Einfluss der Verfütterung des Probiotikums E. faecium NCIMB 10415 im frühen postnatalen Stadium auf die Zusammensetzung und Stoffwechselaktivität der gastrointestinalen Mikrobiota bei Ferkeln | Effect of the probiotic E. faecium (NCIMB 10415) as a diet supplement in the early post-natal period on the composition and the metabolic activity of the gastrointestinal microbiota of piglets

Ziel der vorliegenden Arbeit war es, den Einfluss der frühzeitigen Gabe des Probiotikums Enterococcus faecium NCIMB 10415 (ab dem 1. Lebenstag) auf die Mikrobiota des gastro-intestinalen Traktes von Ferkeln zu untersuchen. Es sollten sowohl der Verbleib des zu-geführten Stammes im Magen-Darm-Trakt der Ferkel als auch die bakterielle Zusammen-setzung der Mikrobiota des Magens, distalen Jejunums und Colon ascendens der Ferkel in allen wichtigen Produktionsphasen (Saugperiode, Übergang zum Festfutter, Absetzen) auf der Gattung- und Spezies-Ebene qualitativ und quantitativ mithilfe von Real-Time PCR unter-sucht werden. Ausgehend von der Hypothese, dass artverwandte bzw. das gleiche Habitat besiedelnde Bakterien am ehesten Kandidaten für einen probiotischen Einfluss sind, galt ein besonderes Augenmerk den gastrointestinalen Populationen der Laktobazillen. Die Arbeit wurde im Rahmen des DFG-Forschungsprojekts 438 „Integrative Analyse der Wirkungs-mechanismen von Probiotika beim Schwein“ zwischen 2005 und 2007 durchgeführt. Für den Versuch wurden Sauen der Rasse Deutsche Landrasse x Duroc und ihre Nach-kommen in zwei Fütterungsgruppen – Kontroll- und Probiotikumgruppe – eingeteilt und räumlich getrennt gehalten. Die Saugferkel wurden am 28. Lebenstag von der Muttersau ab-gesetzt. Die Supplementierung der Ferkel der Probiotikumgruppe mit dem Probiotikum be-gann am ersten Tag und dauerte bis zum 56. Lebenstag. In der Saugperiode wurden Ferkel mit dem Probiotikum ab dem 1. bis zum 14. Tag in Form eines Inokulums (4,8x109 KBE/ml) und ab dem 14. bis zum 34. Tag durch Inokulum und Beimischung zum Prästarter-Futter (1,6x107 KBE/g) versorgt. Ab dem 35. Lebenstag erfolgte die Gabe des Probiotikums ausschließlich als Beimischung zum Starter-Futter (4,1x106 KBE/g). Das Futter der Sauen sowie der Kontrollferkel enthielt kein Probiotikum. Je fünf Ferkel pro Gruppe wurden am 7., 14., 28., 35. und 56. Tag getötet, und es wurden Digestaproben aus Magen, distalem Jejunum und Colon ascendens entnommen. Der spezifische Nachweis und die Quantifizierung des probiotischen Enterococcus faecium NCIMB 10415 in den Digestaproben wurde über eine SYBR-Green Real-Time PCR mit dem spiked-matrix- Standard durchgeführt. Qualitative Veränderungen der gesamten bakteriellen Zusammensetzung in den untersuchten Proben wurden mithilfe der denaturierenden Gradienten-Gelelektrophorese (DGGE) dargestellt. Die Modifizierung der Mikroflora auf der Gattungsebene wurde quantitativ über SYBRGreen- und TaqMan- Real-Time PCR-Assays ebenso mit einem spiked-matrix-Standard und den Primern für Lactobacillus spp., Entero-coccus spp., Bifidobacterium spp. und Escherichia spp. ermittelt. Auf der Spezies-Ebene wurden Lactobacillus acidophilus, L. amylovorus, L. johnsonii, L. mucosae, L. reuteri sowie die mit dem verfütterten probiotischen Stamm eng verwandten E. faecium und E. faecalis quantifiziert. Die Bestimmung der bakteriellen Stoffwechselaktivität erfolgte durch Messung der Konzentrationen von Laktat, Ammoniumionen und flüchtigen Fettsäuren in den unter-suchten Digestaproben. Der untersuchte probiotische Stamm E. faecium NCIMB 10415 kann den Transit durch Magen und Dünndarm der Ferkel überleben und in einem vermehrungsfähigen Zustand das Colon ascendens erreichen. Die höchsten Konzentrationen des Probiotikums in Magen, distalem Jejunum und Colon ascendens wurden nach zweiwöchiger Supplementierung ge- messen, eine weitere