Dynamic induction of the UPR pathway (Unfolded Protein Response) in response to heat stress and DTT treatment in wheat tissues

اظهر المزيد [+] اقل [-]إنجليزي. The quality of wheat, an economically important cereal (800.9 million t/year produced worldwide), is determined by its grain content [1,2,3]. Environmental conditions, such as an increase intemperature due to global warming, negatively impact the wheat yield by affecting physiological and molecular mechanisms in different cellular compartments[4,5]. Modification of proteinhomeostasis inducing a cellular stress that occurs in the endoplasmic reticulum (ER) is one of the cellular events affected in response to heat stress. In response to ER-stress, a signalingpathway called UPR (Unfolded Protein Response) is induced, aiming at regulating the concentration of unfolded proteins in the ER [6]. Among the 2 branches of the UPR pathway, theunconventional splicing of the transcription factor bZIP60 (bZIP60s) by IRE1 is involved in the expression level increase of gene coding protein involved in the folding (Fig1)[7]. In this work, wedecided to use TabZIP60s as a marker of UPR induction in wheat. It is well known that SSP (Seed Storage Protein) are synthesized in the ER during grain filling [8]. Therefore, the expressionpattern of TabZIP60s in wheat seedlings tissues after a chemical inducer of ER-stress (Fig2) and during endosperm development under heat stress was studied to determine the UPR inductionprofile in wheat tissues (Fig3). Finally, the impact of UPR on SSP synthesis and quality during grain development will be observed using TaIRE1 KO mutant (Fig4).
