Immunophenotyping of leukocytes in bovine placenta

Lymphocytes and macrophages are the main white cells involved in fetal-maternal tolerance. Little is known about these leukocytes in bovine placenta, such as the quantity and location of these cells. Thus, the objective of this study was to identify lymphocyte and macrophage populations in bovine placenta using specific markers and flow cytometry. This study analyzed samples of placentomes and intercaruncular regions of cows in the three quarters of pregnancy. In the placentomes, during the first quarter of pregnancy, mean percentage of labeled CD3+ cells was 2.34%; CD8+, 1.28%; CD14+, 1.66%; and CD335+, 0.96%. For the intercaruncular region, percentage of CD3+ cells was 0.71%; CD8+, 1.63%; CD14+, 2.81%; and CD335+, 2.81%. In the second quarter, placentomes showed 0.94% CD3+ cells; 0.77% CD8+; 0.72% CD14+; and 0.51% CD335+. In the intercaruncular region, percentage of labeled CD3+ cells was 0.50%; CD8+, 1.81%; CD14+, 2.64%; and CD335+, 0.51%. In the third quarter, placentomes showed labeling of 0.88% CD3+; 0.66% CD8+; 1.06% CD14+; and 0.74% CD335+ cells. In the intercaruncular region, percentage of labeled CD3+ cells was 0.19%; CD8+, 2.23%; CD14+, 2.43%; and CD335+, 0.16%. The results showed that there was a greater immunomarking of leukocytes CD3+ and CD335+ in the placentome when compared to the intercaruncular region during the third trimester. It can be concluded that leukocytes populations in bovine placenta is reduced, probably because of the syndesmochorial characteristic of bovine placenta. This represents a significant barrier for the immunological system of the mother, sharply decreasing the exposure of the conceptus to the mother’s immune system.

This study was undertaken to investigate the ameliorative effect of sodium selenite and nano-selenium on chicks exposed to heat stress. Ninety one day-old broiler chicks were divided into six groups each of 15 birds. Birds in group one and four were fed on basal diet and served as a control group, while birds in group two and five were fed on basal diet treated with 0.15 mg/Kg diet prepared selenium nanoparticles and birds in group three and six were fed on basal diet treated with 0.15 mg/Kg sodium selenite. On day 37 of experiment birds in 4th, 5th and 6th group were exposed to artificial heat stress at 40°C for 12 hours. Based on the results of total body weight gain and FCR lower parameters of body weight, body weight gain, feed conversion rate and higher feed intake ratio were observed in heat stressed chicks. Nano-selenium and Na selenite supplementation decreased the serum AST, ALT, urea, creatinine and MDA while the serum total protein, SOD, LZM, IgG, IgM and C3 were increased, also nano-selenium has ameliorative effect on hematological and histopathological findings. In conclusion, both nano-selenium and sodium selenite have an essential role in treatment of adverse effect of heat stress on chicks, with superiority of nano-selenium in improving anti- oxidative status and boosting immune response.

As animal welfare issue becomes important, the European Union bans conventional cages for laying hens from 2012. So the alternative housing systems like floor pens, aviaries or free range systems have been suggested. From 2011 to 2014, we monitored 20 welfare-oriented laying hen farms in South Korea to figure out serological status of major viral diseases. During this period, total 3,219 blood samples were collected from the randomly selected chickens to test and evaluate the hemagglutination inhibition titers for low pathogenic avian influenza, Newcastle disease and egg drop syndrome' 76. A total of 2,926 blood samples were tested through enzyme linked immunosorbent assay (ELISA) to assess the serological status of infectious bronchitis (IB). The distribution of ELISA titers for IB was various from almost 0 to 20,000 through the all weeks of age. Also, the antibody coefficient of variation for most of the diseases in this study was higher than those of typical cage layers. As this study was the first surveillance for major avian viral diseases of the animal welfare-oriented farms in South Korea, the results obtained from this study will help to determine what information and resources are needed to maintain better biosecurity and to improve the health and welfare of laying hen flocks.

