Common pathogens diagnosed in pig samples from year 2014 to 2017 by Veterinary Research Institute

A total of 23,322 specimens collected between 2014 and 2017, froma total of 2,592 cases, were received in Veterinary Research Institute, Ipoh (VRI) from various states in Malaysia and testedfor common bacterial, viral, and parasitic diseases in pigs. The highest occurrence of isolated bacteria from 771 samples whichtested positive were Salmonella (47.38%) and Escherichia coli (15.68%), followed by Staphylococcus (6.62%), Streptococcus (5.57%), Klebsiella pneumonia (4.88%), Pseudomona (3.38%), Acinetobacter (3.14%), Aeromonas (2.79%), Enterobacter (2.44%), one each of Bacillus and Pasteurella multocida (1.74%), Enterococcus (1.39%) and Corynebacterium (1.05%). 1.74% of each bacteria detected were Moxarella, Aspergillus, Burkholderia andChromobacterium. Positive samples tested by ELISA was Japanese encephalitis virus (JEV) (9.15%), Aujezsky disease virus (ADV)(5.37%), porcine cirvo-virus-2 (PCV2) (5.09%) and porcine reproductive and respiratory syndrome virus (PRRSV) (4.52%). Positive amples tested by the molecular test wasPCV2 (1.62%), PRRSV (1.32%) and classical swine fever virus (CSFV) (0.4%). Serology tests were conducted on 11,305 samplesand reported positive for Brucella suis (15.32%), Brucella abortus (0.62%), Brucella melitensis (0.85%), and melioidosis (0.05%). Parasitology analyses on 99 samples. revealed presence of 10.1% coccidia and 1% each of helminths and Sarcocystis. Within the 4-year period, there were no positive samples for porcine parvovirus (PPV), Nipah virus, swine influenza virus (SIV), and bacteria of Johne’s disease and leptospirosis. Continuous assessment is required to establish a comprehensive baseline data of swine diseases in Malaysia.

Infectious diseases of livestockare a major threat to global animal health and welfare and their effective control is crucialfor agronomic health, for safeguarding and securing national and international food supplies and for alleviating rural povertyin developing countries. Some devastating livestock diseases are endemic in many parts of the world and threats from old and new pathogens continue to emerge, with changes to global climate, agricultural practices and demography presenting conditions that are especially favourable for the spread of arthropod-borne diseases into new geographical areas. Zoonotic infections that are transmissible either directly or indirectly between animals and humans are on the increase and pose significant additional threats to human health and the current pandemic status of new influenza A (H1N1) is a topical example of the challenge presented by zoonotic viruses (Tomley and Shirley, 2009). Malaysia, being one of the members of the World Organisation forAnimal Health (OIE) which is responsible for setting standards for control of animal diseases. For year 2017, the list included 116 animal diseases, infections and infestations, many of which are zoonotic in nature. As such, this paper discusses the commonzoonotic infections diagnosed in the five Regional Veterinary Laboratories which are spread across the country and entrustedto carry out diagnostic tests to aid in the treatment and control of animal diseases. A total of almost half a million samples weretested comprising more than a million tests to help the Department of Veterinary Services control and eradicate economically important diseases to safeguard the animal population. Of these, zoonotic diseases comprise a small but significant entity which needs careful attention (Chandrawathani et al., 2017) Dora Tan (1981) reported that among the many zoonotic diseases prevalent in Malaysia, are leptospirosis, rabies, influenza, Japanese encephalitis, toxoplasmosis,ornithosis, Q fever and monkeypox which have been investigated at the lnstitute for Medical Research, Kuala Lumpur. The regional laboratories have full capability to conduct tests to confirm parasitic, viral and bacterial infections except for rabies andavian influenza, which was diagnosed in the Veterinary Research Institute. However, preliminary tests for avian influenza wascarried out in regional laboratories.

Chromobacterium violaceum is a facultative anaerobic, gram-negative rodshaped bacteria normally found in soil and stagnant water of tropical and subtropical countries. Infections of Chromobacterium violaceum are rare among mammals, but the first human infection were reported in Malaysia in 1927. This clinical case reports two separate cases of Chromobacterium violaceum infection in two sub-adult male Black-handed Gibbon (Hylobates agilis). Both animals were presented with the history of diarrhea, pyrexia and inappetance. From the physical examination, the findings were high body temperature, dull, inactive, soft fecal stained at the rectum and small open wound at lower limb of one of the case. The treatment given was antipyretic and multivitamin. Unfortunately, due to poor prognosis both patients died within 48 hours after presentation and treatment. Autopsy examinations wereperformed to find out the cause of death. Post mortem examination findings revealed hepatomegaly with multiple size whitish-yellow spots on the liver surface, congestion and enlargement of spleen and lymph node, congestion of lung and loss of demarcation between renal cortex and medulla of the kidney. The cellular changes via histopathological findings of kidney, spleen, lung and liver were suggestive of septicaemia. The gross and histopathologicalfindings were supported by the isolation of Chromobacterium violaceum via bacterial isolation and identification from lung, liver, spleen and kidney. Thus, the cause of death of the two sub-adult male Black-handed gibbon in this case are due to septicaemiadue to Chromobacterium violaceum infection.

