The suitability of present and future climates for 30 Rhipicephalus species in Africa are predicted using a simple climate envelope model as well as a Division of Atmospheric Research Limited-Area Model (DARLAM). DARLAM's predictions are compared with the mean outcome from two global circulation models. East Africa and South Africa are considered the most vulnerable regions on the continent to climate-induced changes in tick distributions and tick-borne diseases. More than 50% of the species examined show potential range expansion and more than 70% of this range expansion is found in economically important tick species. More than 20% of the species experienced range shifts of between 50 and 100%. There is also an increase in tick species richness in the south-western regions of the sub-continent. Actual range alterations due to climate change may be even greater since factors like land degradation and human population increase have not been included in this modelling process. However, these predictions are also subject to the effect that climate change may have on the hosts of the ticks, particularly those that favour a restricted range of hosts. Where possible, the anticipated biological implications of the predicted changes are explored.

The length of the antibiotic withdrawal period after intramammary treatment was influenced by the milk yield of dairy goats during this trial. Shorter withdrawal periods were seen in relatively high yielding dairy goats (production above 1.5 ℓ per day) compared to low producers (less than 1.3 ℓ per day). High yielding goats treated with Curaclox LC (Norbrook [Pharmacia AH]) had a withdrawal period of 42 h, while low yielding goats, treated with the same product, had a withdrawal period of 74 h. The recommended withdrawal period for Curaclox LC for use in cattle is 72 h. Relatively high yielding goats treated with Rilexine 200 LC (Logos Agvet [Virbac]) had a significantly shorter withdrawal period (37 h) than that recommended for use in cattle (96 h). Low yielding goats treated with Spectrazol Milking Cow (Schering-Plough Animal Health) had a significantly longer (95 h) withdrawal period than that recommended for use in cattle (60 h). Withdrawal periods were also influenced by stage of lactation and parity. There was a moderate positive correlation between lactation number and withdrawal period, as measured by TRIS (R2 = 0.621), and a moderate negative correlation between stage of lactation and withdrawal period (R2 = -0.669). In Trials 1, 2 and 3 combined there was a moderate negative correlation between withdrawal period and volume (R2 = -0.511) and a strong positive correlation between withdrawal period and lactation number (R2 = 0.720). The differences in percentage milk fat, protein and lactose before, during and after treatment were not statistically significant except in Trial 3 (Curaclox LC and Rilexine 200 LC) where protein and lactose differed significantly. In Trial 2 (Spectrazol Milking Cow) milk fat percentages differed significantly between treatment and control groups as did protein percentages in Trial 3. These differences are however, not biologically meaningful.

Contagious bovine pleuropneumonia (CBPP), caused by Mycoplasma mycoides var. mycoides small colony (MmmSC), is one of the most important diseases of cattle in Africa. The role of innate or acquired cell mediated and humoral immunity in conferring protection against MmmSC infection has not yet been elucidated. On the other hand, the pathological lesions caused by the aetiological agent have been considered indicative of an immunopathological process. In this study ten naïve cattle were exposed to in-contact infection with animals infected by intubation with a strain of MmmSC. Clinical signs, antibody response, IFNg release and pathological changes at necropsy were analysed and compared with the events following in-contact infection of an equal number of animals kept under daily treatment with cyclosporine for the entire observation period of 84 days. Cyclosporine is a suppressor of the immune response related to the T-cell system. Under the conditions of the experiment, cyclosporine appeared to condition the pathogenesis of CBPP by delaying the events that follow infection, bringing further support to the possibility that the immune response may have an impact on the disease outcome.

A serological survey of bovine babesiosis and anaplasmosis was conducted in the foot-and-mouth disease buffer zone surrounding the Kruger National Park in South Africa between 2001 and 2003 to determine whether the withdrawal of government-subsidized dipping in certain regions had affected the seroprevalence of these tick-borne diseases. Seroprevalence of Anaplasma marginale and Babesia bovis increased during the study period. This increase was greater in Limpopo Province where farmers had to supply their own acaricide than in Mpumalanga Province where dipping materials were provided by the local Veterinary Services. The number of animals testing positive for B. bigemina decreased in both provinces during the study period, which was attributed to possible vector displacement rather than more effective tick control measures. Responses to a questionnaire on ticks and tick-borne diseases revealed local knowledge on the subject to be highly variable and sometimes incorrect.

The sensitivity of LAMP, PCR and microscopy to detect Theileria spp. and Trypanosoma congolense in field-derived bovine blood samples from Tanzania was evaluated and compared. No parasites were detected by microscopy. Furthermore, no bovine Theileria spp. were detected by LAMP and PCR from all the 24 samples collected from Arusha. Four and one out of 24 samples were positive for Theileria congolense infection by LAMP and PCR respectively while, 18 and nine out of 40 samples from Dar es Salaam were positive by LAMP and PCR for Theileria spp. Infection, respectively. Although all samples from Dar es Salaam were negative for Trypanosoma congolense infections by PCR, 12 out of 40 samples were LAMP positive. Whilst PCR is an established gene amplification method for the detection of Theileria and trypanosome parasites, this study introduces LAMP as an alternative molecular diagnostic tool that could be used in large-scale epidemiological surveys.