Verfütterung führte zu keiner Erhöhung der Populationsgröße mehr. Die errechneten prozentualen Anteile von E. faecium NCIMB 10415 an der gesamten bakteriellen Population lassen die Schlussfolgerung zu, dass der untersuchte Stamm nur eine unter-geordnete Rolle in der gastrointestinalen bakteriellen Gemeinschaft spielt. Die nach dem Ab- setzen steigenden prozentualen Anteile des Probiotikums an der gesamten bakteriellen Population im Magen und im distalen Jejunum könnten darauf hindeuten, dass zumindest in diesen Lokalisationen die frühzeitige Supplementierung mit E. faecium NCIMB 10415 die weitere Zusammensetzung der Mikrobiota beeinflussen kann. Hinsichtlich der mittels DGGE ermittelten Zusammensetzung der eubakteriellen Populationen konnten in der Saugperiode nur vereinzelt statistisch signifikante Unterschiede zwischen den beiden Fütterungsgruppen ermittelt werden. Nach dem Absetzen (31., 35. und 56. Tag) jedoch waren die Magen-, Jejunum- und Colon ascendens-Mikrobioten der Probiotikum-Ferkel signifikant diverser als die der Kontrolltiere. Die bakteriellen Populationen waren zwischen den Tieren der Probiotikum-Gruppe in allen Lokalisationen und zu fast allen Beobachtungszeitpunkten ähnlicher als in der Kontrollgruppe, was ebenfalls auf eine höhere Stabilität und Homogentität der gastrointestinalen Mikrobiota in der Probiotikum-Gruppe hindeutet. Auf der Gruppen-Ebene (Lactobacillus spp., Enterococcus spp., Bifidobacterium spp., Escherichia spp.) zeigte der probiotische Stamm E. faecium NCIMB 10415 nur auf Entero-coccus spp. in Magen, Jejunum und Colon ascendens an allen Beobachtungszeitpunkten eine statistisch signifikant fördernde Wirkung. Die Zahl der für den Organismus potenziell günstigen Laktobazillen erhöhte sich im Magen und Colon ascendens in der Probiotikum- Gruppe nur tendenziell in der Saugphase und nach dem Absetzen. Für die Bifidobakterien wurde kein statistisch signifikanter Unterschied zwischen beiden Fütterungsgruppen ermittelt. Die Belastung des Magen-Darm-Traktes mit Escherichia spp. war in der Probiotikumgruppe nur geringfügig geringer als in der Kontrollgruppe. Betrachtet man die Ergebnisse der Analyse der gastrointestinalen Laktobazillen-Populationen, so lässt sich mit Blick auf L. johnsonii eine fördernde Wirkung von E. faecium NCIMB 10415 feststellen. Die Zellzahlen dieser Spezies waren in der Probiotikum-Gruppe sowohl im Magen als auch im distalen Jejunum und im Colon ascendens vor und nach dem Absetzen signifikant höher als in der Kontrollgruppe. Der unter den untersuchten Laktobazillen-Population fast in allen untersuchten Lokalisationen und an allen Beobachtungstagen dominante Stamm – L. reuteri – unterlag dagegen keinem statistisch signifikanten probiotischen Einfluss. Nur tendenziell waren Zellzahlen von L. reuteri und L. amylovorus bei den Probiotikum-Tieren, vorwiegend nach dem Absetzen, höher als jene in der Kontrollgruppe. Obwohl nur wenige untersuchte Proben L. mucosae-positiv waren, war eine statistisch signifikante Wirkung des verfütterten probiotischen Stamms auf diese Spezies im distalen Jejunum festzustellen – fast an allen Beobachtungstagen waren sowohl die absoluten als auch die relativen Zellzahlen in der Probiotikum- Gruppe kleiner als in der Kontrolle, was mit einer möglichen verminderten Abschilferung der Epithelzellen in das Darmlumen verbunden sein könnte. L. acidophilus wurde unter den untersuchten Laktobazillen-Populationen zu den Begleitspezies gezählt; der Stamm wurde an keinem Beobachtungstag und an keiner der untersuchten Lokalisationen von E. faecium NCIMB 10415 nachweislich beeinflusst. Hinsichtlich der Verbesserung der Mikrobiota durch die Produktion von Laktat waren nur wenige statistisch signifikante Unterschiede zwischen den beiden Versuchsgruppen zu registrieren. Tendenziell war die Konzentration an