Enhancer of zeste homologue 2 (EZH2) is the human homologue of Drosophila zeste gene enhancer. The aim of this study was to determine the expression of EZH2 in canine mammary carcinomas (CMCs) and its relationship with clinicopathological features. The expression of EZH2 mRNA and protein in 53 CMC tissue and 8 normal mammary gland tissue samples was measured by quantitative real-time PCR and immunohistochemical staining assay, respectively. The relationship between EZH2 protein expression and clinicopathological features was analysed by χ2 test to further explore the clinical significance of EZH2 in CMCs. Compared with normal mammary gland tissues, EZH2 mRNA expressions were significantly increased in CMC tissues (P < 0.01). Moreover, normal mammary glands did not express the EZH2 protein but carcinomic glands did, and expression increased in CMCs with high histological grades, especially in histological grade II (P < 0.05). However, EZH2 expression was not related to age, tumour size, or metastasis (P > 0.05). The expression of EZH2 in one type of CMC was not significantly different from the expression in any other type (P > 0.05). EZH2 is highly expressed in CMCs, indicating that it can be used as a molecular marker for early diagnosis, prognosis, or therapy of CMCs.

The value of neutrophil gelatinase-associated lipocalin (NGAL), kidney injury molecule-1 (Kim-1), and liver-type fatty acid binding protein (L-FABP) was assessed in early diagnosis of gentamicin-induced acute kidney injury (AKI) in dogs. Subcutaneous gentamicin injection in 16 healthy adult beagles made the AKI model. Blood was sampled every 6 h to detect NGAL, Kim-1, L-FABP, and serum creatinine (SCr) concentrations. Kidney tissue of two dogs was taken before the injection, as soon as SCr was elevated (78 μmol/L), and when it had risen to 1.5 times the baseline, and haematoxylin-eosin staining and transmission electron microscopy (TEM) were used to observe changes. NGAL, Kim-1, and SCr levels were significantly increased (P < 0.05) at 18, 30, and 78 h post injection, but L-FABP concentration was not associated with renal injury. At the earliest SCr elevation stage, findings were mild oedema, degeneration, and vacuolisation in renal tubular epithelial cells in pathology, and mild cytoplasmic and mitochondrial oedema in TEM. At this time point, NGAL and Kim-1 concentrations were significantly increased (P < 0.05), indicating that these two molecules biomark early kidney injury in dogs. Using receiver operating characteristic curve analysis, their warning levels were > 25.31 ng/mL and > 48.52 pg/mL. Plasma NGAL and Kim-1 above warning levels are early indicators of gentamicin-induced AKI in dogs.

In recent years, the high sensitivity and specificity of novel miRNA biomarkers have been utilised for early diagnosis and treatment monitoring of various diseases. Previous reports showed that abnormal expression of miR-208 in mice resulted in the development of an aberrant cardiac conduction system and consecutive arrhythmias. On the other hand, a study on infarcted human heart tissue showed upregulation of miR-208a in subjects with ventricular tachyarrhythmias compared to healthy controls. We prospectively investigated the expression of miR-208a and -208b in the serum of dogs presenting different cardiac arrhythmias. A total of 28 dogs with atrial fibrillation (n = 8), ventricular premature contractions (n=6), conduction system disturbances (n = 7), and free of heart conditions (as controls) (n = 7) were enrolled in the study. Total RNA was extracted from serum samples and miR-208a and -b, miR-16 as well as a cel-miR-39-5p spike-in were analysed with qPCR and ddPCR. miR-208a and miR-208b were not expressed in any of the samples. The calculated ddPCR miR-16 relative expression (normalised with cel-miR-39 spike-in) showed a good correlation (r = 0.82; P < 0.001) with the qPCR results. This outcome warrants further investigation, possibly focusing on tissue expression of miR-208 in the canine heart.

2016
Sainulabeen, Anoop | Nair, Sudheesh | Satheesan, Karthika | Devanand
A 2-year-old (63 days) pregnant female dachshund dog was presented to the Teaching Veterinary Clinical Complex (TVCC) with complaint of a progressively growing swelling in the caudal ventral abdominal area. History, physical examination, radiography and ultrasonography confirmed an inguinal hystrocoele. A ventral midline incisionon the mass was made to relieve the foetuses and reduction of contents, and herniorrhaphy was performed. Sutures were removed on 7th post-operative day and the animal had an uneventful recovery.

Specific contrast ultrasound is widely applied in diagnostic procedures on humans but remains underused in veterinary medicine. The objective of this study was to evaluate the use of microbubble-based contrast for rapid ultrasonographic diagnosis of thrombosis in small animals, using male New Zealand white rabbits (average weight about 3.5 kg) as a model. It was hypothesized that the use of microbubble-based contrast agents will result in a faster and more precise diagnosis in our model of thrombosis. A pro-coagulant environment had been previously established by combining endothelial denudation and external vessel wall damage. Visualization of thrombi was achieved by application of contrast microbubbles [sterically stabilized, phospholipid-based microbubbles filled with sulfur hexafluoride (SF6) gas] and ultrasonography. As a result, rapid and clear diagnosis of thrombi in aorta abdominalis was achieved within 10 to 30 s (mean: 17.3 s) by applying microbubbles as an ultrasound contrast medium. In the control group, diagnosis was not possible or took 90 to 180 s. Therefore, sterically stabilized microbubbles were found to be a suitable contrast agent for the rapid diagnosis of thrombi in an experimental model in rabbits. This contrast agent could be of practical importance in small animal practice for rapid diagnosis of thrombosis.
اظهر المزيد [+] اقل [-]Accuracy of transvaginal ultrasonography for early pregnancy diagnosis in Attappady black goats
2017
Philip, Laiju M. | Abhilash, R. S. | Francis, Bastin P.
Transabdominal ultrasonography has been popularly used in animal reproduction for the assessment of pregnancy status and foetal viability where as transvaginal method for pregnancy diagnosis is rarely used for pregnancy diagnosis in goats. The study was conducted in 74 Attappady Black goats from Government Goat Farm, Attappady and Livestock Research Station, Thiruvizhamkunnu to evaluate the accuracy of trans-vaginal methods and to identify the fetal characteristics from day 20 to 75. Transvaginal ultrasonographic examination was performed using endocavity transducer with frequency of 6.5 to 8 MHz. Observations were made in all the pregnant does at three different stages of pregnancy i.e., 20-35 days, 35-55 days and 55-75 days. Does were diagnosed as pregnant from the observation of gestational sac, embryo or foetus, embryonic or foetal heartbeat, placentomes or foetal skeleton. Sensitivity, specificity, positive predictive value, negative predictive value and overall accuracy along gestation period were 98.03, 92.3, 96.15, 96 and 95.62%, respectively. It is concluded from the present study that transvaginal ultrasonography can be used for herd pregnancy diagnosis at early stages from day 20 to 45 of pregnancy diagnosis in goats.
