Uso estratégico de enzimas exógenas e probiótico para suínos : efeitos sobre metabolismo de energia e nutrientes e saúde intestinal | Strategic use of exogenous enzymes and probiotics for pigs: effects on energy and nutrients metabolism and intestinal health

O objetivo geral desta tese foi avaliar se o uso de β-mananase isolada ou em combinação com arabinofuranosidases e xilanases poderia melhorar a digestibilidade da dieta, o metabolismo dos nutrientes e da energia e saúde intestinal em suínos na fase de crescimento e em leitões pós-desmame. Assim como, avaliar se o fornecimento de um probiótico a matrizes gestantes e lactantes e/ou às suas progênies na fase de creche poderia melhorar a digestibilidade dos nutrientes, o metabolismo de energia e a saúde intestinal de leitões pós-desmame. Ao todo, três experimentos foram realizados para responder estes objetivos. O objetivo do primeiro estudo foi avaliar se a adição de βmananase isolada ou em combinação com um complexo de multi-carboidrases poderia aumentar a digestibilidade da dieta, metabolismo dos nutrientes e da energia e saúde intestinal de suínos em crescimento. Utilizou-se 24 suínos de 35,6 kg alimentados sem ou com a adição de β-mananase (BM; 300 g/ton) ou β-mananase mais uma mistura multienzimática de carboidrases (BM+MCC; 300 + 50 g/ton). A adição da BM e BM+MCC na dieta resultou em maiores (P<0.05) coeficientes de digestibilidade de matéria seca, proteína e energia, e coeficientes de metabolizabilidade de proteína (2,87 e 2,60%) e energia (2,61 e 1,44%) em comparação ao grupo sem adição das enzimas. A adição da BM e BM+MCC reduziu o nível de calprotectina fecal em 52 e 56% em relação ao grupo sem adição das enzimas. O objetivo do segundo experimento, foi avaliar a adição de βmananase isolada ou em combinação com um complexo de multi-carboidrases poderia melhorar a digestibilidade da dieta, metabolismo dos nutrientes e da energia e a saúde intestinal de leitões pós-desmame. Utilizou-se 30 leitões desmamados aos 28 dias (7,9 kg) alimentados sem ou com adição de β-mananase (BM; 300 g/ton) ou β-mananase mais uma mistura multienzimática de carboidrases (BM+MCC; 300 + 50 g/ton) após o desmame. A adição da BM na dieta melhorou (P<0.05) em 64 kcal/kg e o BM+MCC melhorou em 100 kcal/kg a energia metabolizável nos leitões. A adição da BM e BM+MCC às dietas também reduziu o nível de calprotectina fecal dos leitões em 29 e 46% em relação ao grupo controle. O objetivo do terceiro experimento foi avaliar a suplementação de um probiótico multi-especies em dietas para matrizes suínas em gestação e lactação e/ou em dietas de leitões na fase creche poderia melhorar a digestibilidade dos nutrientes e o metabolismo energético, bem como a saúde intestinal dos leitões na fase de creche. Assim, 32 leitões desmamados aos 21 dias (6,2 kg) foram divididos aleatoriamente em um arranjo fatorial 2 × 2, no qual foram consideradas a suplementação probiótica materna (ausente ou presente) na gestação-lactação e a suplementação probiótica no período de creche (ausente ou presente) durante 21 dias. Os leitões nascidos de matrizes alimentadas com probiótico apresentaram maior (P<0.05) digestibilidade e metabolizabilidade da energia, o que resultou em um aumento de 46 kcal no conteúdo de energia metabolizável da dieta. A suplementação com probióticos para as matrizes também reduziu (P<0.05) a permeabilidade intestinal das suas progênies em 16%. A suplementação de enzimas exógenas e probiótico melhoram a digestibilidade e metabolizabilidade dos nutrientes e a saúde intestinal de suínos.

Therefore, the general aim of this thesis was to assess whether the use of β-mannanase alone or in combination with a multi-carbohydrase complex, such as xylanase and arabinofuranosidases, could improve diet digestibility, nutrient metabolism, and intestinal health in post-weaned and growing pigs. In addition, assessing whether providing probiotics to gestation-lactating sows and/or their offspring in the nursery phase could improve the digestibility of nutrients, metabolism, and intestinal health of post-weaned pigs. Three trials were conducted to address these objectives. The first study aimed to evaluate whether the addition of β-mannanase alone or in combination with the multicarbohydrase complex could act additively to improve diet digestibility, nutrient and energy metabolism, and gut health in growing pigs. Twenty-four 35.6 kg of pigs were fed with or without β-mannanase (BM; 300 g/ton) or β-mannanase plus a multi-enzyme mixture of carbohydrases (BM + MCC; 300 + 50 g/ton). The addition of BM and BM+MCC resulted in higher (P<0.05) dry matter, protein, and energy digestibility coefficients, as well as protein (2.87 and 2.60%) and energy (2.61 and 1.44%) metabolizability coefficients, compared to the unadded group. The addition of BM and BM + MCC reduced the level of fecal calprotectin by 52 and 56%, respectively, compared to the unadded group. The second experiment aimed to assess whether the addition of β-mannanase alone or in combination with a multi-carbohydrase complex could improve diet digestibility, nutrient and energy metabolism, and intestinal health in post-weaned pigs. Thirty pigs weaned at 28 days of age (7.9 kg) were fed without any addition (control) or addition with βmannanase (BM; 300 g/ton) or β-mannanase plus multi-carbohydrase complex (BM+MCC; 300 + 50 g/ton) after weaning. BM improved metabolizable energy by 64 kcal/kg (P<0.05) and BM + MCC by 100 kcal/kg. The addition of BM and BM +MCC to the diets also reduced the level of fecal calprotectin by 29 and 46%, respectively, compared to the control group. The third experiment aimed to assess whether supplementation of a multi-species probiotic in diets for gestating and lactating sows and/or in diets for pigs in the nursery phase could improve the digestibility of nutrients and energy metabolism, as well as the intestinal health of pigs in the nursery phase. 32 pigs weaned at 21 days (6.2 kg) were randomly divided into a 2 × 2 factorial arrangement, which considered maternal probiotic supplementation (absent or present) during gestation-lactation, and probiotic supplementation during the nursery period (absent or present) for 21 days. Pigs born to sows fed probiotics showed greater (P<0.05) digestibility and metabolizable energy, which resulted in an increase of 46 kcal in the dietary metabolizable energy content. Supplementing sows with probiotics also reduced the intestinal permeability of their offspring by 16 % (p < 0.05). Supplementation with exogenous enzymes and probiotics in various strategies improves the digestibility and metabolizability of nutrients and the intestinal health of pigs.
