Bestimmung der Ceruloplasminkonzentration im Blutserum und Blutplasma von Milchkühen | Determination of the concentration of Ceruloplasmin in blood serum and blood plasma of dairy cows

Die Bestandsbetreuung ist Teil der tierärztlichen Arbeit im Milchviehbetrieb. Sie umfasst die Überwachung verschiedener Parameter zur Einschätzung der Tiergesundheit. Eine wichtige Rolle spielt in diesem Zusammenhang die Kupferversorgung der Rinder. Zu deren Untersuchung kann die Messung des Akute- Phase- Proteins Ceruloplasmin in Blutproben hilfreich sein. Bisher wurden verschiedene Verfahren zu dessen labordiagnostischer Bestimmung beschrieben. Die große Anzahl an entwickelten Untersuchungsmethoden legt nahe, dass noch kein Verfahren gefunden werden konnte, welches allen Anforderungen genügt. Zielstellung der vorliegenden Arbeit war die Ceruloplasminbestimmung im Rinderblut und die Etablierung eines automatisierten Verfahrens auf der Grundlage der manuellen Methode nach Sunderman und Nomoto (1970). Die Probengewinnung für diese Untersuchung erfolgte im Rahmen der Bestandsbetreuung durch die Klinik für Klauentiere der Freien Universität Berlin. Der Zeitraum der Probensammlung erstreckte sich von 2014 bis 2017. Indikationen waren Routinebesuche zur Erhebung eines Status quo und dem Erkennen eines möglichen Verbesserungspotentials bis hin zur Lösung spezifischer Bestandsprobleme. Die untersuchten Tiere wurden nach Laktationsstatus in vier Gruppen eingeteilt. Zunächst wurden Untersuchungen zur Präzision des manuellen und automatischen Messverfahrens durchgeführt. Die ermittelten intraday- Variationskoeffizienten für Serumproben betrugen für die manuelle Methode 0,012 und 0,015 sowie 0,010 und 0,024 für die automatische Methode. Für die Plasmaproben lauteten die entsprechenden Koeffizienten für die manuelle Methode 0,020 und 0,032 sowie 0,011 für die automatische Methode. Die für die Präzision von Tag zu Tag ermittelten Variationskoeffizienten waren von der zugrunde gelegten Untersuchung und dem betrachteten Zeitraum abhängig. Die niedrigsten Koeffizienten konnten über den Lagerungsversuch bei Kühlschranktemperatur ermittelt werden. Für die Serumproben betrug der interday- Variationskoeffizient 0,029 und 0,021 für die entsprechenden Plasmaproben. Die aus den Kontrollproben gewonnenen Werte betrugen 0,038 bzw. 0,047. Nach der Etablierung der Methode konnten interday- Variationskoeffizienten unter 0,088 aus Serum- bzw. unter 0,086 aus Plasmaproben erzielt werden. Die Variationskoeffizienten des Lagerungsversuches wurden über 12 Tage, also einen sehr viel kürzeren Zeitraum als bei den anderen hier durchgeführten Untersuchungen (ca. 1 Jahr) ermittelt. Alle ermittelten interday- Variationskoeffizienten waren kleiner als 0,09. Sowohl für die manuelle als auch die automatische Methode konnte eine hohe Präzision in der Serie und von Tag zu Tag nachgewiesen werden. Dies gilt auch im Vergleich mit in anderen Studien untersuchten Methoden zur Ceruloplasminbestimmung. In bereits erfolgten Untersuchungen wurde herausgefunden, dass Chlorid einen hemmenden Einfluss auf die Reaktion hat. Dieser Effekt konnte in der vorliegenden Arbeit bestätigt werden. Es wurde nachgewiesen, dass der hemmende Einfluss linear mit der Chloridkonzentration zunimmt. Die