Bioassay techniques and high-performance liquid chromatography for detection of oxytetracycline residues in tissues from calves.

Tissue specimens from muscle, liver, kidney, and injection sites were collected, and serum was obtained from 3 calves euthanatized on each of posttreatment days 5 and 22. Calves were treated with 6.7, 13.4, or 20 mg of oxytetracycline (OTC)/kg of body weight, IM, once daily for 3 days; these dosages are 1, 2, and 3 times the label dose, respectively. One control calf was euthanatized on each of posttreatment days 5 and 22. In treated male calves killed 2 days after the last injection, OTC residues were detected in all tissues and serum, using high-performance liquid chromatography. Tissues from all injection sites also were considered positive for antimicrobial residues, using swab test on premises (STOP), microbial inhibition test (MIT), and thin-layer chromatography-biautography (TLCB) test. Kidney tissues from a calf given 13.4 mg of OTC/kg and kidney and liver tissues from a calf given 20 mg of OTC/kg also were considered positive, using the MIT and TLCB. Results of the STOP only were considered positive for the liver and kidney of a calf given 20 mg of OTC/kg, but substitution of Saskatoon antibiotic medium-3 for the original medium (antibiotic medium-5) allowed the STOP to detect residues in these tissues from all treated calves. In female calves killed 19 days after the last injection, the STOP, MIT, and TLCB procedures revealed positive results for tissues from some injection sites, but revealed negative results for other tissues. High-performance liquid chromatographic analyses detected OTC in tissues from injection sites from all treated calves, in muscle and liver from a calf given 20 mg of OTC/kg, and in kidneys from calves given 13.4 or 20 mg of OTC/kg. The STOP, MIT, and TLCB procedures lacked the sensitivity of high-performance liquid chromatography for detection of OTC residues.

DNA hybridization assay using probes prepared from liver was carried out to identify species characterization of the domestic animals. Gel electrophoresis showed that the target DNA extracted from raw muscle were 1kb and uniform pattern while fragments size of heated muscle were irrgular. Hybridizatino was performed by adding 200ng/ml probe in hybridization solution and incubating for 12 hours at 68 degrees centigrade. To obtain good discrimination, applied washing buffer and washing step differently depending on the species. The probes of pig, horse and dog formed the specific hybrids with each target DNA respectively. Although cross reaction ws detected in cattle, goat and sheep but signal intensity among these species made the discrimination possible each other. Such pattern was the same in the cases of chicken, turkey and duck. The hybridization pattern of heated muscle was similar to that of raw muscle in general, but the signal intensity was inferior to that of raw muscle. Species identification between closely related animal species, hybridized using the target DNA of such closely related animal species as a blocking agent, remarkable increase of discrimination from the evident decrease of non specific reaction compared with the control group. In addition, in the admixture where certain meat was included in the beef, pork, chicken meat, we colud find whether any unjust meat was admixed or not. In this case, detection limit of certain meat in admixture was 1%.

Different selective culture media were evaluated for isolation of Aeromonas spp. from 120 poultry meat samples. The recovery ofAeromonas isolates was highest fromAmpicillin DextrinAgar (89.39%), followed by Aeromonas Starch DNAse agar (68.18 %) andAeromonas isolation media (18.18 %).

In the conditions of the Republic of Belarus there were presented the results of a veterinary and sanitary assessment and bacteriological investigation of sheep slaughter products, which showed meat serviceability for consumption after application of Triclafen suspension. Application Triclafen suspension did not render negative influence on an organism of animals. It was proved by organoleptic and laboratory research of meat of slaughter animals of experimental and control groups. The meat obtained from healthy sheep which were administrated the suspension in accordance with microbiological indicators proved to be of good quality and corresponded to the established norms (Sanitary regulations and standards 11 - 63 Republic of Belarus 98): it had the general bacterial content within admissible limits and did not contain agents of anthropozoonosts, alimentary toxicosis and toxinfections and it can be sold without any restrictions.

In the conditions of the Republic of Belarus there was realized a complex of studies of quality swine slaughter products against the background of combine applications of the albuminous-vitamin-mineral additive Immovit and an extract of hill-growing saltwort for the preventive maintenance and treatment of a toxic dystrophy of liver. Research results showed the analysed preparations did not render negative influence on quality and safety of produced meat products. Against the background of application of the albuminous-vitamin-mineral additive Immovit in combination with an extract of hill-growing saltwort there was stated the improvement of swine slaughter product quality. In products from the animals which were administrated with a complex of examined preparations there was stated an optimized moisture content, the increasing of a relative biological value of a product (101,3-103,8%), and according to the indexes of bacterial safety the experimental pork considerably surpassed the meat indexes from animals of control groups. At the same time, in slaughter animal products of control groups there were revealed coliform bacillus bacteria.

