Chlamydiae – what’s new?

This paper provides an overview of the current knowledge of chlamydiae. These intracellular microorganisms belonging to the Chlamydiaceae family are widely distributed throughout the world. Constant development of culture-independent approaches for characterisation of microbial genomes enables new discoveries in the field of Chlamydia. The number of new taxa is continuously increasing as well as the range of hosts. New species and genotypes are constantly being discovered, particularly new avian and reptilian agents, which are discussed in this article. Interestingly, wild animals are the main hosts for new Chlamydia species including different species of bird, turtle and snake. The availability of next-generation sequencing opens up a new prospect for research and leads to deeper knowledge of these interesting microorganisms about which much is still to discover.

Antimicrobial resistance is a global health threat. It has been studied in humans and domestic animals, but there is a lack of data on wild animals. The objective of this study is the elucidation of its patterns in Staphylococcus spp. isolated from wild mammals of the Autonomous Community of Aragón (Spain). A total of 103 mammals (Artiodactyla, Carnivora, Chiroptera, Erinaceomorpha, and Lagomorpha) were studied. A recovery centre provided 32 and hunting 71. Nasal and faecal samples yielded 111 staphylococci, which were identified by matrix-assisted laser desorption/ionization–time of flight mass spectrometry. A susceptibility test to 11 antibiotics was carried out, and statistical analysis was performed. Some differences were detected in bacterial prevalence depending on how the mammal fed. Artiodactyla, mainly hunted, were predisposed to carry coagulase-positive staphylococci. The staphylococci species recovered were resistant to at least two classes of antibiotics, and were disseminated in all of the geographical areas studied. Resistant staphylococci are widely distributed in the wild mammals in the areas of the study, but the resistance quantified in them is lower than that to be expected if the use of antibiotics in farms had a direct influence on the wildlife and its environment. On the other hand, resistance to antibiotics restricted to human use was widely disseminated in various wild animal species.

Coronaviruses are extremely susceptible to genetic changes due to the characteristic features of the genome structure, life cycle and environmental pressure. Their remarkable variability means that they can infect many different species of animals and cause different disease symptoms. Moreover, in some situations, coronaviruses might be transmitted across species. Although they are commonly found in farm, companion and wild animals, causing clinical and sometimes serious signs resulting in significant economic losses, not all of them have been classified by the World Organization for Animal Health (OIE) as hazardous and included on the list of notifiable diseases. Currently, only three diseases caused by coronaviruses are on the OIE list of notifiable terrestrial and aquatic animal diseases. However, none of these three entails any administrative measures. The emergence of the SARS-CoV-2 infections that have caused the COVID-19 pandemic in humans has proved that the occurrence and variability of coronaviruses is highly underestimated in the animal reservoir and reminded us of the critical importance of the One Health approach. Therefore, domestic and wild animals should be intensively monitored, both to broaden our knowledge of the viruses circulating among them and to understand the mechanisms of the emergence of viruses of relevance to animal and human health.

Archival specimens of gastric mucosa of 10 raccoons (Procyon lotor), 9 porcupines (Erethizon dorsatum), 6 grey foxes (Urocyon cinereoargenteus), 6 bobcats (Lynx rufus), 4 skunks (Mephitis mephitis), and 3 black bears (Ursus americanus) were microscopically examined for evidence of Helicobacter-like organisms. Such organisms were seen in the specimens from the grey foxes and bobcats only. Histochemical stains (modified Steiner and carbol fuchsin methods) revealed long spiral organisms within lumina of gastric glands; however, neither gross nor microscopic lesions were observed. By electron microscopy (EM), the organisms were found to be free in the glandular lumina and were seen occasionally in the cytoplasm of gastric epithelial cells. Morphologically, 2 different phenotypes of spiral organisms were identified by EM. The organisms associated with bobcats appeared to be more tightly coiled than those seen in grey foxes. The presence of Helicobacter-like organisms in the gastric mucosa of grey foxes has not previously been described.

Ovine progressive pneumonia is caused by a lentivirus of known infectivity only for sheep and goats. Virus susceptibility of 11 other species of animals was examined. Species included cattle, chickens, deer, dogs, goats, hamsters, horses, mice, pigs, rabbits, and rats. Of these species, only goats and rabbits could be experimentally infected with the virus. The infection in rabbits was acute, and virus did not persist or induce antibody production as it does in sheep and goats. Sera obtained from several people working in close contact with the virus and from several wild species, with unknown exposure history, were tested for antibodies to viral antigens. All results were negative. Knowledge of the host range of this virus is important for scientific studies and for virus eradication programs.

