Detection of autoantibodies against thyroid peroxidase in serum samples of hypothyroid dogs

Objective-To establish a sensitive test for the detection of autoantibodies against thyroid peroxidase (TPO) in canine serum samples. Sample Population-365 serum samples from dogs with hypothyroidism as determined on the basis of serum concentrations of total and free triiodothyronine (T3), total and free thyroxine (T4), and thyroid-stimulating hormone, of which 195 (53%) had positive results for at least 1 of 3 thyroid autoantibodies (against thyroglobulin Tg, T4, or T3) and serum samples from 28 healthy dogs (control samples). Procedure-TPO was purified from canine thyroid glands by extraction with detergents, ultracentrifugation, and precipitation with ammonium sulfate. Screening for anti-TPO autoantibodies in canine sera was performed by use of an immunoblot assay. Thyroid extract containing TPO was separated electrophoretically, blotted, and probed with canine sera. Alkaline phosphatase-conjugated rabbit anti-dog IgG was used for detection of bound antibodies. Results-TPO bands were observed at 110, 100, and 40 kd. Anti-TPO autoantibodies against the 40-kd fragment were detected in 33 (17%) sera of dogs with positive results for anti-Tg, anti-T4, or anti-T3 autoantibodies but not in sera of hypothyroid dogs without these autoantibodies or in sera of healthy dogs. Conclusions and Clinical Relevance-The immunoblot assay was a sensitive and specific method for the detection of autoantibodies because it also provided information about the antigen. Anti-TPO autoantibodies were clearly detected in a fraction of hypothyroid dogs. The value of anti-TPO autoantibodies for use in early diagnosis of animals with thyroid gland diseases should be evaluated in additional studies.

Results on determination of residual quantities of fenbendazole and its metabolites in cattle meat, organs and tissues after administration of preparation Febolvet in dose of one bolus per one animal realized in the conditions of the Republic of Belarus were presented. Animals were killed in 35 and 80 days after administration of preparation. There were no strangulates in animals organs. Pieces of nephros, liver, cordis, fat and muscle tissue were chosen for residual quantity of fenbendazole and its metabolites evaluation. The level of carcasses bleeding was good at application of boluses of infested activity with fenbendazole. In 24-48 hours of keeping in refrigerator the drying crust was well-defined, muscle tissue in cut had firm texture and from light red to red color, cut areas were wet. Tendon was form and elastic. Soup was clear, flavored and without extraneous odors. Study of physical-chemical indexes estimated peroxidase activity, pH, presence of products of primary breaking of protein in 24 and 72 hours after killing. Biological value decreased slightly (on 1,04%). Veterinary-sanitary evaluation of cattle meat was conducted in the investigation. Research results showed no residual quantities of fenbendazole in the analyzed samples after application of long lasting boluses. Application of the analyzed boluses in case of cattle strongylatosis did not lower the veterinary and sanitary indexes of meat and did not render substantial effect on its quality

Realization of veterinary and sanitary examination of meat and slaughter products of calves treated with hyperimmune serum against cattle colibacillosis was conducted in the conditions of the Republic of Belarus. There were formed two groups calves of 3-5 days old. Calves of the first group were treated with serum biological preparation of trial series produced at Vitebsk Biofabrika (Belarus). Calves of the second group were intact animals. Calves of each group were killed in 14 days after preparation application for realization of veterinary-sanitary examination. Slaughter meat was studied in accordance with the following indexes: pH; peroxidase activity; presence of products of primary protein breakdown; moisture content; relative biological value of meat; bacterial examination. The research results of organoleptic indicators showed that meat of both groups corresponded to the standards. Hyperimmune serum against colibacillosis of live-stock animals did not render negative influence on organoleptic, physical and chemical indexes of meat. Indexes of the relative biological value in meat of calves treated with the examined serum were higher than the similar indexes of intact animals. Meat of all examined animals did not have any germs of toxico-infections and toxicoses