Stabilität der Ceruloplasminkonzentration in Blutproben wurde unter verschiedenen Bedingungen in einem Lagerungsversuch untersucht. Gewonnene Proben konnten über einige Tage (5- 7 Tage) stabil bei Raumtemperatur (20°C) oder im Kühlschrank (8°C) gelagert werden. Auch mehrfaches Einfrieren und Auftauen der Proben beeinflusste die Konzentration nicht wesentlich. Eine Lagerung für ein Jahr bei -20°C verminderte die Ceruloplasminaktivität um ca. 10- 14 %, bzw. fiel die Ceruloplasminkonzentration in Serumproben in 30 Tagen um ca. 1,2 und in Plasmaproben um ca. 1,3 mg /l ab. Die nach Laktationsgruppen getrennte Auswertung der untersuchten Betriebe zeigte, dass die Ceruloplasminkonzentration nach der Kalbung am höchsten war. Der Mittelwert der mit der manuellen Methode bestimmten Konzentrationen in Serumproben betrug 160± 22,6 bzw. 227± 31,6 mg/ l in Plasmaproben. Mit der automatischen Methode wurden Werte von 123± 18,8 bzw. 166± 21,8 mg/ l ermittelt. Der Vergleich der beiden Messmethoden lieferte hohe Pearson- Korrelationskoeffizienten. Für die mit der manuellen und automatischen Methode bestimmten Konzentrationen in Serumproben betrug der Korrelationskoeffizient 0,922 (p< 0,001) bzw. 0,868 (p< 0,001) in Plasmaproben. Zudem wurden Korrekturfaktoren ermittelt, um die Ergebnisse der manuellen mit denen der automatischen Methode vergleichen zu können. Sie lagen zwischen 1,30 und 1,42 und waren somit niedriger als der theoretische Korrekturfaktor von 1,74. Die Bland- Altman- Analyse ohne Anwendung der Korrekturfaktoren ergab, dass die mit der automatischen Methode ermittelten Konzentrationen in Serumproben 34± 9,6 mg/ l unter den mit der manuellen Methode ermittelten Ceruloplasminkonzentrationen lagen. Für Plasmaproben waren es entsprechend 55± 14,2 mg/ l. Nach der Anwendung der ermittelten Korrekturfaktoren wurde mit der Bland- Altmann- Analyse kein statistisch signifikanter Unterschied zwischen den mit beiden Methoden parallel bestimmten Ceruloplasminkonzentrationen nachgewiesen. Der Vergleich von Serum- und Plasmaproben zeigte, dass die Ceruloplasminkonzentrationen in Serumproben geringer als in den zugehörigen Plasmaproben waren. Das Verhältnis der Konzentrationen in gepaarten Serum- und Plasmaproben war sehr variabel. Die ermittelten Quotienten aus den Konzentrationen beider Probenarten lagen zwischen 1,07 und 1,91. Aus den Betriebsmessungen ergaben sich über alle Laktationsgruppen Quotienten von 1,43 für die manuelle und 1,37 für die automatische Methode. Zwischen einzelnen Betrieben waren die Abweichungen größer als zwischen den Laktationsgruppen innerhalb der Betriebe. Es wurden die Konzentrationen von separaten Plasmaproben und den daraus hergestellten Poolproben ermittelt. Aus den Mittelwerten der separaten Proben und den Ergebnissen der Poolproben resultierte ein Pearson- Korrelationskoeffizient von 0,978 (p< 0,001). Das automatisierte Messverfahren bietet gegenüber der manuellen Methode sowohl ökonomische als auch ökologische Vorteile. Es lieferte präzise reproduzierbare Ergebnisse und kann unter Berücksichtigung von Korrekturfaktoren als Alternative zum manuellen Verfahren nach Sunderman und Nomoto (1970) zur routinemäßigen Bestimmung der Ceruloplasminkonzentration aus Rinderblutproben verwendet werden.

