Development of a simple print-capture PCR method for extracting DNA from asparagus

1. DNA can be quickly extracted from asparagus cladodes at low cost by combining the print-capture method for sampling and grinding with the previously reported single-step DNA extraction method. 2. Filter paper impregnated with sap and then air-dried could be stored for 2 months at room temperature. Extracted DNA solutions were also stable and usable as PCR templates even after 1 month of storage at 5℃ 3. This extracted DNA can be used for PCR with a wide range of markers, including the common marker A01-900 with a PCR product size of 880 bp as well as the previously reported gender determination marker.