In the conditions of the Republic of Belarus there was studied the veterinary and sanitary quality and safety of poultry slaughter products after application of a natural biological corrector VitoLAD produced in Belarus on the basis of cultivation of nonpathogenic strain of fungi Fusarium sambuninum MKF-2001-3 used for restoration of dysfunction disorders. As a result of the realized bacteriological research there were marked no pathogenic and opportunistic organisms in all experimental samples of meat and internal organs. Indices of biological value of meat of three experimental and control group had no authentic differences. Displays of toxicity for infusorians were not marked. Poultry meat of investigated samples in control, and all three experimental groups (doses of 0,25 ml/head - the second experimental group, a dose of 0,5 ml/head - the third experimental group and a dose of 1 ml/head - the fourth experimental group) in diets of which there was added the natural biological corrector VitoLAD, proved to be good-quality and harmless. In accordance with organoleptic, physical and chemical, bacteriological indexes, as well as biological value and harmlessness did not concede to meat of the control group. The complex veterinary and sanitary estimation of bird carcasses of three experimental and control groups did not show any deviations from existing standards that made it possible to organize production realization without restriction.

Research results on usage of DNA-markers (RYRI, FABP, ESRF18/FUTI) and other factors for industrial pork production were presented. It was determined that DNA-technologies let conduct interbreed crossing on industrial scale and get diverse (fat or meat, with different fat content) products, as well as increase pork production in general. The best variants of crossing combinations were pigs of the Belarus large white breed with Belarus meat and Dyurok, and also the Belarus large white breed with Landras and Dyurok. Research results showed, for in course of the whole period of breeding the daily average weight gain of hybrids of the large white with the Belarus meat and breeds Landras was higher than with the Estonian bacon on 25 and 18 g, respectively. Fattening of stores was finished at the period of achievement of body weight by 100-102,5 kg. The slaughter yield of meat products by groups (without intestinal fat) did not essentially differ and varied within 69,9 - 71,1%. There were noted no considerable distinctions in weight of internals (heart, lungs, kidneys, spleen, liver). Thickness of the salted pork fat at hybrids with animals of the Belarus meat breed was 1-2 mm lower than in other. The average meat yield of diverting weir was 61,9%, fat weight - 27,9%, bone weight - 10,2%. The organoleptic evaluation of beef-infusion broth quality showed that indicators of habit, aroma and taste and nourishing were at high level and compose 4,67; 4,59; 4,55 and 4,71 points, respectively. Tenderness, juiciness and taste of boiled meat also were at level 4,69; 4,76 and 4,87 points, respectively; fried meat - 4,57; 4,68 and 4,66 points

There was carried out the veterinary and sanitary examination of meat of young pigs vaccinated against swine erysipelas by a vaccine of a pilot batch. The meat testing results from animals under investigation show that the used vaccines reveal no negative influence on organoleptic and certain physical-chemical characteristics of meat; and by relative biological value the meat of young pigs at the use of concentrated vaccine of Konev matrix and the depot vaccine against swine erysipelas somewhat surpasses the analogous parameter of meat taken from the intact animals. Meat from animals of all groups is free from food borne toxic infections agents and toxicosis which characterizes it as a quality and safe food product. In the tabular form some physical and chemical and biological indicators of meat from young pigs involved in experiences are presented. | Целью работы явилось проведение ветеринарно-санитарной экспертизы мяса молодняка свиней, вакцинированного против рожи свиней вакциной опытной серии. Полученные результаты исследований мяса от подопытных животных указывают на то, что применяемые вакцины не оказывают отрицательного влияния на его органолептические и некоторые физико-химические показатели, а по показателям относительной биологической ценности мясо молодняка свиней при использовании концентрированной вакцины из матрикса Конева и депонированной вакцины против рожи свиней несколько превосходит аналогичный показатель мяса от интактных животных. Мясо от животных всех групп не содержит возбудителей пищевых токсикоинфекций и токсикозов, что характеризует его как качественный и безопасный пищевой продукт. В табличной форме представлены некоторые физико-химические и биологические показатели мяса от молодняка свиней, задействованных в опытах.

There were studied the veterinary-sanitary indicators of sheep slaughter products in the conditions of application of microcelements in various forms. Studying of the influence chelate forms of microelements on quality of cattle-breeding production is spent in comparison with preparations in which microelements are in a kind of salts. As a model preparation, for studying of chelate influence on quality of cattle-breeding production used of the chelate zinc form - Tsinkovet. Results of research show, that use of chelate and inorganic forms of zinc does not reduce high quality of received production. Meat of sheep of trial groups on organoleptic, bacteriological and physical and chemical indicators, and also biological value and harmlessness is characterized as good-quality and corresponds to veterinary-sanitary requirements. In tables results of physical and chemical researchs of meat, its biological value and harmlessness are presented. | Целью исследований явилось изучение ветеринарно-санитарных показателей продуктов убоя овец при назначении им микроэлементов в различных формах. Изучение влияния хелатных форм микроэлементов на качество животноводческой продукции проведено в сравнении с препаратами, в которых микроэлементы находятся в виде солей. В качестве модельного препарата, для изучения влияния хелатов на качество животноводческой продукции использовали хелатную форму цинка - Цинковет. Результаты исследований показывают, что использование хелатных и неорганических форм цинка не снижает доброкачественности получаемой продукции. Мясо овец опытных групп по органолептическим, бактериологическим и физико-химическим показателям, а также биологической ценности и безвредности характеризуется как доброкачественное и соответствует ветеринарно-санитарным требованиям. В таблицах представлены результаты физико-химических исследований мяса, его биологическая ценность и безвредность.
