Entwicklung eines indirekten ELISA-Testsystems zur Serodiagnostik der caninen Protothekeninfektion und nachfolgender Untersuchung der Prävalenz der caninen Protothekose | The development of an indirect ELISA testsystem for serodiagnosis of canine prototheca infection and the subsequent investigation of the prevalence of the canine protothecosis

Chlorophylllose, Hefe-ähnliche Mikroalgen der Gattung Prototheca (P.) sind die einzigen bekannten Pflanzen, welche infektiös für Menschen und Tiere sein können. Bis heute sind 5 verschiedene Spezies beschrieben, wobei Prototheca zopfii Genotyp 2 ursächlich für die Protothekenmastitis von Milchkühen und für die sporadisch auftretende Protothekeninfektion beim Hund ist. Das klinische Erscheinungsbild betroffener Hunde ist häufig assoziiert mit einer hämorrhagischen, therapieresistenten Enterokolitis, welche später in der Generalisierung der Erreger mit okularen und zentralnervösen Störungen vergesellschaftet ist. Da die Hundeprotothekose in den meisten Fällen durch P. zopfii Genotyp 2 ausgelöst wird, war die Zielstellung der vorliegenden Arbeit die Entwicklung eines ELISA-Testsystems, zum Nachweis von Prototheken- spezifischen Antikörpern der Gattung P. zopfii Genotyp 2 in Hundeseren und darauf aufbauend anschließend die Ermittlung der Prävalenz von Protothekeninfektionen in einer größeren Hundepopulation. Hierzu wurden 363 Hunde aus der Patientenklientel der Kleintierklinik der FU Berlin kulturell und serologisch untersucht. Eingeteilt in 3 Gruppen beinhaltete Gruppe 1 (n=236) als Kontrollgruppe gesunde Hunde, akut erkrankte und orthopädisch erkrankte Patienten sowie Hunde mit nachgewiesenen benignen Neoplasien. Gruppe 2 (n=65) umfasste Hunde mit gastrointestinaler Symptomatik und Gruppe 3 (n=62) involvierte eine Gruppe von Hunden mit chronischen, malignen bzw. immun- mediierten Erkrankungen ohne gastrointestinale Symptome. Zunächst konnte ein indirektes ELISA-Testsystem entwickelt und etabliert werden, mit dem der Nachweis von P. zopfii Genotyp 2-spezifischen Antikörpern bei enteritischen, an Protothekose erkrankten Hunden gelang. Dabei wies der entwickelte ELISA mit einer Spezifität und Sensitivität von jeweils 100 % eine sehr gute diagnostische Eignung auf. Im Rahmen des in dieser Studie untersuchten Patientenkollektivs konnte die canine Protothekose sowohl kulturell als auch serologisch bei zwei Hunden (Fälle FU 1 und FU 2) nachgewiesen werden. Dies entspricht einer Prävalenz von 3,1 % der enteritisch erkrankten Hunde der Gruppe 2. Bei Hunden der Kontrollgruppe 1 und bei chronisch, maligne bzw. immun-mediiert erkrankten Hunden (Gruppe 3) konnten keine Prototheken in den Fäzes oder Prototheken-Antikörper im Blutserum nachgewiesen werden. Infolge der in dieser Studie nachgewiesenen Prävalenz der caninen Protothekose von 3,1 % bei enteritisch erkrankten Hunden, sollte die Protothekeninfektion beim Enteritisgeschehen des Hundes, und hier insbesondere bei therapieresistenten, chronischen, hämorrhagischen Enteritiden, differentialdiagnostisch berücksichtigt werden. Aufgrund der intermittierenden Erregerausscheidung in den Fäzes und der damit verbundenen Gefahr möglicher falsch negativer Kulturbefunde kann der serologische Nachweis von Prototheken-spezifischen Antikörpern, verglichen mit den bisherigen diagnostischen Möglichkeiten, von großem Nutzen sein. So ermöglicht der entwickelte indirekte Prototheken-ELISA eine schnelle und sichere Identifikation von an Protothekose erkrankten Hunden.

Achlorophyllous, yeast-like microalgae of the genus Prototheca (P.) are the only known plants, able to cause infections in humans and animals. Until now five different species have been described. One of them –Prototheca zopfii genotype 2– was identified to be responsible for inflammation of the mammary gland in dairy cows as well as for the sporadically occurring prototheca infection in dogs. In the case of affected dogs the clinical signs are often associated with hemorrhagic, therapy-resistant enterocolitis, which later, in the generalized manifestation of the pathogens, occur in combination with ocular and central nervous disorders. Due to these observations, the present study was focused on the following objectives: (1) to develop an ELISA test system in order to detect prototheca-specific antibodies of the genus Prototheca zopfii genotype 2 in dog serum and based on this (2) to investigate the prevalence of Prototheca infection in a large dog population. For this purpose 363 dogs, divided into three groups, were culturally and serologically tested. The first group (n=236) included healthy dogs, acutely ill and orthopedic ill patients as well as dogs with proven benign tumors. The second group (n=65) consisted of dogs with gastrointestinal symptoms and the third group (n=62) involved dogs that had chronic, malign or immune-mediated diseases without any gastrointestinal signs. In addition an ELISA test system could be established that succeeded in demonstrating species-specific P. zopfii genotype 2 antibodies in enteritic ill patients suffering from protothecosis. With a specificity and sensitivity of 100 % the developed ELISA test indicated a very good diagnostic suitability. Within the scope of this study, the canine protothecosis was confirmed culturally and serologically in 2 cases (case FU 1 and FU 2) of the investigated dog population. This corresponds to a prevalence of 3.1 % in the enteritic dogs from group 2. In dogs from group 1 and 3 no Prototheca species were found in the feces or in the blood serum. The proven prevalence of 3.1 % of the canine protothecosis in enteritic dogs suggests that the Prototheca infection should be considered as one differential diagnosis of dogs showing clinical signs of gastrointestinal disease. Due to intermittent pathogen shedding in the feces and thereby the possible danger of false negative results, the established serological test can be very useful. Compared to previously described methods, the indirect test system, depicted in this study, enables a fast and secure identification of Prototheca antibodies in blood serum and therefore provides a promising tool for Prototheca diagnostic in the clinical practice.