Neurofascin, one of the members of L1CAM, has been known to have some important roles during the development of nerve fibers. In order to investigate the role of neurofascin associated with the formation of Schmidt-Lantermann incisure in the sciatic nerve, the localization of neurofascin was studied with electron microscopy, immuno-fluorescence and immuno-electron microscopy. In the electron microscopy, the first formation of Schmidt-Lantermann incisure was checked at postnatal day 6 and the complete form of incisures traversing the whole myelin sheath began to be observed at postnatal day 8. In the immunofluorescence, neurofascin immunoreactive Schmidt-Lantermann incisures were first checked at postnatal day 6 and dramatically increased with aging by postnatal day 56. In the immunoelectron microscopy, neurofascin immunoreactive gold particles at the incisure forming sites were first observed at postnatal day 6 and the number of gold particles was increased as the animal was getting old by postnatal day 56. According to the present study, neurofascin is likely to have some relationships with Schmidt-Lantermann incisure formation.

Objective-To describe the prevalence of antibodies against Salmonella spp in swine marketed in Iowa. Animals-Swine marketed by 1,044 low-volume producers and 45 high-volume producers. Procedure-Samples of diaphragm muscle collected from swine carcasses were tested by an indirect ELISA based on lipopolysaccharides from Salmonella spp, in particular Salmonella serovar Typhimurium. Prevalence of positive results for antibodies against Salmonella spp for carcasses, lots, and swine for each producer was determined. Producer-level seroprevalence was used to classify swine from producers as having negligible, low, moderate, or widespread evidence of previous or historical exposure to Salmonella spp. Results-From low-volume producers, 23,609 of 25,478 (92.7%; 95% confidence interval CI, 92.4% to 92.9%) samples had negative results, and 1,863 (7.3%; 95% CI, 7.05% to 7.56%) had antibodies against Salmonella spp. Of the 6,299 lots of swine tested, 1,191 (18.9%) contained at least 1 sample with positive results. From high-volume producers, 203 of 2,486 (8.1 %; 95% CI, 6.8% to 9.3%) samples had antibodies against Salmonella spp, and 124 of 629 lots had at least 1 sample with positive results for antibodies against Salmonella spp. Conclusions and Clinical Relevance-Less than 10% of pigs marketed in Iowa are apparently exposed to Salmonella spp. Most swine marketed by low-volume producers had negligible or little evidence of exposure to Salmonella spp, whereas a higher percentage of swine marketed by high-volume producers had positive results when tested to detect antibodies against Salmonella spp.

Objective--To evaluate the immunologic response of a killed tachyzoite vaccine against Neospora caninum and its effectiveness in preventing fetal loss associated with experimentally induced neosporosis in sheep. Animals--30 Dorset ewes. Procedure--Ewes were randomly allocated to receive vaccination on days 1 and 60 of the study with a killed N caninum tachyzoite preparation in a commercially available adjuvant or a saline-adjuvant mixture. A ram was placed on pasture with the ewes from days 15 to 60. Blood was collected from ewes before primary and booster vaccinations and prior to experimental challenge with N caninum tachyzoite performed on day 90; sera were assessed via Neospora agglutination (NA) and immunofluorescence antibody (IFA) assays. Blood was collected from lambs before they suckled, and sera were tested for antibodies against N caninum. Results--Of the 14 vaccinated ewes that became pregnant, 12 gave birth to live-born lambs; in contrast, 5 of 11 pregnant control ewes gave birth to live-born lambs. Whereas vaccination improved fetal survival in pregnant ewes challenged with N caninum tachyzoites, it did not appear to have any appreciable effect on transmission of N caninum to offspring, as indicated by results of NA and IFA assays. Conclusions and Clinical Relevance--The N caninum tachyzoite vaccine used in this study appeared to provide protection against fetal loss associated with experimentally induced neosporosis in a high proportion of pregnant ewes.

