Florescimento em aveia hexaplóide : análise fenotípica e molecular | Flowering time in hexaploid oat: phenotypic and molecular analysis

O florescimento exerce um grande efeito no sucesso reprodutivo e adaptativo das plantas. A resposta a sinais ambientais, tais como fotoperíodo e vernalização, representa uma estratégia reprodutiva importante utilizada pela aveia (Avena sativa L.) e outros cereais. Esta estratégia visa garantir que o florescimento ocorra na época mais favorável dentro da estação de crescimento. Os objetivos deste estudo foram avaliar a variação fenotípica em uma população de linhagens recombinantes de aveia derivada do cruzamento entre genitores contrastantes para o florescimento e caracterizar sequências de nucleotídeos de aveia associadas com genes de vernalização. Uma população de 108 linhagens, derivada do cruzamento entre ‘UFRGS 8’ (insensível ao fotoperíodo e à vernalização) e ‘UFRGS 930605’ (sensível ao fotoperíodo e à vernalização) foi utilizada. Os experimentos foram conduzidos em duas épocas de semeadura, nos anos de 2013 e 2014. O número de dias ao florescimento e a resposta à semeadura no cedo foram avaliados. Pares de primers específicos para o gene Vrn1 desenvolvidos a partir do alinhamento de sequências de trigo, cevada e azevém foram utilizados para amplificar e clonar sequências de aveia. A partir dos resultados do número de dias ao florescimento, as linhagens recombinantes foram classificadas como insensíveis, intermediárias e sensíveis a ambos sinais de fotoperíodo e vernalização. As linhagens recombinantes exibiram relação similar no número de dias ao florescimento, nas diferentes épocas de semeadura e anos avaliados. A resposta à semeadura no cedo apresentou valores negativos nos dois anos de avaliação para todas as linhagens e, associação negativa com o número de dias ao florescimento. A variação fenotípica do florescimento observada entre as linhagens recombinantes demonstrou a existência de diferentes mecanismos genéticos que controlam a resposta ao fotoperíodo e à vernalização. A análise molecular revelou a presença de pelo menos duas variações alélicas para o gene Vrn1 nos genitores UFRGS 8 e UFRGS 930605. Entretanto, estas variações alélicas precisam ser validadas quanto ao seu potencial de uso em estudos que visam o mapeamento molecular do gene Vrn1 em aveia.

Flowering time exerts a great effect in the reproductive and adaptive success of plants. The response to environmental cues, such as photoperiod and vernalization, represents an important reproductive strategy used by oat (Avena sativa L.) and other small grain cereals. This strategy aims to ensure that flowering occurs at the most favorable time within the growing season. The objectives of this study were to assess the phenotypic variation in a population of oat recombinant inbred lines derived from the cross between contrasting parental lines for flowering time and to characterize oat nucleotide sequences associated with vernalization genes. A population with 108 lines, derived from the cross between ‘UFRGS 8’ (insensitive to photoperiod and vernalization) and ‘UFRGS 930605’ (sensitive to photoperiod and vernalization) was used. The experiments were carried out in two growing seasons, over the years of 2013 and 2014. The number of days to flowering and the response to early sowing were evaluated. Specific primer pairs for the Vrn1 gene developed from sequence alignments of wheat, barley and ryegrass were used to amplify and clone oat sequences. Considering the number of days to flowering, recombinant inbred lines were classified as insensitive, intermediate, and sensitive to both photoperiod and vernalization cues. The recombinant lines exhibited a similar relation in the number of days to flowering in different sowing dates and years evaluated. The response to early sowing showed negative values in the two years of evaluation for all the lines and, negative association with the number of days to flowering. Flowering phenotypic variation observed among the recombinant lines demonstrated the existence of different genetic mechanisms controlling the response to photoperiod and vernalization. The molecular analysis revealed the presence of at least two allelic variations for the Vrn1 gene in the parental lines UFRGS 8 and UFRGS 930605. However, these allelic variations need to be validated as to its potential use in studies that aim the molecular mapping of the Vrn1 gene in oat.
