Sequence analysis of VP2 gene of infectious bursal disease virus field isolate and vaccine strains

The VP2 full gene of Korean infectious bursal disease virus (IBDV) strain, SH/92, three attenuated vaccine strains, Bur706, Bursine-2 and CEVAC strains, were amplified by reverse transcriptase-polymerase chain reaction and sequenced and compared with published PV2 gene sequences of IBDVs. The VP2 nucleotide sequence similarity between SH/92 and three vaccine stains was 95.6~96.5% whereas the nucleic acid similarity among three vaccine strains was 97.5~98.5%. The amino acid sequence similarity of VP2 of SH/92 compared with three vaccine strains was between 94.4 and 97.6% while the amino acid similarity among three vaccine strains was between 97.4 and 98.4%.

Infectious bursal disease virus (IBDV) is a member of the Avibirnavirus genus of the Birnaviridae family which genome consists of two segments (A and B) of double stranded RNA. Segment A gene of KNU08010 isolate, which was isolated from a 15-day-old chicken flock in 2008, was sequenced and compared with other IBDV isolates including SH/92 strain, the first Korean very virulent (vv) IBDV isolate. The amino acid sequences of segment A gene showed that KNU08010 had 99.2% homology with SH92 strain. KNU08010 isolate had specific amino acids A222, I242, I256, I294 and S299 which are highly conserved among vvIBDV strains. Phylogenetic analysis based on the nucleotide sequences of variable region of the VP2 gene of 18 IBDV strains revealed that KNU08010 was grouped with vvIBDVs and was closely related to Korean vvIBDVs isolated from wild birds.

The level of antibody by ELISA,immunsosuppressive effect baes on the response of birds to Newcastle disease vaccination and damage to bursa of Fabricius by IBD vaccination were studied. The efficacy of six different IBD vaccination schedules were studied using intermediate and intermediate plus strains of vaccines either alone or in combination. In vaccinated groups, the sero-conversion of the vaccine virus was noticed during fourth week, reaching to the peak between eight to twelve weeks of age in different groups. Afterwards, there was a gradual decrease in the titres, by the end of 20th week (maximum period tested). There was no significant difference in the titres of different treatment groups. However, all the groups showed titres above protective level during the entire period of study. There was significant difference in bursa body weight (B-BW) ratios of vaccinated groups in comparison with control group. Histopathological studies of bursal sections revealed depletion of lymphoid follicles, presence of cystic spaces, edema and hemorrhages. The birds vaccinated with hot strain of IBD vaccine showed metaplastic changes, presence of foam cells with pronounced interfollicular fibrosis. The bursal scores were maximum in the groups vaccinated with hot strains of IBD vaccines.

In the conditions of the Republic of Belarus there was realized a morphological study of bone marrow aspirates at different terms chicken of immunization against Gumboro disease which made it possible to analyze the state of immune system for the objective evaluation of immunological status. Research results showed that chicken immunization with a national liquid chick embryo viral vaccine produced on the basis of KMIehV-13 strain against Gumboro disease (infectious bursal disease) promoted the development of immunomorphological remodeling of bone marrow. Application of immunostimulants Alveozan and Nukleovit for poultry vaccination caused more expressed changes revealing in statistically reliable increasing of total number of myeloblastic cells in the majority of cases by means of increasing of immature cells, and then mature cells of granulocytic series, as well as quantity of lymphocytes and plasmacytes