L(+)-Laktat in der Probiotikum-Gruppe sowohl im Jejunum als auch im Colon an fast allen Beobachtungstagen höher als in der Kontrolle. Jedoch bestand keine Korrelation zwischen den Zellzahlen von E. faecium NCIMB 10415 bzw. Enterokkoken und dem gemessenen Laktat. Dagegen konnte eine solche Korrelation für Zellzahlen von Lactobacillus spp. und einzelne andere Lactobacillus- Spezies festgestellt werden. Dies, ebenso wie die erhöhten L. johnsonii- Zellzahlen, die tendenzielle Reduzierung von Escherichia spp. sowie niedrigere Ammoniak-Konzentrationen an allen Beobachtungszeitpunkten und erhöhte Essig- und n-Buttersäure-Mengen nach dem Absetzen in der Probiotikum-Gruppe deuten darauf hin, dass E. faecium NCIMB 10415 eine zumindest indirekte Wirkung auf die intestinale Mikrobiota der Ferkel aufweist.

This study examined the effects of the probiotic Enterococcus faecium NCIMB 10415 as a diet supplement (administered from the 1st day post natum) on the intestinal microbiota of piglets. Using different PCR methods, we investigated (both qualitatively and quantitatively) the bacterial composition of the microbiota in the stomach, distal jejunum, and colon ascen-dens of piglets throughout the suckling period, the transition to solid food, and the weaning period, thereby keeping track of the applied probiotic within the gastrointestinal tract. Based on the expectation that similar bacteria and those occupying the same habitat are the most likely candidates for a probiotic influence, the main focus was on the gastrointestinal popula-tions of lactobacilli. For the trial, sows (Landrace x Duroc) and their litter were divided into a probiotic- and a control group and were kept separately. Piglets were weaned on the 28th day of life. The sup-plementation of piglets in the probiotic group began on day 1 and lasted until the 56th day of life. During the suckling period, from day 1 to 14, piglets received the probiotic via inoculum (4,8x109 CFU/ml) and via supplement to the pre-starter diet (1,6x107 CFU/g) from day 14 to 34. Starting on day 35, the probiotic was administered along with the starter diet (4,1x106 CFU/g). The sow and piglet diet of the control group contained no probiotic. Five piglets per group were euthanised on day 7, 14, 35, and 56, respectively, and digesta samples were re-moved from the stomach, distal jejunum and colon ascendens. SYBR-Green real time PCR with spiked matrix standards was applied for the specific detection and quantification of the probiotic E. faecium NCIMB 10415 in the digesta samples. Qualitative changes of the overall bacterial composition in the samples under examination were identified using the denaturing gradient gel electrophoresis (DGGE) method. Quantita-tive modifications to the microbiota on the genus level were examined by SYBR-Green and TaqMan real time PCR assays, using spiked matrix standards and primers for Lactobacillus spp., Enterococcus spp., Bifidobacterium spp., and Escherichia spp. On the species level, Lactobacillus acidophilus, L. amylovorus, L. johnsonii, L. mucosae, and L. reuteri, as well as E. faecium und E. faecalis were quantified. Concentrations of lactate, ammonium ions and short chain fatty acids were measured in order to evaluate the degree of metabolic activity. E. faecium NCIMB 10415 survived the transit through the stomach and small intestine of the piglets and was found in the colon ascendens in high cell numbers. The highest concentration of the examined probiotic in the gastrointestinal tract was measured two weeks into the trial – subsequent supplementation led to no further increase in the size of the population. The ratio of E. faecium NCIMB 10415 within the overall bacterial population suggested that the exam-ined strain plays a subordinate role in the gastrointestinal bacterial