Herd supervision belongs to veterinary work in the dairy farming. It includes the monitoring of various parameters for the assessment of animal health. In this context, the copper supply of cattle plays an important role. The measurement of the acute- phase- protein ceruloplasmin in blood samples may be helpful for its investigation. So far, several methods have been described for laboratory diagnostic analysis of ceruloplasmin. However this supports that no method has been found which meets all requirements. The aim of the present study was to determine ceruloplasmin in cattle blood and to establish an automated procedure based on the manual method according to Sunderman and Nomoto (1970). Samples for this study have been collected during regular visits by the veterinary ambulance of the clinic for claw animals of the Free University of Berlin. The period of the sample collection was between 2014 to 2017. Indications included routine visits, visits to identify options for improval or to solve specific questions. The examined animals were divided into four groups by lactation status. First of all, the precision of the manual and automated measuring methods was investigated. The intra- assay coefficients of variations for serum samples were 0.012 and 0.015 for the manual method and 0.010 and 0.024 for the automated method. For plasma samples, the corresponding coefficients for the manual method were 0.020 and 0.032 as well as 0.011 for the automated method. The coefficients of variations determined for day- to- day precision depended on the underlying study and the period considered. The lowest coefficients could be determined for the storage experiment at refrigerator temperature. For serum samples, the interday variation coefficient was 0.029 and 0.021 for the corresponding plasma samples. The values calculated from the control samples were 0.038 and 0.047, respectively. After establishing the method, interday coefficients of variations below 0.088 could be obtained from serum samples and less than 0.086 for plasma samples. The coefficients of variations of the storage experiment were determined over 12 days, i.e. a much shorter period of time than in the other tests of this study (about 1 year). All interday coefficients of variations were less than 0.09. For both the manual and the automated method, high precision in the series and from day to day could be proven. This also applies in comparison with methods for ceruloplasmin analysis which have been investigated in other studies. In previous studies, it has been found that chloride has an inhibitory effect on the reaction. This effect was confirmed in the present work. It has been shown that the inhibitory influence increases linearly with the chloride concentration. The stability of ceruloplasmin concentration in blood samples was tested under various conditions in a storage trial. Samples collected could be stored stable for a few days (5- 7 days) at room temperature (20°C) or in the refrigerator (8°C). Multiple freezing and thawing of the samples did not considerably affect the concentration. Storage for one year at -20°C reduced the ceruloplasmin activity by approx. 10- 14 %, or the ceruloplasmin concentration decreased by approx. 1.2 in serum samples and by approx. 1.3 mg / l in 30 days in plasma samples. Dividing the samples into lactation groups showed that the ceruloplasmin concentration was highest after calving. The mean value measured by the manual method in serum samples was 160± 22.6 and 227± 31.6 mg / l in plasma samples, respectively. Values of 123± 18.8 and 166 ± 21.8 mg / l were determined by the automated method. The comparison of the two methods provided high Pearson correlation coefficients. For the concentrations determined by the manual and automated method in serum samples, the correlation coefficient was 0.922 (p< 0.001) and 0.868 (p< 0.001) in plasma samples, respectively. Correction factors were determined to compare the results of the manual methods with those of the automated method. They were between 1.30 and 1.42 and were thus lower than the theoretical correction factor of 1.74. The Bland- Altman analysis without applying the correction factors showed that the serum concentrations of the automated method was 34± 9.6 mg / l lower than the ceruloplasmin concentrations determined by the manual method. For plasma samples it was 55± 14.2 mg / l, respectively. Following the application of the correction factors determined, the Bland- Altmann analysis showed no statistically significant difference of the ceruloplasmin concentrations when comparing both methods. The comparison of serum and plasma samples showed that the ceruloplasmin concentrations in serum samples were lower than in the corresponding plasma samples. Moreover the ratio of concentrations in paired serum and plasma samples was very variable. The quotients from the concentrations of both types of samples were between 1.07 and 1.91. The measurements of the farms showed quotients of 1.43 for the manual method and 1.37 for the automatic method across all lactation groups. Between individual farms the differences were greater than between the lactation groups within the farms. The concentrations of separate plasma samples and the pool samples prepared therefrom were determined. The mean values of the separate samples and the outcome of the pool samples resulted in a Pearson correlation coefficient of 0.978 (p <0.001). The automated measuring method offers both economic and ecological advantages over the manual method. It provides precise and reproducible results and can be used with correction factors as an alternative to the manual method of Sunderman and Nomoto (1970) for the routine analysis of ceruloplasmin concentrations from bovine blood samples.
