Single nucleotide polymorphisms in the bovine Histophilus somni genome; a comparison of new and old isolates

Histophilus somni, a causative agent of the bovine respiratory disease complex, can also cause a variety of systemic disorders, including bronchopneumonia, myocarditis, pericarditis, arthritis, pleuritis, and infectious thrombotic meningoencephalitis. The purpose of this study was to determine if currently circulating strains differ from those of the 1980s by identifying genomic changes. Single nucleotide polymorphisms (SNPs) and insertion and deletion (INDEL) sites were examined by whole-genome sequencing in 12 samples, 6 old and 6 new. The 31 028 SNP/INDELs recorded were compared against the reference genome sequence of the pathogenic H. somni strain 2336. The distribution of about 75% of these SNPs within a specified gene differed between old and new isolates and did not follow any particular pattern. The other 25% clustered into 2 groups containing the same SNPs in various genes: group I included 5 old isolates and 1 new isolate; group II included 5 new isolates and 1 old isolate. For putative virulence genes there were more SNPs in group I compared with strain 2336, itself an older isolate, than in group II. Although only 25% of all the SNPs formed 2 clusters, the results suggest some genetic difference in various genes between old and new strains.

Objective—To characterize the impact of Mannheimia haemolytica infection on feed intake and weight gain in feedlot heifers and to evaluate the clinical efficacy of isoflupredone acetate administered in combination with oxytetracycline. Animals—96 weanling heifers in a research feedlot facility. Procedures—Bronchopneumonia was induced by intrabronchial infusion of M haemolytica. Control heifers underwent a sham procedure. Infected heifers were treated with oxytetracycline alone or in combination with isoflupredone acetate (OXY-ISO) or with nothing. Clinical variables were recorded daily for 7 days following disease induction, and feedlot performance indices were measured over a 12-week period. Results—Infection caused a reduction in dry-matter intake and average daily gain (ADG) in heifers that received no treatment. Oxytetracycline treatment alone did not prevent reductions in feed intake and ADG during the first week after infection was induced, whereas OXY-ISO treatment did prevent these reductions. Treatment with OXY-ISO also resulted in faster clinical improvement. No significant differences were evident between the oxytetracycline and OXY-ISO groups with respect to dry-matter intake or ADG throughout the study period. Conclusions and Clinical Relevance—Isoflupredone acetate appeared to be a useful clinical adjunct to treatment with oxytetracycline in cattle with acute M haemolytica bronchopneumonia.

Objective-To validate a radioimmunoassay for measurement of procollagen type III amino terminal propeptide (PIIINP) concentrations in canine serum and bronchoalveolar lavage fluid (BALF) and investigate the effects of physiologic and pathologic conditions on PIIINP concentrations. Sample Population-Sera from healthy adult (n = 70) and growing dogs (20) and dogs with chronic renal failure (CRF; 10), cardiomyopathy (CMP; 12), or degenerative valve disease (DVD; 26); and sera and BALF from dogs with chronic bronchopneumopathy (CBP; 15) and healthy control dogs (10 growing and 9 adult dogs). Procedure-A radioimmunoassay was validated, and a reference range for serum PIIINP (S-PIIINP) concentration was established. Effects of growth, age, sex, weight, CRF, and heart failure on S-PIIINP concentration were analyzed. In CBP-affected dogs, S-PIIINP and BALF-PIIINP concentrations were evaluated. Results-The radioimmunoassay had good sensitivity, linearity, precision, and reproducibility and reasonable accuracy for measurement of S-PIIINP and BALF-PIIINP concentrations. The S-PIIINP concentration reference range in adult dogs was 8.86 to 11.48 microgram/L. Serum PIIINP concentration correlated with weight and age. Growing dogs had significantly higher S-PIIINP concentrations than adults, but concentrations in CRF-, CMP-, DVD-, or CBP-affected dogs were not significantly different from control values. Mean BALF-PIIINP concentration was significantly higher in CBP-affected dogs than in healthy adults. Conclusions and Clinical Relevance-In dogs, renal or cardiac disease or CBP did not significantly affect S-PIIINP concentration; dogs with CBP had high BALF-PIIINP concentrations. Data suggest that the use of PIIINP as a marker of pathologic fibrosis might be limited in growing dogs.

To evaluate the relationship between pneumonic lesions and distribution of bacteria, lungs from calves inoculated with Pasteurella multocida were examined histologically by use of immunoperoxidase technique. Pneumonic lesions fundamentally consisted of bronchopneumonia with fibrinopurulent pleuritis. The lesions were confirmed to be associated with inoculated P multocida, using the immunoperoxidase technique. The P multocida antigen was detected not only in the bacterial clusters in the lesions, but also in the cytoplasm of infiltrating neutrophils and macrophages. Further, immunoelectron microscopy indicated that the inoculated bacteria generally were phagocytosed and digested by neutrophils.

Objective: This study was carried out to assess the pathogenicity of local isolates of Mycoplasma ovipneumoniae and M. arginini in West African dwarf goats (kids) in Nigeria. Materials and methods: A total of 22 goats aged less than 1-year were purchased from markets. The goats were divided into six groups comprising of four experimental groups (EG; 4 in each) and two control groups (CG; 3 in each). The goats were fed ad libitum with standard diets and safe water. Groups EG1 and EG2 were infected with M. ovipneumoniae through trans-tracheal (TT) and intravenous (IV) routes, respectively, while those in groups EG3 and EG4 were infected with M. arginini through the same routes. Goats in groups CG1 and CG2 were inoculated with sterile Mycoplasma broth through TT and IV routes, respectively. In all cases, the amount of bacteria inoculated was 1.5x108 cells/mL. After the onset of the disease in goats, re-isolation of Mycoplasma was performed by culturing on mycoplasma agar supplemented with mycoplasma supplement G. The goats were monitored for 14 days post-infection (PI) to observe respiratory signs and mortality. Post-mortem (PM) examination was performed on each animal that died, while one surviving goat from each of the groups was sacrificed at 14 days PI for PM. After PM, histopathology was performed to observe the changes in tissues. Results: Cough and nasal discharges were observed in all the experimentally infected goats seven days PI. Mortalities were recorded in goats in EG1 (two goats), EG2 (one goats), EG3 (two goats) and EG4 (one goat). At PM, pneumonic lesions were observed in the lungs of all the experimentally infected goats. Conclusion: This study provides evidence that the local isolates of M. ovipneumoniae and M. arginini strains are pathogenic for goats in Nigeria. [J Adv Vet Anim Res 2016; 3(3.000): 242-251]

The bronchopneumonia of calves represents a risk to national supply chain because it is an ecopathy and weakens the more intensive production systems. It is characterized by inflammatory changes in the bronchi, bronchioles, lung parenchyma, and pleura. It is a disease of multifactorial traits called Bovine Respiratory Disease (BRD). The association of infectious agents with host defense and management to which the animal is subjected leads to the emergence of major clinical manifestations of the disease. The clinical evolution of BRD can also have serious secondary changes such as pulmonary edema, sepsis, and pulmonary hypertension, or even be consequent to the involvement of other structures, such as in cases of myocarditis leading to congestive heart failure. Although this report refers to a non-experimental framework, the circumstances that caused the calf to be subjected to a protocol-specific respiratory assessment involving non-routine reviews has made it possible to associate circulatory and respiratory conditions, rarely considered in ruminant clinic. The focus of this report was pulmonary edema. Modern clinical vision requires of the veterinarian work with cost-benefit relation, so that the more accurate and the earlier the clinical diagnosis the less expensive the treatment.