community. However, in-creasing ratios of the probiotic regarding the total bacterial population in stomach and distal jejunum samples after weaning may point to modifying effects on the composition of the mi-crobiota in these localisations, which resulted from an early supplementation with E. faecium NCIMB 10415. Regarding the eubacterial composition, only sporadic statistically significant differences be-tween the two trial groups became visible before weaning. After weaning (days 31, 35, and 56), however, the microbiota of the stomach, distal jejunum and colon ascendens of the probi- otic piglets was significantly more diverse than that of the control piglets. At the same time, bacterial populations were more similar between animals of the probiotics group in all local-isations and at almost all points of observation, which also indicates a higher consistency and homogeneity of the gastrointestinal microbiota within that group. On the bacterial group level (Lactobacillus spp., Enterococcus spp., Bifidobacterium spp., Escherichia spp.), the probiotic strain showed a significant beneficial effect only on Entero-coccus spp. in all locations and at all points of observations. The number of potentially bene-ficial lactobacilli tended to increase in the stomach and colon ascendens of probiotic piglets in both the suckling and the post-weaning period. As for bifidobacteria, no statistically signifi-cant difference could be found. The number of harmful Escherichia spp. was slightly lower in the probiotics group compared to the control group. Concerning the results of the analysis of gastrointestinal lactobacilli, an effect of E. faecium NCIMB 10415 on Lactobacillus johnsonii could clearly be discerned. The cell numbers of this species were significantly higher in the probiotics group in all three locations and both before and after weaning. The dominant lactobacillus strain – L. reuteri – was, however, not significantly affected by the administered probiotic. Cell numbers of L. reuteri and L. amylo-vorus tended to be higher in the probiotics group, but not by a significant margin. For L. mu-cosae, despite reduced presence in sample extracts (it is being detected in only a few samples), a statistically significant effect was detected: on almost all days of observation, both absolute and relative cell numbers of L. mucosae were smaller in the probiotic group compared to the control group. As this strain is known to adhere to intestinal mucus, this may be connected to a reduced cell turnover of intestinal epithelium. L. acidophilus was counted among the accompanying species within the examined population of lactobacilli – at no point in time and in none of the localisation was it demonstrably affected by E. faecium NCIMB 10415. With a view to the possible improvement of the microbiota through augmented production of lactate, only isolated significant differences between the two trial groups were registered. On almost all days of observation, there was a tendency for the concentration of l-lactate to be higher in the probiotics group, both in the jejunum and in the colon ascendens. There was, however, no correlation between the cell numbers of E. faecium NCIMB 10415 – or enteroc- coci, respectively – and the amount of lactate registered. Yet such a correlation was detected between the cell numbers for Lactobacillus spp. and single other lactobacillus species. This observation, along with the increase in cell numbers for L. johnsonii, the tendency towards a reduction of Escherichia spp., a lower concentration of ammonia at all points in time, and a quantitative increase of acetic and n-butyric acid after weaning in the probiotics group, sug-gests that E. faecium NCIMB 10415 does exert an indirect effect on the composition and ac-tivity of the intestinal microbiota of piglets.
