Caracterização genética e química e atividade biológica do óleo essencial de populações naturais de Elionurus muticus Humb. & Bompl Ex Willd | Genetic and chemical characterization and biological Activity of essential oil of natural populations of Elionurus Muticus Humb. & Bompl Ex Willd

O mercado mundial de óleos essenciais vem apresentando um crescimento aproximado de 11% ao ano, sendo que o Brasil está entre os 15 maiores exportadores, correspondendo a 1% das exportações mundiais. Um dos compostos de óleos essenciais mais comercializados é o citral, que apresenta um forte odor cítrico e é empregado nas indústrias de perfumarias, alimentos e cosméticos. Elionurus muticus é uma gramínea nativa do bioma Pampa e tem se destacado pela produção de um óleo essencial rico em citral; entretanto, poucos trabalhos têm sido realizados com esta espécie. Os objetivos desse trabalho foram caracterizar morfologica, química e geneticamente populações naturais de E. muticus coletadas no RS, bem como avaliar a atividade biológica do óleo essencial e do extrato etanólico destas populações. As populações coletadas foram cultivadas em vasos e mantidas ao ar livre. As amostras de folhas, colmos e inflorescências para todas as análises foram obtidas destas plantas. A caracterização genética usou marcadores do tipo AFLP e a citogenética avaliou mitose, meiose e grãos de pólen. As análises químicas foram realizadas com as técnicas de cromatografia gasosa e espectrometria de massas. A atividade biológica foi avaliada com os fungos Botrytis cinerea, B. allii e Penicillium expansum; com o microcrustáceo Artemia salina e com diásporos de cebola e alface. A atividade de captura de radicais livre foi baseada no método DPPH. Os resultados demonstraram que as populações estudadas têm variabilidade genética para diferentes caracteres, apontando para a possibilidade de seleção de plantas com caracteres relevantes, como alto rendimento de óleo essencial e de citral. A alta variabilidade também sugere a ocorrência preferencial de alogamia nas populações naturais. Através de análises citogenéticas foi possível concluir que a meiose na espécie é regular e que o número cromossômico é de 2n=20. O óleo essencial de E. muticus pode ser utilizado no controle de fungos fitopatogênicos e no controle da germinação e desenvolvimento de outras plantas. Testes preliminares sugerem uma possível atividade citotóxica. Além disso, o extrato metanólico de E. muticus apresenta grande potencial como antioxidante.

The world market for essential oil is increasing about 11% per year, where Brazil is among the 15 largest exporters, accounting for 1% of world exports. One of the essential oil compounds most commercialized is the citral, which has a strong citrus odor and is used in perfumery, food and cosmetics industries. Elionurus muticus is a native grass of the Pampa biome and has been highlighted by the production of an essential oil rich in citral. However, few studies have been done on this species. The aims of this study were to characterize morphologically, chemically, and genetically natural populations of E. muticus collected in RS, as well as to evaluate the biological activity of the essential oil and the ethanol extract of these populations. Collected populations were cultivated in pots and maintained outdoors. Leaf, stem, and flower samples for all analyzes were obtained from these plants. Genetic characterization used AFLP markers and cytogenetics assessed mitosis, meiosis and pollen. Chemical analyzes were performed using the techniques of gas chromatography and mass spectrometry. The biological activity was evaluated with the phytopathogenic fungus Botrytis cinerea, B. allii, and Penicillium expansum, with the shrimp Artemia salina, and with onion and lettuce seeds. The scavenging of free radicals was based on DPPH. The results showed that the studied populations have genetic variability for several traits, indicating the possibility of plant selection for high essential oil’s yield and citral content. The high varibility also suggests the occurrence of allogamy in natural populations. Through cytogenetic analysis it was concluded that the species meiosis is regular and that the chromosome number is 2n = 20. The essential oil of E. muticus can be used for controlling pathogenic fungi and to supress germination and growth of other plants. Preliminary tests suggest a possible cytotoxic activity. Moreover, the methanol extract of E. muticus can be used to scavenge free radicals.
