Evaluation of the Effect of Vanadium-Zinc Complex on the Changes in Histological and Stereological Structure, Functional Enzymes, and Oxidative Stress Indices of Liver Tissue in Adult Male Rats with Type 2 Diabetes

BACKGROUND: Vanadium and zinc are identified as low-concentration elements in the body of living organisms with a wide range of activities. Their insulin-like activity, through regulating the metabolism of carbohydrates, lipids, and eliminating the secondary symptoms of the disease, clearly demonstrates the ability of these elements to improve diabetes.OBJECTIVES: The present study aimed to evaluate the effect of vanadium-zinc complex on the structural, functional, and oxidative stress changes in liver tissue in adult diabetic male rats.METHODS: Herein, we recruited 40 adult male rats with the same weight range. They were randomly divided into four groups of 10, namely control, diabetic, healthy mice receiving vanadium-zinc complex, diabetic mice receiving vanadium-zinc complex) and received this combination at the rate of 10 mg / kg once a day for 60 days by oral gavage. At the end of the course, following blood sampling, part of the liver tissue was removed from the body to measure oxidative stress and the rest for stereological and histological studies. The serum isolated from the animals was also used to measure liver tissue functional enzymes (aspartate aminotransferase, alkaline phosphatase, alanine aminotransferase).RESULTS: The results of this research revealed that vanadium-zinc complex has no side effects on liver tissue in most case, but can greatly prevent structural damage to liver tissue by lowering blood glucose levels in diabetic rats and improving oxidative stress.CONCLUSIONS: Vanadium-zinc complex can be utilized with a certain mechanism in order to control blood sugar and inhibit oxidative stress. It could be regarded as an appropriate approach to preventing liver damage following chronic hyperglycemia in diabetic patients.

BACKGROUND: Wheat sprout contains a high amount of antioxidants, vitamins (especially vitamin E), minerals and phytoestrogen compounds. Use of medicinal herbs in reducing heavy metal toxicities has increased worldwide. In recent years, negative effects of lead on the male reproductive system and sperm fertility parameters have been shown broadly. OBJECTIVES: This study investigated the effects of wheat sprout extract (WSE) and vitamin E on sperm parameters and testicular oxidative stress in rats exposed to lead acetate. METHODS: Thirty-five rats were divided randomly into seven groups: G1 (control group) received 1 ml/kg/day of normal saline, G2 received 20 mg/kg/day of lead acetate, G3 and G4 received 100 mg/kg/day and 200 mg/kg/day of WSE respectively, G5 and G6 received 100 mg/kg/day and 200 mg/kg/day of WSE respectively with 20 mg/kg/day of lead acetate, and G7 received 100 mg/kg/day of vitamin E with 20 mg/kg/day of lead acetate. After 35 days, rats were sacrificed and blood, sperm, liver and testicle tissue samples were collected for histomorphological and histochemical studies. RESULTS: Results showed that count, motility and viability of sperms increased following the administration of 200 mg/kg/day of WSE (p<0.01). Histomorphological studies showed a significant increase in tubular differentiation index (TDI), Repopulation index (RI), number of Sertoli cells, and epithelium of seminiferous tubules in groups receiving 200 mg/kg/day of WSE (p<0.001). CONCLUSIONS: Results of the current study show that dose dependent WSE significantly prevents testicular toxicity and oxidative stress effects of lead acetate.

BACKGROUND: Lead (Pb) induced oxidative stress is known to suppress growth performance in broiler chickens. The current study was carried out in an attempt to describe the specific underlying mechanisms of such phenomenon. OBJECTIVES: The objective of this study was to investigate the effect of Pb-induced oxidative stress on performance, antioxidant status and behavioral responses of broiler chicken. METHODS: Eighty day-old broiler chicks were randomly assigned to 2 dietary treatment groups of 4 pen replicates, namely i) basal diet containing no lead supplement (control) and ii) basal diet containing 200 mg Pb/kg of diet. RESULTS: The results showed that addition of lead decreased body weight gain (p<0.01) and feed conversion ratio (p<0.01). Also, consumption of contaminated diet significantly increased MDA and H/L Ratio and significantly decreased SOD and GPx activity (p<0.01). Moreover, addition of 200 mg/kg diet significantly increased sitting pasture and aggression behavior and decreased feeding behavior (p<0.01). CONCLUSIONS: Our data conclude that Pb-induced oxidative stress adversely suppressed feed conversion ratio and growth performance. The proposed underlying mechanism for such phenomenon is Pb-induced oxidative stress by impaired antioxidant status and feeding behavior decreased the growth performance.

