounds are part of complex regulation systems. The aim of this dissertation was to investigate the influence of iromycin A, a bioactive compound, isolated from Streptomyces bottropensis, on the penetration process of Streptomyces spp. onto potatoes. It is known that the ability to penetrate host plants by Streptomyces spp. is directly correlated with their ability to synthesise the phytotoxin thaxtomin A. One essential step in the biosynthesis of the toxin thaxtomin A is performed by a bacterial nitrogen monoxide synthase (NOS). Feeding experiments with iromycin A, a known NOS inhibitor, have shown a dose dependent inhibition of thaxtomin A production on two Streptomyces strains. Therefore we hypothesize that iromycin A may have a regulative character on thaxtomin A biosynthesis that influences the pathogenicity of Streptomyces. We consequently investigated the production dynamic of iromycins and thaxtomins in planta and documented the influence of single metabolites on plant growth. As in the feeding experiments, the detection of secondary metabolites was performed by HPLC-DAD and HPLC-MS and quantification of bacterial biomass by Real-Time PCR.In addition to the biosynthesis regulation by secondary metabolites, another aspect of this work was dedicated to investigate how efficient plant extracts might be against grape downy mildew (Plasmopara viticola). Next to an already established bioassay, that monitors the influence of plant extracts on development of P. viticola, the main focus was located on extraction optimisation of bioactive plant ingredients as well as on the influence of plant extracts towards intracellular growth of the Oomycetes.