Bovine haemoparasites are cosmopolitan in distribution and are known to cause substantial losses to the cattle industry. In spite of their economic importance, there remains a dearth of information on their molecular epidemiology in many parts of the world including Malaysia. To ascertain the molecular prevalence and species co-infection of bovine haemoparasites in the country, blood samples were collected from 1,045 heads of beef and dairy cattle on 43 farms from six geographical zones throughout Peninsular Malaysia. Samples subjected to PCR amplification of parasite species-specific genetic fragments revealed that Anaplasma marginale was the most prevalent haemoparasite (72.6%),followed by Theileria orientalis(49.8%),Candidatus Mycoplasma haemobos ( 47. 0 % ),Babesia bovis(32. 5%), Babesia bigemina (30.5%) and Trypanosomaevansi(17.9%). A high percentage (92.1%) of cattle was infected with either one or more haemoparasites. Triple haemoparasite species co-infection was the most prevalent (25.6%), followed closely by double species co-infection (25.1%). The most common (8.8%) and significantly correlated(rs= 0.250; p<0.01) combination was A. marginale+ T.orientalis. The present study constitutes the first attempt in the country to document the molecular prevalence and species co-infection of bovine haemoparasites over a wide spatial distribution. The data obtained will facilitate treatment, control and prevention measures to improve the local cattle industry.

A total of 65 Pasteurella multocida were isolated and identified from various animal’s samples received by Veterinary Research Institute (VRI) during the period of 2014 to 2016. These animals comprises of cattle, goat, pig, chicken, duck and rabbit. The serogroup of Pasteurella multocida were carried out using designation system of Carter’s capsular typing and molecular serogrouping method. Based on cases submitted to VRI, the prevalence of pasteurellosis in Malaysia ranging from 1.0% to 3.2% (2014 to 2016). It is low compared to previous reports and the pattern of predominant serogroups and animal hosts were found to be changing every year. In 2014, 80% (12/15) of the isolates were Pasteurella multocida Carter’s type D where all were isolated from goats. In 2015, the predominant serogroup changed to Pasteurella multocida Carter’s type A with a prevalence rate of 40.6% (13/32) which were mostly isolated from duck and cattle. While for Pasteurella multocida Carter’s type D, the prevalence in 2015 reduced to 21.9% (7/32) compared to the previous year and it was isolated from various animal species. Interestingly, in 2015 there was one isolate of Pasteurella multocida Carter’s type B isolated from goat with no reported history of outbreak. In 2016, the prevalence of Pasteurella multocida Carter’s type A increased to 72.2% (13/18), with a high percentage (92.3%) infection in young calves showing clinical signs with high mortality and morbidity in infected farms. Furthermore, during these 3 years of study, 3 isolates of Pasteurella multocida serogroup F were also identified each from pig, goat and chicken, respectively. In conclusion, this study revealed that pasteurellosis had become sporadic in Malaysia and the distribution of serogroups were diverse in all species of animal with no definitive host.

Septicaemic pasteurellosis is a fatal, sometimes epidemic, bacterial disease of domestic and wild animals including deer, bison, elk, and pronghorn antelope caused by Pasteurella multocida. This is the case report of septicaemic pasteurellosisin a captive sambar deer. The carcass was sent from Royal Endurance Stable, Bachok, Kelantan to the Kota Bharu RegionalVeterinary Laboratory for post-mortem. Gross examination of organs was followed by collection of specimens from lung, kidney,liver, spleen and heart for histopathology and bacterial examination. Pooled organ samples with rumen content were collected and sent to the nearest Chemistry Department for investigation. For histology, the liver, lung, spleen, kidney, and heart specimens were fixed in 10% neutral formalin, and routinely embedded in paraffin. Fivemicrometer sections were stained with H&E. Other tests such as worm and ectoparasiteidentification were conducted to identify the parasites. Post-mortem lesions revealed generalised haemorrhage in the organs.Pasteurella multocida serogroup B and E. coli were isolated from multiple tissues of the animal. Histological examination alsorevealed severe congestion and haemorhage of multiple tissues with infiltration of the inflammatory cells. The most likely mode of transmission of these bacteria is through an infected wound and into the bloodstream, thereby causing severe septicemia and death to the animal.

The small ruminant industry in Malaysia is plagued by several significant problems related to health and management. Due to the hot, wet tropical climate, parasitic, bacterial and viral diseases are rampant and conveniently spread and enhanced by poor management. The Veterinary Research Institute has initiated a working group to
identify and control critical diseases in small ruminants as an educational cum advisory programme for farmers on the lower economic rung so that productivity and profits may be heightened. Farms are attended to base on the complaint of high mortality and morbidity, whereby a complete and holistic approach to disease control is taken. All the farms sampled were in the vicinity of the laboratory
that is within 150 km radius. Samples collected were faeces, blood (serum and whole blood) and nasal swabs as well as water and feed samples. A questionnaire on the management practices as well as
history of the farm was recorded. Tests on all samples were conducted according to routine procedures. Results from this study
on 7 farms involving 175 goats and sheep revealed that the common parasitic diseases encountered are helminthiasis, coccidiosis and blood protozoans. More than 80% of the animals showed strongyle and coccidial infections. The prominent bacterial infections are Caseous Lymphadenitis (34.3%), Brucellosis (Brucella melitensis), Leptospirosis (1.7%), Melliodosis and QFever whereas the viral diseases are Caprine Arthiritis Encephalitis (1.1%) and Blue Tongue (15.4%). Early detection for these diseases is critical so that production losses can be minimised. Poor nutrition and hygiene is a contributing factor for the high incidence of disease in small
ruminants. Farmer awareness on latest farming methods and services provided also contributes to general wellbeing of animals. One of the outcomes of this programme is to create awareness and getting farmers to be independent and confident in problem solving with regards to management and disease in their herds.