BACKGROUND: Oxidative stress reflects an imbalance between the reactive oxygen species (ROS) and a biological system's ability to readily detoxify the oxidative agents. Disturbances in the normal redox state of cells can cause toxic effects through production of peroxides and free radicals that damage all components of the cell, including proteins, lipids, and DNA. ObjectiveS: The purpose of this study was to evaluate the oxidative status in calves within 3 days of birth. Methods: Twenty calves from Zagros dairy farm were selected. Blood samples were collected from the jugular vein at 0 (before administration of colostrum), 24, 48 and 72 hours after birth for measurement of serum levels of TBARS, Superoxide dismutase and catalase. Results: The results indicated higher levels of serum malondialdehyde concentration at birth that subsequently decreased at 24 and 48 hours after birth (p<0.05). There were lower levels of serum concentration of catalase at birth that followed by increasing in it at 24 and 48 hours after birth (p<0.05). Also, the concentration of superoxide dismutase was lower at the birth that subsequently increased at 24 hours after birth (p<0.05). Conclusions: Based on this study, it seems that antioxidant capacity of calves has been increased within 3 days of birth. Improvement of oxidative status in calves could be due to gradual increasing of their antioxidative capacity and also beneficial effects of colostrum.

BACKGROUND: Ischemia-reperfusion injury (IRI) can induce major changes in the function of different organs, including the liver. Studies have indicated that ischemic post-conditioning (HIPO) can protect the tissues against ischemia-reperfusion injury.OBJECTIVES: To investigate the antioxidant and anti-inflammatory effects of ischemia post-conditioning against the IRI of the rat liver through four 30-second cycles of alternating ischemia and reperfusion, before 24-hour persistent reperfusion.METHODS: Fifteen rats were randomly divided into 3 groups 1) operation control group, 2) ischemia-reperfusion (IR) group whose liver was exposed to 60-miute ischemia of by 24-hour reperfusion and 3) ischemic post-conditioning (IR+IPO) group underwent the same procedure as the second group except that before persistent reperfusion, the rats were subject to post-conditioning by four 30-second cycles of alternating ischemia and reperfusion. The changes induced by IRI and the antioxidant and anti-inflammatory effects of ischemic post-conditioning were assessed by the serum level of IL-6 using the ELISA method, malondialdehyde (MDA), and total antioxidant capacity (TAC). RESULTS: Ischemic post-conditioning potentiated antioxidant effects and reduced the inflammation caused by the IR in the liver. The serum level of IL-6 reduced from 394.4±126.4 to 124.4±29.07 pg/ml (post-conditioning group), and the tissue MDA reduced from 431.4±76.53 to 207.2±25.77 nmol/g) compared to the IR group. The data revealed that the levels of the indices returned almost to the level of the operation control group (P<0.001). Additionally, the total antioxidant capacity of the liver significantly improved (P<0.01) from 11.58±1.87 (in the IR group) to 17.53±2.51 mmol/mg (in the IR+IPO group).CONCLUSIONS: The study demonstrates that the protective effect of ischemic post-conditioning against IR-induced injury may be mediated through decreasing inflammation and improving antioxidant activities.

Dairy cows may infrequently give milk tinged with blood after calving, which is a condition termed haemolactia. Economic losses for dairy farmers are caused by cases of haemolactia because of the condemnation of such milk, potential contamination of good bulk tank milk with haemolactic milk, and need for veterinarian intervention. This study was performed to elucidate the oxidative status of dairy cows with haemolactia during the peripartum period.

