Serodiagnosis of porcine reproductive and respiratory syndrome virus infection with the use of glycoprotein 5 antigens

Glycoprotein 5 (GP5) of porcine reproductive and respiratory syndrome virus (PRRSV) has been studied extensively as a target for vaccine development. This study evaluated the serodiagnostic application of PRRSV GP5 by enzyme-linked immunosorbent assay (ELISA). Two immunodominant peptides (VR #1 and VR #2) and two neutralizing ectodomain-containing peptides (Ecto #1 and Ecto #2), as well as recombinant GP5 (rGP5) as a control, were prepared. Serum from unvaccinated pigs was screened for the antibodies that bind to these peptide and protein antigens. The results were compared with those from a commercially available diagnostic ELISA kit (HerdChek), which uses the nucleocapsid (N) protein as an antigen. Only VR #1+#2 showed a result statistically similar to that of N protein. Ecto #1 and Ecto #2 had a lower sensitivity than VR #1+#2 and rGP5. The peptides and rGP5 showed significant associations with the N protein (P < 0.05 or 0.01), which suggests that GP5 may also be a candidate serodiagnostic antigen. Since antibodies against GP5 persist much longer than those against the N protein, GP5 itself and some of its fragments are thought to be good targets for serodiagnosis. In addition, the presence of antibodies against the PRRSV structural antigens showed significant antigen-dependent differences.

The objective of this study was to determine the association between space allowance and in-transit loss of finishing pigs going to select abattoirs in Ontario during summer weather conditions. The study included data from 2- or 3-tiered trailers transporting >or= 130 pigs in June, July, and August 2003 to 3 packers that processed 76% of Ontario market pigs. Daily in-transit loss data were merged with packer data to determine the number of pigs on each trailer. Space allowance (in square meters per pig) was estimated from the percentage of each trailer’s capacity that was filled by the load size. Actual pig weights were not available. Hourly temperature and relative humidity were obtained from 2 local Ontario weather stations. In-transit loss increased with environmental temperature, by 6.6 times at temperatures between 28°C and 34.2°C compared with < 17°C. At space allowances between 0.44 and 0.43 m2/pig compared with >or= 0.515 m2/pig, in-transit losses increased 2.12 times when environmental temperatures were < 21°C. Temperature is likely a more important determinant of in-transit loss than space allowance. However, in-transit losses in hot weather are likely to be reduced by increasing space allowance or by adding a cooling device.

A clinical field trial was performed to determine the effectiveness of an autogenous Salmonella Typhimurium bacterin compared with a commercial live S. Choleraesuis vaccine in pigs. The association between Salmonella shedding and weight gain was also investigated. Nine cohorts of weaned pigs, (330 to 350 pigs per cohort), were randomly assigned to 1 of 3 treatment groups (injection with S. Typhimurium bacterin, vaccination via water with S. Choleraesuis vaccine, or a control group receiving no vaccine). In each cohort, the average daily gain was calculated for a selected pen throughout the production stage. Pen (pooled) fecal samples were collected bi-weekly and cultured. The odds of Salmonella shedding in both vaccinated groups was higher than in the control group (P < 0.05). The prevalence of Salmonella shedding declined overall as pigs aged (P = 0.04). However, the control pigs showed the smallest decrease in Salmonella shedding over the entire production stage, while prevalence of Salmonella shedding in the vaccinated groups decreased twice as much as the control group over the entire production stage. Salmonella Typhimurium var. Copenhagen DT104, S. Cerro, and S. Agona, which had been isolated on the study farm previously, were recovered from pigs in this study. Shedding of S. Typhimurium var. Copenhagen decreased over time in both vaccine treatment groups. On the other hand, S. Cerro shedding rate was lower in the control pigs compared with vaccinated pigs and S. Agona could be recovered only from the samples collected from S. Choleraesuis vaccinated pigs. The pigs from pens with a higher Salmonella recovery rate experienced slower growth compared with pigs from pens where Salmonella was not isolated. This latter finding indicates that there might be an economic incentive for producers to try to control endemic salmonellosis if effective programs could be developed.

The objective of this study was to determine the capsular serotypes and potential virulence factors of Streptococcus suis isolated from pigs with polyserositis. Among the 24 isolates evaluated, serotype 3 [7 (29%) of the isolates] and serotype 4 [5 (21%)] were the most common. The isolates were also studied for the presence of the genes mrp, epf, and sly, which encode muramidase-released protein (MRP), extracellular factor (EF), and suilysin (SLY), respectively. Of the 24 isolates, 8 carried mrp: 4 of serotype 3, 2 of serotype 2, and 2 of serotype 4. One mrp+ isolate (serotype 2) also carried the epf gene. All 24 isolates carried the sly gene. The serotype and genotype distribution greatly differed from that reported for isolates from pigs with other clinical manifestations of S. suis infection in other countries.