Tetrameres numida n. sp. from the proventriculus of Helmeted guinea fowls, Numida meleagris, in South Africa is described from eight male and four female specimens. The new species shares some characteristics with other Tetrameres species, but can be differentiated by a unique combination of characters. It bears two rows of cuticular spines extending over the whole length of the body and possesses two spicules. The left spicule measures 1 699-2 304 ?m and the right one 106-170 mm. Caudal spines are arranged in three ventral and three lateral pairs and the tail is 257-297 mm long. Diagnostic criteria of some of the previously described species of the genus Tetrameres from Africa and other parts of the world have been compiled from the literature and are included here.

Krimpsiekte, a chronic form of cardiac glycoside poisoning, is an important plant-induced intoxication of small stock in South Africa. It is caused by cumulative, neurotoxic bufadienolides, such as cotyledoside. A cotyledoside-bovine serum albumin conjugate was synthesized to immunize animals. The efficacy of the cotyledoside-conjugate in inducing an immunological response was ascertained in rabbits (n = 4) and sheep (n = 4) by determining cotyledoside antibody titres with an ELISA using cotyledoside-hen ovalbumin as antigen. The formation of anticotyledoside antibodies was induced in both rabbits and sheep following immunization with the cotyledoside-protein conjugate. Protection provided by the vaccine was demonstrated by challenging sheep (n = 4) with repeated, daily doses of cotyledoside (0.015 mg / kg) administered intravenously, commencing 45 days after the initial vaccination. One control animal died on Day 3 of the challenge period and the other was severely affected after administration of the third cotyledoside dose. The immunized ewes (n = 2) remained clinically unaffected and the challenge was suspended following six daily injections. Vaccination as a means of preventing krimpsiekte seems to be quite feasible and deserves further investigation.

Objective—To determine whether selection for the homozygous A136R171 genotype that confers resistance to classic scrapie infection negatively affects production traits in sheep. Animals—996 commercial lambs obtained from 2 flocks at separate locations across 3 consecutive years. Procedures—Genotyping at codon 136 and 171 was performed by use of commercially available testing or a single-nucleotide polymorphism assay. Carcass data were collected without knowledge of genotype approximately 24 hours after slaughter by an experienced grader. The model to analyze associations between prion protein (PRNP) genotype and production traits was based on genotype, breed, or both as fixed effects and days on feed as a covariate. Results—Average daily gain was significantly associated with only combined codons 136 and 171. In flock 1, weaning average daily gain was significantly greater in AA136 sheep than heterozygotes; the difference between QR171 and RR171 sheep, compared with QQ171 sheep, were not significant although QR171 and RR171 sheep had higher values. However, in flock 2, average daily gain was significantly greater in AV136 sheep than AA136 sheep and in QR171 sheep than QQ171 sheep. Conclusions and Clinical Relevance—Findings suggest there is an advantage for average daily gain in lambs with an arginine allele at codon 171, but there were no other genotype effects on production traits. Thus, selection for the resistant arginine allele at codon 171 to comply with USDA scrapie eradication guidelines should not be detrimental to lamb production in commercial flocks. Effects of codon 136 on average daily gain were ambiguous.

Thirty samples of Mozzarella cheese (15 made traditionally from raw milk in Tripoli city (Libya) and other 15 imported samples sold in markets related to different brand names) were examined bacteriologically for their total bacterial count, psychrophilic count, coliform count (MPN/g), presumptive Staphylococcus aureus count, as well as enterococci count. Higher counts were found in locally manufactured Mozzarella cheese. Salmonellae were absent in all examined samples for both types, while Escherichia coli were isolated from 3 samples (20%) of locally made samples. According to the suggested Libyan Standards of such samples, most of examined locally manufactured Mozzarella cheese samples were found unacceptable.

A total of 620 egg samples from different species (chickens, ducks and ostriches) and 1615 poultry samples (chickens, ducks, pigeons, quails, turkeys and ostriches) were examined for salmonella infection. 12 salmonella isolates were obtained from the egg samples (1.9%) and 67 isolates from poultry samples (4.1%). Salmonella isolates were serotyped into S. enteritidis (25 isolates), S. typhimurium (17 isolates), S. infantis (12 isolates), S. montivideo (7 isolates), 3 isolates for each of S. rubislaw and S. cerro , 2 isolates for each of S. virginia, S. agona, S. poona, and S. derby and 1 isolate for each of S. sandiago and S. kentucky. The incidence of isolation from different poultry species was discussed in details. Antibiogram of the isolated salmonellae against 10 different antibiotics revealed that norofloxacin, ciprofloxacin, cepheridin and gentamycin gave the highest activity against different salmonella isolates while amoxicillin, tetracycline, and nitrofurantoin showed the highest resistance rate. Pathogenicity of the isolated serovars was tested in chickens. All isolates were found pathogenic with various degree of virulence. SDSPAGE protein analysis for the salmonella isolated form different poultry species revealed 12 protein bands ranged from 22-289 kDa. The differences were insufficient for reliable differentiation between the isolates and accordingly, it could be used beside other molecular techniques in differentiation between the salmonella strains.
