Avaliação da atividade antifúngica do óleo essencial de Origanum Vulgare L. frente a fungos de importância em veterinária com ênfase em candida spp. | Evaluation of the antifungal activity of Origanum vulgare L. against important fungi in veterinary medicine with emphasis in Candida spp

O estudo teve como objetivos avaliar a ação antifúngica in vitro e in vivo do óleo essencial do Origanum vulgare (orégano), determinar a composição química dos óleos estudados e verificar a toxicidade em ratos Wistar. Inicialmente, oito amostras de O. vulgare foram extraídas por hidrodestilação e os óleos essenciais analisados por cromatografia gasosa, sendo posteriormente testados in vitro para escolha do óleo para os ensaios in vivo. Os testes in vitro seguiram as normas estabelecidas pelo CLSI-M27-A2, sendo utilizados sete isolados de Sporothrix schenckii e 16 isolados de Candida spp. A toxicidade do O. vulgare foi avaliada em 30 ratos wistar, fêmeas, tratadas via oral e vaginal com óleo a 3%, por 30 dias, através de parâmetros clínicos, hematológicos e histopatológicos. Para o teste in vivo, foram utilizados 48 ratos wistar, machos, distribuídos em quatro grupos: T1:óleo (1,5%); T2:óleo (3%); T3:fluconazol (10mg/Kg/dia); T4:controle negativo (emulsão); os óleos foram emulsionados com 0,001% de Tween 80. Para a reprodução experimental da candidíase sistêmica foi utilizado isolado canino e o inóculo (106céls/mL deC. albicans) foi administrado pela veia lateral da cauda em todos os animais. Os fármacos e o diluente foram administrados diariamente por via oral, com auxílio de sonda oro-gástrica, durante 30 dias. Foi realizada avaliação clínica semanal e pesagem dos animais, e após a necropsia foi realizada análise macroscópica, pesagem dos órgãos e retroisolamento de C. albicans. O óleo essencial do O. vulgare utilizado nos testes in vitro apresentou como componentes majoritários o 4-terpineol, carvacrol, timol e α-terpineol. A Concentração Inibitória Mínima (CIM) do O. vulgare para S. Schenckii foi de 0,25% (250μL/mL-1) para todos os isolados testados. Para Candida spp. os resultados foram expressos em CIM e CFM (Concentração Fungicida Mínima), obtendo-se CIM de 2,72μL mL-1 e CFM de 5μL mL-1 para C.albicans isolada de animais; CIM de 2,97μL mL-1 e CFM 3,54μL mL-1 para cepas padrões e CIM igual 2,10μL mL-1 e CFM igual 2,97μL -1mL para espécies não-albicans. A utilização do óleo essencial de O. vulgare a 3%, uma vez ao dia, 30 dias, por via oral e vaginal, não causou toxicidade nos ratos wistar. Na candidíase experimental sistêmica, os animais tratados com óleo a 3% e com fluconazol resultaram em menor freqüência de alterações clínicas. Não houve diferenças entre os grupos quanto a alterações macroscópicas nos órgãos, com exceção dos rins, onde o grupo T3 diferiu dos demais grupos (T1,T2,T4) sendo aquele com menor alterações. Quanto ao retroisolamento, foi obtido menor quantidade de Unidades Formadoras de Colônias no grupo T3 e maior no T4. Dessa forma é possível concluir que o óleo essencial do O. vulgare possui atividade in vitro frente ao S. schenckii e Candida spp. e que sua composição química interfere nos resultados. A utilização do óleo à 3%, via oral e vaginal por 30 dias não causou alterações toxicológicas relevantes em ratas Wistar. O uso do óleo a 3% demonstrou bons resultados no tratamento da candidíase experimental sistêmica, porém são necessários maiores estudos que possibilitem introduzir esta substância como opção terapêutica para candidíase.

The aims of this study were to evaluate the in vitro antifungal activity of the essential oil Origanum vulgare (oregano), to determine the chemical composition of the oils tested and to verify their toxicity to rats. Eight samples of Origanum were extracted by hydrodestilation, and the composition of the essential oils was determined by gaseous chromatography. In vitro antifungal sensitivity was performed with CLSI-M27-A2 protocol. Seven isolates of Sporothrix schenckii and 16 isolates of Candida species were tested. The animal model consisted of female wistar rats, which were treated orally and vaginally with 3% oil for 30 days. Animals were analyzed by clinical, hematological and hystopathologic parameters. For the in vivo test, 48 rats wistar were used, distributed in four groups: T1/n=12: oil 1,5%; T2/n=12:oil 3%; T3/n=12: Fluconazole 10mg/Kg; T4/n=12: Control (emulsion); the oils had been emulsified with 0,001% of Tween 80. The experimental disease was carried with one canine isolate, which was prepared with a suspension of C. albicans (106 céls/mL), and inoculated for the lateral vein of the tail. The drugs and diluent had been administered daily orally during 30 days. Weekly clinical evaluation and weighing of the animals were performed, and to at the end of experiment autopsies were done with macroscopic analysis, weighing of the tissues and retro isolation of C. albicans. The O. vulgare essential oil used in the tests in vitro showed 4-terpineol, carvacrol, timol and alfa-terpineol as majority component. The Minimum Inhibitory Concentration (CIM) of the O. vulgare for S. schenckii was 0,25% (250μL/mL), for all the isolates tested. For Candida spp. the results had been express in CIM and CFM (Concentration Fungicidal Minimum), showing CIM 2,72μL mL-1 and CFM 5μL mL-1 for C.albicans isolates from animals; CIM 2,97μL mL-1 and CFM 3,54μL mL-1 for standards strains and CIM 2,10 μL mL-1 and CFM 2,97μL -1mL for species non-albicans. The use of the 3% essential oil, daily for 30 days, orally and vaginally, did not cause toxicity in the rats. In systemic experimental candidiasis, animals treated with 3% oil and fluconazole showed lesser frequency of clinical alterations. Differences between the groups weren’t observed in the organs macroscopic alterations, with exception of the kidneys that presented more lesions in the group T3. The retro isolation was higher in the T4 group and in the group T3 was of minor. Thus, we conclude that the O. vulgare essential oil have activity in vitro against S. schenckii and Candida spp. and its chemical composition intervenes in the results. The use of the 3% oil for 30 days does not cause oral and vaginal significative alterations in wistar rats. The use of the 3% oil demonstrated good results in the treatment of systemic experimental candidiasis, however other studies are necessary to introduce this substance as a therapeutic option for candidiasis.
