Validation of an HPLC/PDA Quantification Method for Vanillic Acid from the Leaf and Stem of Curcuma longa

This study developed an analytical method to identify and quantify vanillic acid (VA), a compound that belongs to the hydroxybenzoic acid group, in Curcuma longa (CL) using reversed-phase high-performance liquid chromatography. The analyses were conducted employing a Sunfire® C18 column and a gradient elution system coupled with a photodiode array detector , and after optimizing the parameters, the method was validated. Results indicated that the limits of detection and quantification for VA are 0.170 and 0.515 µg/mL, respectively. The calibration curves showed a strong linear relationship (r² = 1.0000) over the concentration range of 1 to 16 µg/mL. In the accuracy analysis, we achieved a high recovery rate (95.3–107.2%), while maintaining precision values below 1.28 (% RSD). These findings substantiate the suitability of our analytical method for the precise identification and quantification of VA in CL. Moreover, our methodology holds promise for the determination of hydroxybenzoic acid derivatives in routine analyses and large-scale extraction processes.
