Molecular Characterization of Strongyle Nematodes of Goats in Western Uttar Pradesh of India

Strongyle infection in goats is responsible for economic losses due to reduced productivity and increased mortality. The current study’s goal is to molecular- based methods to determine the kind of strongyle eggs from naturally infected goats in four selected districts of Western Uttar Pradesh, including Meerut, Saharanpur, Baghpat, and Muzaffarnagar. Faecal samples were examined through direct faecal wet smears and flotation methods to identify strongyle - eggs. Between March 2023 to February 2024, 1117 goats faecal samples were taken from various villages in chosen district. Out of these samples, 360 were found to be positive for strongyle resulting in an overall prevalence of 32.22% in goats. The positive faecal samples were pooled separately and subjected to larval culture with DNA extraction performed on the larvae. Polymerase chain reaction amplification of the ITS-2 region of the rDNA, conducted for identification purposes confirming the presence of Haemonchus contortus and genus of Trichostrongylus and Oesophagostomum. The prevalence rates of Haemonchus contortus, Trichostrongylus spp. and Oesophagostomum spp. prevalence rates were recorded as 54.16%, 31.66% and 14.16%. The finding of this study indicated that Haemonchus contortus was predominant among the strongyle spp. The pathogenicity of Haemonchus contortus was significant due to its blood sucking nature, leading to clinical signs such as anemia, submandibular edema and lethargy. There is a need to develop prevention and control strategies for the strongyle infections in goats in the study area.