Evaluation of the myelography was studied in miniature pigs. Radiographs and computed tomographic (CT) images of the whole spine were obtained at clinically healthy twelve miniature pigs of 4 (8.7-10 kg) and 12 (26-31 kg) months. The assessments of the spinal cord were made in accordance with the Pavlov's method and compared area ratio [at spinal cord (SC), vertebral canal (VC) and vertebral body (VB)]. The Pavlov's ratio in the cervical spine was significant larger than that of thoracolumbar in radiographic myelography. On CT myelography, the area of the spinal cord had a significant difference between the cervical and thoracolumbar spine. Among the cervical spine, the ratios of spinal cord and vertebral body (SC : VB), vertebral canal and vertebral body (VC : VB) were minimum at the level of 4th cervical spine in both ages, while maximum at the level of 6th cervical spine in both months. In case of lumbar spine, the ratios of spinal cord and vertebral body (SC : VB) were the largest at the level of 4th lumbar spine in 4 months and at the level of 3rd lumbar spine in 12 months. In addition, the ratio of spinal cord and vertebral body (SC : VB) of the cervical spinal cord was significant lower at 4 months but the lumbar spinal cord showed lower pattern at 12 months old miniature pigs.

Torque teno virus (TTV), a species of Anellovirus, is a non-enveloped single stranded DNA virus with a wide range of animal hosts. The incidence of TTV is quite ubiquitous throughout the world. A total of 235 serum samples obtained from 137 pigs and 98 cattle at slaughterhouses in Korea during April 2005 to May 2005 were tested by TTV-specific PCR as to monitor prevalence of TTV among swine and cattle. As a result, the prevalent rates of TTVs in pigs and cattle were 43.1% and 4.1%, respectively. It seems that TTV infection is quite prevalent in swine population.

To determine the prevalence of Escherichia (E.) coli O157 : H7 from pigs after the Hazard Analysis and Critical Control Point (HACCP) system has been applied to Korean swine farm since 2006, 291 fecal samples were tested between May and December in 2008. Four E. coli O157:non-H7 (1.4%) were isolated from 4 different non-HACCP-accredited farms and they didn't have virulent genes which can cause illness for human. Also, Clostridium (C.) perfringens, Salmonella spp. and E. coli enterotoxins were tested using multiplex PCR. The positive rate for these pathogens of non-HACCP-accredited farms was higher than that of HACCP-accredited farms, and especially in case of C. perfringens, E. coli enterotoxins LT and STa, it was statistically significant (p less than 0.05). Thus, the early implementation of the HACCP program is expected to greatly contribute to the safety of livestock products as well as food hygiene.

Practicing veterinarians play an important role in detecting the initial outbreak of disease in animal populations. A pilot study was conducted to determine the feasibility of a veterinary-based surveillance system for the Ontario swine industry. A total of 7 practitioners from 5 clinics agreed to submit information from July 1, 2007 to June 30, 2008. The surveillance program was evaluated in terms of timeliness, compliance, geographic coverage, and data quality. Our study showed that the veterinary-based surveillance system was acceptable to practitioners and produced useful data. The program obtained information from 25% of pig farms in Ontario during this time period. However, better communication with practitioners, more user-friendly recording systems that can be adapted to each clinic’s management system, active involvement of the clinics’ technical personnel, and the use of financial incentives may help to improve compliance and timeliness.

The spectrum of acaricides changes continuously and improves. At the same time, big attention is given to research of new, more effective preparations meeting modern requirements. Research on studying the efficiency of Akaribil preparation against swine sarcoptosis (Sarcoptidae) was realized in the conditions of swine breeding farm in the Republic of Belarus. The preparation possesses high insect-acaricidal activity. In the conditions of the preparation storage and repeated opening of a ware in the course of tests there were stated no changes in drug smell or color. The scheme of the preparation application is applied in industrial production technology. It possesses anti-inflammatory properties, antiseptic stimulating healing of skin damages, does not irritate integuments, renders weak irritating effect on mucous membranes of eyes. In recommended doses Akaribil does not cause any adverse effects and complicating disease. There were stated no contraindications to preparation application. High therapeutic efficiency against arachnoentomosis was proved. Application the preparation made it possible not only to reduce disease incidence and animal death, but also to facilitate the severity of scabietic diseases. As a result of realized research it was established that efficiency of Akaribil against swine sarcoptosis was 100%.

In the conditions of the Republic of Belarus there was realized a complex of studies of quality swine slaughter products against the background of combine applications of the albuminous-vitamin-mineral additive Immovit and an extract of hill-growing saltwort for the preventive maintenance and treatment of a toxic dystrophy of liver. Research results showed the analysed preparations did not render negative influence on quality and safety of produced meat products. Against the background of application of the albuminous-vitamin-mineral additive Immovit in combination with an extract of hill-growing saltwort there was stated the improvement of swine slaughter product quality. In products from the animals which were administrated with a complex of examined preparations there was stated an optimized moisture content, the increasing of a relative biological value of a product (101,3-103,8%), and according to the indexes of bacterial safety the experimental pork considerably surpassed the meat indexes from animals of control groups. At the same time, in slaughter animal products of control groups there were revealed coliform bacillus bacteria.
