Morphometry, granulosa cells and nuclear chromatin of preantral follicles associated or not to vitrification of bovine ovarian tissue | Morfometria, células da granulosa e cromatina nuclear de folículos préantrais em associação ou não com a vitrificação de tecido ovariano bovino

إنجليزي. The objective of this study was to evaluate the effect of ovarian fragments vitrification in the morphometry, number of granulosa cells and nuclear  chromatin of preantral ovarian follicles of bovines. Ovarian was obtained at a local slaughterhouse and cortex fragments were removed. One third wereimmediately fixed and processed for histology (T0 - control). The remaining fragments were divided in two treatments, TI - toxicity test and TII -  vitrification. In TI the fragments were exposed to only cryoprotectants and fixed in Carnoy and in TII were utilized DMSO and ethylene glycol supplemented with sucrose. The results indicated that the primordial follicles when vitrified or exposed to cryprotectants did not show variation in morphometry and amount of cells of the granulosa (p > 0.05), exception to primary follicles (p<0.05) when compared to T0. The analysis of the nuclear chromatin of follicles vitrified or cryprotectants exposed showed significant difference (p<0.05) when compared to T0 with the control treatment, particularly in TII. In conclusion the vitrification technique affected negatively the morphology of bovine follicles enclosed in ovarian tissue.

البرتغالية. Busca-se no presente estudo, avaliar o efeito da vitrificação de fragmentos ovarianos na morfometria, número de células da granulosa e cromatina nuclear de folículos pré-antrais bovinos. Ovários foram obtidos em abatedouro e fragmentos do córtex retirados, enquanto uma parte foi fixada imediatamente e processada para histologia (T0 - controle). Outros fragmentos foram distribuídos em dois tratamentos: TI - toxicidade e TII – vitrificação; no TI, os fragmentos foram expostos apenas aos crioprotetores e fixados em Carnoy, e no TII, usou-se a associação dos crioprotetores DMSO e etilenoglicol acrescido de sacarose. Os resultados indicaram que folículos primordiais, quando vitrificados ou expostos aos crioprotetores, não apresentam variações morfométricas e na quantidade de células da granulosa (p > 0,05), exceção feita aos folículos primários (p < 0,05) quando comparados com o T0. A análise da morfologia da cromatina nuclear de folículos vitrificados ou apenas expostos aos crioprotetores, mostrou diferenças significativas (p < 0,05) quando  comparados com o T0; conclui-se, então, que a vitrificação causou alterações morfológicas de folículos bovinos inclusos em tecido ovariano.
