Molecular characterisation of Mycobacterium bovis isolated from African buffaloes (Syncerus caffer) in Hluhluwe-iMfolozi Park in KwaZulu-Natal, South Africa

Bovine tuberculosis (BTB), a chronic disease of mammals caused by Mycobacterium bovis, is a threat to South African wildlife. It has been reported that African buffaloes (Syncerus caffer) are reservoir hosts of BTB in South African wildlife populations. This study reports on the molecular identification and typing of 31 M. bovis isolates collected between 1993 and 2008, mainly from buffaloes but also from two lions and a bush pig, in the Hluhluwe-iMfolozi Park (HiP) in KwaZulu-Natal. To study the dynamics of BTB in the buffalo populations, 28 M. bovis isolates from the HiP and epidemiologically related parks were characterised using regions of difference deletion analysis for species identification and spoligotyping, variable number of tandem repeats (VNTR), polymorphic G-C-rich sequences and IS6110 restriction fragment length polymorphism (RFLP) genotyping methods. At least three distinct M. bovis genotypes were found amongst HiP samples. The combination of VNTR typing (using a 16-loci panel) and IS6110 RFLP revealed the presence of three additional genetic profiles in individual buffaloes, demonstrating that the highest level of discrimination was achieved by these typing methods. One of the observed spoligotypes (SB0130) was dominant and represented 75% of isolates from buffaloes. A novel M. bovis spoligotype (SB1474), which is reported for the first time in this study, was observed in 14.3% of isolates from buffaloes. Based on the observed genetic relationships, the findings suggest independent introductions from at least three unrelated sources. This study investigated the prevalence, risk factors and species composition of ticks, fleas and lice infesting dogs and cats in and around Hawassa in southern Ethiopia. In total, 200 dogs and 100 cats were examined from November 2008 to April 2009. Of the dogs and cats examined, 99.5%and 91.5%, respectively, were infested with one or more species of ticks, fleas or lice. The overall prevalence was higher in dogs than in cats. A total of six different species of ectoparasites were collected and identified from dogs, but only three species from cats. Ctenocephalides felis was the predominant species amongst the animals, with a prevalence of 82.9% on dogs and 67% on cats. Other prevalent species on dogs included Ctenocephalides canis (73.8%), Heterodoxus spiniger (4%), nymphs of Amblyomma spp. (3.5%), Pulex irritans (2.5%) and Haemaphysalis leachi (0.5%). C. canis (18%) and P. irritans (6%) were also found on cats. More female than male fleas and lice were observed. The study revealed that the prevalence of fleas, ticks and lice on dogs was not significantly different between male and female animals or between young and adult dogs. However, the prevalence of these ectoparasites was significantly higher in female than in male and in adult than in young cats. The study showed that the prevalence of ectoparasites on both dogs and cats was significantly higher on animals with flea allergy dermatitis (FAD) than those without FAD, and on animals with lesions on their skin compared with those without lesions.

Two 4-day-old Thoroughbred foals with acute clinical signs of depression, elevated pulse and respiratory rates, anemia, and jaundice were referred to the equine hospital of Korea Racing Authority. The foals were seemingly normal at birth, but showed clinical signs such as abnormal behavior, jaundice and anemia suddenly after ingestion of the mare's colostrum, followed by death at 4 days after birth. According to the results of the hematology, serum chemistry analysis, jaundice foal agglutination test, and blood groups test, the foals were diagnosed as neonatal isoerythrolysis (NI) caused by the mare's antibodies. These results suggested that the pregnant mares have a potential for NI.

Eosinophilic granuloma is a common hypersensitive inflammatory skin disease in cats, and rare in dogs and horses. The skin biopsies of 5 years old female Cocker spaniel and 2 years old female mixed dog had the clinical signs of skin nodules with alopecia were submitted for diagnosis. Solitary skin nodules and papillary nodules were presented on the left external ear and back of Cocker spaniel and on the external ear of mixed dog, respectively. Histopathologically, epidermis of skin showed mild to severe hyperplasia with multifocal ulceration. Small to large irregular, brightly eosinophilic foci with degenerating eosinophils and homogeneous degenerated collagens were existed in the dermis of both ear and back skin. Typical 'flame figures', a mixture of degenerated collagen and degranulated eosinophils, were observed in both cases. Based on the histopathologic findings and special staining characters, 2 cases were diagnosed as canine eosinophilic granuloma. This is the first report for the eosinophilic granuloma of dogs in Korea.

