Determining the Effect of Different EMS (Ethyl Methane Sulphonate) Doses on Safflower | Aspir’de Farklı EMS (Ethyl Methane Sulphonate) Dozlarının Etkisinin Belirlenmesi

The basic principle in mutation breeding is to identify and select the ones that are suitable for the purpose among the negative and positive variations that will occur with different mutagen doses applied by different methods. Creating hereditary differences that will increase variation in desired qualities depends on the selection of appropriate chemicals, their use in certain doses, and their effectiveness. This study was conducted in Dicle University Faculty of Agriculture Biotechnology laboratory and greenhouse conditions. The aim of the study is to determine the effect of different EMS doses on the properties examined in safflower. Seeds were treated with 0 (control), 10, 20, 30, 40, 50, 60, 70, 80, 90, 100 mM EMS. Seeds treated with EMS were subjected to a germination trial in 4 replications according to the random plot trial plan, with 20 seeds in each petri dish. Under greenhouse conditions, 20 seeds were sown in viols in 4 replicates and characteristics such as emergence rate, root length, seedling height, first leaf length, seedling fresh and dry weight were examined. As a result of the examined features, it was determined that there was a significant difference between the doses, and it was concluded that the highest values in all features were in the control group. It has been reported that increasing EMS doses cause a decrease in the examined property values, and the lowest values and deaths are obtained from the maximum dose of 100mM.

Mutasyon ıslahında temel ilke, farklı yöntemlerle uygulanan değişik mutagen dozlarının ortaya çıkaracağı olumsuz ve olumlu varyasyonlar içerisinden amaca uygun olanların tespit edilip seçilmesidir. İstenilen niteliklerde varyasyonu arttıracak kalıtsal farklılıkları oluşturabilmek, uygun kimyasalların seçimi belirli dozlarda kullanılabilmesine ve etkinliğine bağlıdır. Bu çalışma, Dicle Üniversitesi Ziraat Fakültesi Biyoteknoloji laboratuvarında ve sera koşullarında yürütülmüştür. Çalışmanın amacı farklı EMS dozlarının aspirde incelenen özellikler üzerindeki etkisini belirlemektir. Tohumlara 0 (kontrol), 10, 20, 30, 40, 50, 60, 70, 80, 90, 100 mM EMS uygulaması yapılmıştır. EMS uygulaması yapılan tohumlar her petri kabında 20 tohum gelecek şekilde tesadüf parselleri deneme planına göre 4 tekerrürlü olarak çimlendirme denemesine alınmıştır. Sera koşullarında ise viyollere 4 tekerrürlü olarak 20 adet tohum ekilmiş ve çıkış oranı, kök uzunluğu, fide boyu, ilk yaprak uzunluğu, fide yaş ve kuru ağırlığı gibi özellikler incelenmiştir. İncelenen özellikler neticesinde dozlar arasında önemli denecek derecede farklılık olduğu belirlenirken, tüm özelliklerde en yüksek değerlerin kontrol grubunda olduğu sonucu ortaya konulmuştur. Artan EMS dozlarının incelenen özellik değerlerinde düşüşe neden olduğu, en düşük değerlerin ve ölümlerin en fazla 100mM dozundan elde edildiği sonucu rapor edilmiştir.