The present study was carried out to investigate the prevalence of mycoplasmal pneumonia of slaughter pigs in Youngnam area during the period from 1995 to 1997. The prevalence and pathomorphology of gross lung lesions were studied from 682 slaughter pigs in 8 swine herds. Gross lesions of pneumonia were recorede in the lungs of 442(64.8%), from 367 out of them(83.0%) were diagnosed as mycoplasmal pneumonia. Microbiological examination was performed with 197 lungs with gross lesions of mycoplasmal pneumonia of slaughter pigs from 8 differentswine herds. M hyopneumoniae, P multocida, A pleuropneumoniae, Streptococcus spp, Corynebacterium spp, and H parasuis were detected in 24.4%, 48.2%, 2.5%, 11.2%, 3.6%, and 1.0% of the pneumonic lungs, respectively. A total of 48 strains of M hyopneumoniae was investigated for thier in vitro susceptibility to antibiotics. Among the drugs tested, lincomycin, oxytetracycline, tiamulin and tylosin showed the high activity in minimal inhibitory concentration(MIC) of 0.04-5 micro gram/ml while erythromycin showed low activity in MIC values(1.25~40micro gram/ml).

This investigation was carried out to study morphological and chronological aspects of the development of the Harderian gland in the Mongolian gerbil(Meriones unguiculatus). Male and female Mongolian gerbils were sacrificed on days 1, 3, 5, 10, 30 and 60 after birth and their Harderian glands were processed for light microscopic observation. The result obtained were summarized as follows; 1. In 1-day-old Mongolian gerbil, Harderian gland was well distinguished from other tissue structures. It was composed of several immature tubules, and these tubules were separated each other by undifferentiated mesenchymal connective tissues. 2. In 3-day and 5-day-old Mongolian gerbils, the arrangement of tubules in the gland was more condensed than that of 1-day-old Mongolian gerbil. The excretory ducts started to appear in the connective tissues located between lobes. 3. In-10-day-old Mongolian gerbil, small lipid vacuoles began to be found in the cytoplasm of the secretory cells of the Harderian gland. There were some mucus-secreting cells within the epithelium of the excretory duct found in the interlobar connective tissues. 4. In 30-day-old Mongolian gerbil, there was markdely increased number of the tubules in the glands. The epithelial cells of the tubules were typically columnar in shape. Most of the columnar epithelial cells contained many small lipid vacuoles, although a few cells contained large lipid vacuoles. 5. In 60-day-old Mongolian gerbil, the Harderian gland exhibited the typical structural characteristicsl of the adult gland. The mature glandular structrues were more distinct than those of 30-day-old animals.

A species-specific 760 base pair(bp) BamHI to EcoRI DNA fragment(fMG-2) of lipoprotein gene was isolated from a Mycoplasma gallisepticum (M gallisepticum) genomic library. Based on the DNA sequence data of fMG-2, a pair of 25bp primers was synthesized. When used in the polymerase chain reaction(PCR), 732bp DNA products were amplified from 6 standard strains and 10 field isolates of M gallisepticum, but not from 2 Mycoplasma synoviae and 7 other Mycoplasma species. The lower detection limit was 100fg of the genomic DNA. Identity of the PCR products was confirmed by comparison of patterns of restriction endonuclease analysis with AseI, DraI, EcoRV and SspI.

Ontogeny, distributions and relative frequencies of somatostatin-immunoreactive cells were investigated in the proventriculus of the chicken embryos with incubation periods. Samples were taken from 10 groups(10 days of incubation to hatching) and studied by immunogistochemical methods. The findings were as follows. Somatostatin-immunoreactive cells were observed from 12 days of incubation in the proventricular glands and after that increased with incubation periods. The first observation time of these cells in the epithelium were at 15 days of incubation in the basal portion but in 16 and 17 days of incubation, no immunoreactive cells were observed in the epithelium but after that a few immunoreactive cells were observed in the basal portion and gastric gland regions. The shpaes of these cells were spherical to spindle in the proventricular glands and spherical to round in the epithlium and gastric gland.
