Untersuchungen zur Embryoletalität, Genotypisierung und Resistenzlage aktueller Ornithobacterium rhinotracheale-Isolate | Investigations into the embryo lethality, genotyping and resistance to antibiotics of current Ornithobacterium rhinotracheale isolates

Untersuchungen zur Embryoletalität, Genotypisierung und Resistenzlage aktueller Ornithobacterium rhinotracheale-Isolate Mittels Kultivierung sowie Anwendung biochemischer und molekularbiologischer Methoden wurden 117 Isolate, die aus erkrankten Mastputen zwischen 2003 und 2006 in Deutschland und Frankreich isoliert wurden, als ORT identifiziert. Unterschiede wurden in der Koloniemorphologie und einigen biochemischen Merkmalen beobachtet. Die Serotypisierung zeigte eine heterogene Verteilung, in der der Serotyp A am häufigsten auftrat. Auffällig war das vermehrte Auftreten des Serotyps C sowohl in Deutschland als auch in Frankreich. Der zweite Teil der Arbeit befasste sich mit der Genotypisierung mittels Fingerprinting. Hierzu wurde die Eignung der Primer ERIC 1R und ERIC 2 sowie des Primers M 13 und der zugehörigen PCR-Methode (ERIC-PCR bzw. RAPD-PCR) ermittelt. Unter Nutzung des Primers M 13 wurden unterscheidbare Fragmentmuster der Standardserotypen A, B, D, E, F, G, I, K, M und O produziert. Die Fragmentmuster der Standardstämme C, H und Q, J und L sowie N und P ließen aufgrund großer Ähnlichkeit eine Differenzierung nicht zu. Ausgewählte deutsche ORT-Isolate der Serotypgruppen A, B, C und E wurden der PCR mittels Primer M 13 unterzogen. Die größte genetische Ähnlichkeit wurde innerhalb der Serotypgruppe A festgestellt. Die als Serotyp B charakterisierten Feldisolate wiesen eine geringe genetische Ähnlichkeit zu ihrem Standardstamm auf und zeigten unterschiedliche Fragmentmuster, die wiederum eine große Vergleichbarkeit mit den Mustern der Feldisolate des Serotyps A offenbarten. Für die Isolate der Serotypgruppen C und E wurde eine Vielzahl an Fragmentmustern erstellt. Eine sichere Typisierung mittels Fingerprinting unter Verwendung des Primers M 13 scheint nur für die Isolate des Serotyps A möglich. Die Embryoletalität der deutschen ORT-Isolate wurde mittels Infektion von Hühnerembryonen mit ca. 1000 KBE pro Ei via Allantoishöhle bestimmt. Anhand der Parameter Mortalitätsrate und Überlebensindex wurden 54 bzw. 69 Isolate als niedrig pathogen sowie 34 bzw. 19 Isolate als mäßig pathogen bewertet. Es wurden keine hoch pathogenen Isolate nachgewiesen. Gegenstand des vierten Teils der Arbeit war die Bestimmung der minimalen Hemmstoffkonzentration (MHK) aller 117 ORT- Feldisolate unter Anwendung der Mikrobouillondilutionsmethode. Unter Nutzung kommerzieller Mikrotiterplatten wurden MHK-Werte von 9 antimikrobiellen Wirkstoffen gegen ORT ermittelt und diese aufgrund fehlender Grenzwerte in Form von MHK50, MHK90 sowie Minimal-, Maximal- und Modalwerten dargestellt. Alle Isolate wiesen gegenüber Tiamulin niedrige MHK50 und MHK90 -Werte auf und ließen sich als empfindlich einstufen. Für die verbleibenden Wirkstoffe Penicillin G, Ampicillin, Enrofloxacin, Erythromycin, Ceftiofur, Lincomycin, Tetracyclin und Trimethoprim/Sulfamethoxazol wurden MHK-Werte in hohen Konzentrationsbereichen ermittelt, sodass auf eine Unempfindlichkeit von ORT gegenüber diesen Substanzen geschlossen werden kann.

Investigations into the embryo lethality, genotyping and resistance to antibiotics of current Ornithobacterium rhinotracheale isolates The first part of the present study focussed on the identification and characterisation of German and French ORT isolates, which were isolated between 2003 and 2006 from affected turkeys of different age. By using cultivation as well as biochemical and molecular biological methods, 117 field isolates were unambiguously identified as ORT. Differences were observed in colony morphology and some and biochemical characteristics. Serotyping by AGP test. German isolates were classified as serotypes A, B, C, D, E, H, I or J, while the French isolates were classified as serotypes A, C, I or J. Within these heterogeneous distributions the serotype A appeared most often. The frequent occurrence of serotype C within the German and the French isolates was surprising. In the second part two fingerprinting methods for ORT genotyping were evaluated. The suitability of primers ERIC 1R and ERIC 2 as well as of primer M 13 and the corresponding PCRs (ERIC-PCR or RAPD-PCR) was determined. RAPD-PCR by using primer M 13 was able to distinguish reference strains of serotypes A, B, D, E, F, G, I, K, M and O based on fragment patterns. In contrast fragment patterns of reference serotypes C, H and Q, J and L, as well as N and P were very similar so that a differentiation between these serotypes was not possible. Based on these results selected German isolates of serotype A, B, C and E were investigated by RAPD-PCR. Of all serotypes the patterns of field isolates of serotype A showed the highest genetic similarity. The field isolates characterized as serotype B showed two different patterns which could hardly be differentiated from patterns of serotype A field isolates and had only a slight similarity to the reference serotype B. For isolates of serotypes C and E a large number of fragment patterns were produced. A reliable fingerprinting by RAPD-PCR seems to be possible only for isolates of serotype A. The third part comprised the determination of embryo lethality of the 88 German ORT isolates. Eggs were inoculated with 1000 cfu Ornithobacterium rhinotracheale via allantoic cavity. Based on mortality rate and survival rate 54 and 69 isolates respectively were categorized as lowly pathogenic and 34 and 19 isolates respectively were categorized as moderately pathogenic. No highly pathogenic isolates were detected. Subject of the fourth part was the determination of the minimal inhibitory concentration (MIC) of 88 German and 29 French ORT field isolates using the broth microdilution method. By using commercial micro titre plates MIC values of 9 antimicrobial substances against ORT were estimated. Because of missing breakpoints these values were presented as minimal-, maximal- and modal-value as well as MIC50 and MIC90. All isolates showed low MIC50 and MIC90 for tiamulin. For the remaining substances penicillin G, ampicillin, enrofloxacin, erythromycin, ceftiofur, lincomycin, tetracycline and trimethoprim/sulfamethoxazole MIC values of high concentrations were estimated. Consequently a resistance and insusceptibility of ORT in Germany as well as in France could be assumed.
