Acclimatization and Hardening of Oil Palm Materials Obtained by Techniques of Plant Tissue Culture | Aclimatación y endurecimiento de materiales de palma de aceite obtenidos mediante técnicas de cultivo de tejidos vegetales

The conditioning of plantlets obtaining by plant tissue culture techniques is a transitional stage between the laboratory and the field, which objective is to keep the seedlings from an in vitro to ex vitro conditions. During this stage exists a gradual return to autotrophic functioning and a recovery of normal physiological and morphological characteristics. Cenipalma has standardized a methodology of propagation oil palm by plant tissues culture, which opens the possibility of delivering oil palm clonal materials to the Colombian palm cultivation. However, one of the bottlenecks of the cloning process might be the acclimatization and hardening stages that could be developed for palm growers to have clonal plants in their plantations. Therefore, in the Experimental Field Palmar de la Vizcaina - CEPV (Barrancabermeja) a methodology was adapted for acclimatization of clones as final stage of plant tissue culture propagation, with the future aim of training palm growers interested in planting clones, so they can carry out this acclimatization on their plantations. In this experiment we managed to obtain percentages of acclimatization over 85% in the seedlings classified as type 3 laboratory (good looking, good number of roots and leaves, and larger than 10 cm) and type 2 (intermediate appearance seedlings). Likewise, similar values were obtained for some type seedlings showed abnormalities AC, Curved Double, Double and Triple. Also, the behavior in pre-nursery demonstrated the importance of the quality of the root for the survival of the plant in this stage.

El acondicionamiento de plántulas, obtenidas por técnicas de cultivo de tejidos, es una fase transitoria entre el laboratorio y el campo, cuyo objetivo es llevar las plántulas desde un cultivo in vitro a condiciones ex vitro. Durante esta etapa se produce un retorno gradual al funcionamiento autotrófico, así como la recuperación de las características morfológicas y fisiológicas normales. Cenipalma ha estandarizado la metodología de propagación de la palma de aceite por tejidos vegetales in vitro, lo cual abre la posibilidad de entregar a la palmicultura nacional materiales clonales. Sin embargo, uno de los cuellos de botella de la clonación es esta fase de aclimatación y endurecimiento, que puede ser desarrollada por los palmicultores que vayan a sembrar plantas clonales en sus plantaciones. Por lo tanto, en el Campo Experimental Palmar de la Vizcaína - CEPV (Barrancabermeja), se adaptó la metodología para la aclimatación de clones como fase final del proceso de cultivo de t ejidos in vitro, con el ánimo de, en el futuro, capacitar a los palmicultores interesados en sembrar clones para que puedan llevar a cabo esta aclimatación en sus plantaciones. Se logró obtener porcentajes de aclimatación por encima de 85% en plántulas clasificadas a la salida del laboratorio como tipo 3 (con buena apariencia, buen número de raíces y de hojas, y de tamaño mayor a 10 cm) y tipo 2 (plántulas de apariencia intermedia). De igual forma se obtuvieron valores similares para algunas plántulas que presentaban anormalidades tipo AC, Curved Doble, Doble y Triple. Asimismo, el comportamiento en previvero demostró la importancia de la calidad de la raíz para lograr la supervivencia de la planta en esta fase.
