Identifisering av potensielle virulensfaktorer hos Pasteurella atlantica gv. salmonicida

Pasteurellose hos atlantisk laks (Salmo salar) forårsaket av P. atlantica genomvar salmonicida har siden 2018 blitt en betydelig trussel mot fiskevelferden og den bærekraftige produksjonen i norsk akvakultur. Det er derfor et stort behov for en kommersiell vaksine mot sykdommen. Til nå har man lite kunnskap om overflateproteiner, virulensfaktorer og immunogene komponenter hos denne bakterien. Denne kunnskapen er kritisk for valg av vaksineisolater og strategier, og for å forstå hvordan denne bakterien forårsaker sykdom. I denne masteroppgaven ble potensielle virulensfaktorer og immunogene komponenter hos P. atlantica gv. salmonicida identifisert og produsert rekombinant i Escherichia coli. Antigenisitet av rekombinante proteiner ble undersøkt ved Western blotting. Ett rekombinant YopT-lignende protein ble undersøkt for funksjon ved transfeksjon av cellelinjen BK (en hittil ubeskrevet cellelinje ved Fiskeimmunologigruppen ved Universitetet i Bergen). Isolat av P. atlantica gv. salmonicida og P. atlantica gv. cyclopteri ble dyrket ved ulike temperaturer for å undersøke hvordan temperatur påvirket vekst og proteinuttrykk. Studien avdekket at P. atlantica gv. salmonicida og P. atlantica gv. cyclopteri vokser best ved 20 - 30 °C. Videre ble in silico analyser benyttet for å etablere en liste over potensielle virulensfaktorer og immunogene komponenter hos P. atlantica gv. salmonicida. Basert på disse resultatene ble et YopT-lignende protein og et YadA-lignende protein valgt for å produseres rekombinant. Det YopT-lignende proteinet ble transfektert inn i BK-celler, for å undersøke cytotoksiske effekter gjennom ødeleggelse av aktinfilamentene i cellene. Bare det YopT-lignende proteinet ble vellykket produsert rekombinant i Escherichia coli og bundet til 6xHisTag-antistoff. Dette proteinet viste ingen reaktivitet mot serum fra P. atlantica gv. salmonicida vaksinert fisk, noe som indikerer at dette proteinet ikke representerer de primære antigenene som gjenkjennes av fiskens immunforsvar. Resultatene kunne ikke vise destruksjon av aktin i BK-celler transfektert med rekombinant YopT-lignende protein, heller ikke påvirkning i cellenes evne til fagocytose. Utfordringene knyttet til å analysere preparatene gjør at denne effekten likevel ikke kan utelukkes. Videre analyser er nødvendig for å ytterligere undersøke funksjonen til YopT-lignende protein i P. atlantica gv. salmonicida.

Pasteurellosis in Atlantic salmon (Salmo salar) caused by P. atlantica genomvar salmonicida has become a significant threat to fish welfare and sustainable production in Norwegian aquaculture since 2018. Therefore, there is a great need for a commercial vaccine against the disease. So far, there is limited knowledge about surface proteins, virulence factors, and immunogenic components of this bacterium. This knowledge is critical for the selection of vaccine isolates and strategies, and to understand how this bacterium causes disease. In this master's thesis, potential virulence factors and immunogenic components in P. atlantica gv. salmonicida were identified and produced recombinantly in Escherichia coli. The antigenicity of recombinant proteins was investigated by Western blotting. One recombinant YopT-like protein was examined for function by transfection of the BK cell line (a so far undescribed cell line at the Fish Immunology Group at the University of Bergen). Isolates of P. atlantica gv. salmonicida and P. atlantica gv. cyclopteri were cultured at different temperatures to investigate how temperature affected growth and protein expression. It was found that P. atlantica gv. salmonicida and P. atlantica gv. cyclopteri grow best at 20 - 30 °C. Furthermore, in silico analyses were used to establish a list of potential virulence factors and immunogenic components in P. atlantica gv. salmonicida. Based on these results, a YopT- like protein and a YadA-like protein were chosen to be produced recombinantly. The YopT- like protein was transfected into BK cells, to examine cytotoxic effects through the destruction of the actin filaments in the cells. Only the YopT-like protein was successfully produced recombinantly in Escherichia coli and bound to the 6xHisTag antibody. This protein showed no reactivity against serum from vaccinated fish with P. atlantica gv. salmonicida, indicating that this protein does not represent the primary antigens recognized by the fish's immune system. We could not demonstrate destruction of actin in BK cells transfected with recombinant YopT-like protein, nor an effect on the cells' ability to phagocytize. However, challenges associated with analyzing the preparations cause that this effect cannot be ruled out. Further analyses are necessary to learn more about the function of the YopT-like protein in P. atlantica gv. salmonicida.