Ochratoxin A (OTA) is a mycotoxin notably produced by Aspergillus and Penicillium spp. Bacillus subtilis fermentation extract (BSFE) contains specific enzymes which hydrolyse OTA. This study evaluated the efficiency of BSFE in ameliorating the immunotoxic and nephrotoxic effects of OTA in broiler chickens.

Canine brachycephalic obstructive airway syndrome (BOAS) is a conformation-related respiratory disorder of dog breeds having congenitally flattened facial and skull anatomy. The aim of the study was to determine oxidative stress parameters, the lipid peroxidation product malondialdehyde, and antioxidant enzymes superoxide dismutase (SOD) and glutathione peroxidase in BOAS patients before and after surgical treatment and in healthy brachycephalic dogs. Nine healthy brachycephalic dogs that had not undergone surgery and 39 BOAS patients were included in the study. The BOAS patients were classified as grade 1 (5/34), grade 2 (16/34), and grade 3 (13/34) based on the decrease in the radius of the airway in the larynx. In BOAS patients, oxidative stress parameters were determined before and two weeks after surgery, while in control dogs, blood samples were collected only on inclusion to the study. All BOAS patients showed various degrees of improvement in clinical signs after surgery. Significantly lower (P < 0.05) SOD activity was found in grade 2 and 3 BOAS patients than in grade 1 patients. Two weeks after surgery, a significant (P < 0.05) increase in SOD activity in grade 2 and 3 patients was observed. Antioxidant enzyme SOD may play an important role in BOAS and can be used as a biomarker of antioxidant status assessment in BOAS patients.

The aim of the study was to demonstrate a link between uncomplicated Babesia canis infection in dogs and blood concentrations of zinc and copper and erythrocytic antioxidant defence – activities of glutathione (GSH), superoxide dismutase (SOD) and catalase (CAT). The study was based on 15 naturally occurring cases of canine babesiosis with anorexia, pyrexia, depression, pale mucous membrane, splenomegaly and dark red urine. Microscopic examination of Giemsa-stained peripheral blood smears and the results of PCR confirmed B. canis infection. Seven apparently healthy dogs brought in for either a check-up or vaccination were used for comparison. The levels of the erythrocytic antioxidant enzymes - SOD and CAT - were significantly higher in the infected dogs than in cytologically negative dogs. The levels of blood micronutrients were significantly lower in the infected dogs (0.478 μg of zinc per mL vs 1.241 μg/mL and 0.722 μg of copper per mL vs 1.392 μg/mL). Oxidative stress can be posited as one of the mechanisms leading to anaemia in dogs with babesiosis, and therefore antioxidant biomarker and copper and zinc concentrations could be used as indicators of disease severity and prognostic markers.

The study measured the hormonal and protein markers of acute stress, those of oxidative stress and total antioxidant capacity (TAC) in swine oral fluid, determined which of these parameters would be the most appropriate for future livestock welfare assessment and established the time when the samples should be taken. Stress was induced in 7 out of 14 castrated six-week-old Danbred×Duroc pigs by immobilisation on a nasal snare at 8 a.m., 1 p.m., and 6 p.m. and samples were taken both directly after the stressor was applied and 30 min later. The remaining pigs were the control group, which were not immobilised; their samples were taken at the same times. The concentrations of hormones and malondialdehyde (MDA) were measured using liquid chromatography with tandem mass spectrometry, while those of alpha-amylase and TAC were measured using spectrophotometry. The levels of cortisol and cortisone increased with statistical significance immediately after the acute stress response and 30 min later. A cut-off value set at 0.25 ng/mL cortisol concentration was capable of distinguishing between the stressed and control groups with 100% accuracy in evening samples and 95% accuracy overall. Prednisolone was not present, and the levels of testosterone and corticosterone were low and not distinctive. Alpha-amylase became significantly more concentrated during stress induction and 30 min later. The TAC and MDA levels rose after the stress but without statistical significance. The most suitable markers of acute stress were cortisol, cortisone and alpha-amylase. Oral fluid is a reliable material for monitoring the level of pigs’ stress and should be collected in the evening.