A two-year old lioness (Panthero leo) was referred to our clinic with nasal and oral bleeding, vomiting and anemia. As being informed by the owner; we learned that the lion had a history of anorexia, nasal discharge, fatigue, dehydration and apathy just one month prior to admitting the clinic. At the laboratory examination Urea, Creatinine and RBC (red blood cell) were higher than normal. Coronavirus, Feline Leukemia Virus, Feline Immunodeficiency Virus (Rapid FIV/Ab FeLV Ag Test Kit, Bionote®) agents were checked with commercial immunochromatographic rapid test kit and found as negative. Heamobartonella sp was found by blood smear. Enrofloxacin (Baytril-K % 5, Bayer®) 10 mg/kg dose for 10 days once a day and vitamin K and supportive fluid were given for treatment. All clinical signs and abnormal blood values returned to normal levels after 17 days. This report indicated that Heamobartonella sp is important agent also for wild cats and all cats should be controlled for this to infection even if suffered from renal failure.

OBJECTIVE To develop a noninvasive biomarker-based detection system specific for Mycobacterium bovis for monitoring infection in wild animals. SAMPLE Serum samples from 8 experimentally infected yearling white-tailed deer (Odocoileus virginianus) and 3 age-matched control deer and from 393 Minnesota Department of Natural Resources hunter-harvested white-tailed deer in northwest Minnesota. PROCEDURES 8 yearling deer were inoculated with 2 × 10(8) CFUs of virulent M bovis strain 1315 (day 0), and sera were obtained on days 0, 19, 48, and 60; sera were obtained from 3 uninoculated control deer on those same days. Sera from these deer and 9 M bovis-positive hunter-harvested deer were tested for 3 Mycobacterium-specific biomarkers (MB1895c, MB2515c, and polyketide synthase 5) by use of an indirect ELISA. That same ELISA was used to test sera obtained from 384 exposed noninfected deer in northwest Minnesota from 2007 through 2010, concurrent with an outbreak of tuberculosis involving cattle and deer in that region. RESULTS ELISA results revealed that tuberculosis infection could be detected as early as 48 days after inoculation in experimentally infected deer. Results for 384 deer sera revealed that prevalence of tuberculosis decreased over the 4-year period. CONCLUSIONS AND CLINICAL RELEVANCE Results suggested that the prevalence of tuberculosis in Minnesota deer decreased after 2009 but tuberculosis may have persisted (as subclinical disease) at extremely low levels, as indicated by the presence of low concentrations of circulating biomarkers. Biomarker-based diagnostic tests may offer a specific approach for early identification of M bovis infection.

Six groups of 5 dogs each were fed dilutions of canine adenovirus-2, either as raw liquid or after insertion into cornmeal baits. By the fourth week after vaccination, 29 of the 30 dogs developed high titers of serum-neutralizing antibodies to the virus.

A survey of 95 wild rats which were captured from various locations was conducted to determine the diversity and distribution of ectoparasites and endoparasites infesting wild rat population around the city of Ipoh and Kuala Lumpur. The rodents captured were Rat tus norvegicus and post mortem was carried out immediately after capture, with skin and organs examined for parasite infection. Ectoparasites recovered were blood sucking louse (Polyplax spinulosa) and mites (Myocoptes musculinus). Endoparasites recovered were nematodes (Aspiculuris tetraptera, Trichuris sp., and two strongyles, one of which is Strongyloides sp.) And three intestinal protozoan parasites (Blastocystis sp., Trichomonas sp., and a coccidia). Low diversity of ecto- and endoparasites were observed infecting wild rat population caught in Ipoh as compared to Kuala Lumpur.

Nematode eggs in liver tissues of two wild rats were recorded atthe Regional Veterinary Laboratory, Kota Bharu from 2014 to 2015. A total of 15 (2014) and 48 (2015) wild rats were examined by the laboratory for routine screening of zoonotic pathogens such as Leptospira sp. and others. On histological examination of the haematoxylin and eosin (H&E) stained liver tissues, masses of parasitic nematode eggs were observed. The shell of the eggsis striated with shallow polar prominences at either end. Numerous mini-pores can be seen on the outer shell as well. The eggswere identified as Capillaria hepatica (C. hepatica) nematode eggs, which causes hepatic capillariasis in rodents and numerous other mammal species, including humans. The wild rats were alsoshown to harbour Salmonella enteritidis from the intestine, E. coli from the lung and liver but none had leptospirosis by PCR. The purpose of this report is to highlight a common nematode of wildrats that is Capillaria hepatica infection in wild rats in Kelantan diagnosed by the Kota Bharu Regional Laboratory, in orderto create the awareness on concurrent parasitic infections which may cause reduced immunity thereby creating higher risk for other zoonotic pathogens such as leptospirosis.