This study surveyed the prevalence of traumatic reticular diseases (TRD) of slaughter cattle in Korea, the typology of the causative foreign bodies and the effects on beef quality and carcass weight. The overall prevalence of TRD in 3,121 slaughter cattle was 5.5%. However, the prevalence was significantly higher in Korea indigenous cattle Hanwoo (5.8%, p less than 0.05) and female cattle (20.75%, p less than 0.001). The prevalence significantly increased in aged cattle (p less than 0.001). Major lesions related to foreign bodies were reticulitis (96%) and peritonitis (86%). Most causative foreign bodies were made of iron including nails, wires, steel rods, screw nails, and syringe needles. Cattle affected with TRD produced significantly lower grade quality of beef compared to normal cattle (p less than 0.0001), but TRD did not affect carcass weight. The data will be useful in the management of TRD, with the aim of increasing beef productivity in Korea.

Bovine tuberculosis (bTB), caused primarily by Mycobacterium bovis, continues to exert an economic loss, even in countries with active control measures, and is one of zoonotic diseases enable to be transmitted to human. The control and eradication of bTB are mainly based on a test and slaughter policy and/or abattoir surveillance. Various factors including limitation of diagnostic tests have been considered as major constraints to eradication. Single intradermal test (SIT) is the official diagnostic test. New diagnostic methods are needed to be developed, because of limitations of the test. In the present study SIT was compared with single intradermal comparative cervical test (SICCT) and interferon (IFN)-γ assay. There was very low correlation between SIT and SICCT. However, high correlation was shown between SIT and IFN-γ assay while no correlation was observed between SICCT and IFN-γ assay. Therefore, our results suggest the possibility of replacement of SIT with IFN-γ assay for the diagnosis of bovine tuberculosis.

The study evaluated whether a transgenic carrot vaccine could induce a K88-specific immune response in sows and whether the resultant maternal antibody could protect piglets against enterotoxigenic Escherichia coli (ETEC) K88ac infection. Sows (n = 15) selected randomly from a farm in Korea were assigned to three groups (n = 5 per group: control [untreated]), group A (orally inoculated with a non-transgenic and transgenic carrot vaccines at 2 and 4 weeks ante partum, respectively), and group B (conventionally vaccinated according to the manufacturer’s instructions). After 7 days of lactation, 5 piglets selected randomly from each group were challenged with 1 × 10∨10 colony forming units/mL ETEC K88ac. Group C had the lowest mean fecal consistency score on post-challenge days 1 and 7. Histiologically, On post-challenge day 7, group C showed an increased duodenum and ileum villus:crypt ratio, compared to group A in the duodenum, with group B displaying the highest ratio. Groups B and C had more increased villus width than group A in the jejunum. Group C displayed the greatest increase in villus width in the ileum. The colostrums and serum from groups B and C displayed higher concentrations of IgA and IgG against ETEC K88, compared to group A. Based on the results, it was concluded that the transgenic carrot vaccine in sow per oral may have an effect on preventing piglet diarrhea as good as commercial recombinant vaccine.

Barium suspension, oral iodine contrast medium and water were applied in eight dogs to evaluate (1) distension of gastrointestinal tract, (2) the effect of the oral contrast media on the identification of the pancreas from surrounding organs, and (3) image quality and the presence of artifacts in canine pancreas computed tomography (CT) images. Oral iodine contrast medium, gastrografin, produced significant artifacts that deteriorated the CT images of the pancreas. The use of water did not provide the fullness of the gastrointestinal lumens. Barium suspension was effective for the identification of the pancreas from the surrounding gastrointestinal tract, without significantly increasing image noise. Barium suspension can be used as an optimal contrast medium that will not cause an adverse effect on the pancreatic density and image quality.

Malignant Sertoli cell tumor was diagnosed in a 5-year-old male Shih Tzu dog. Clinical features of the dog were anorexia, urinary incontinence, constipation, anemia, alopecia, and epistaxis. The dog also had unilateral cryptorchid testis in the abdomen. Several abdominal and thoracic masses were identified on radiography. Grossly, the cryptorchid testicular mass was markedly enlarged to 8 cm in diameter. On cut surface, firm and well demarcated milk-white neoplastic areas were irregularly separated by white fibrous bands. Histologically, the testicular mass was diagnosed as tubular pattern Sertoli cell tumor. In addition, abdominal and mediastinal lymph nodes metastasis were found. Immunohistochemically, the tumor cells were strongly positive for vimentin and neuron specific enolase, but negative for S-100 and cytokeratin.

Trimethyltin chloride (TMT) has been used as a neurotoxin for inducing brain dysfunction and neuronal death. Neuronal death in the hippocampus by TMT may generate excessive nitric oxide, but there are few studies about nitric oxide synthase enzyme involved in the synthesis of nitric oxide. The purpose of present study is to analyze the TMT toxicity in each region of rat hippocampus. To evaluate the involvement of nitric oxide, we analyzed the effects of aminoguanidine known as a selective inhibitor for inducible nitric oxide synthase on behavioral changes and the hippocampus of rat by TMT toxicity. 6-week-old male Sprague-Dawley rats were administered with a single dose of TMT (8 mg/kg b.w., i.p.) and the control group was similarly administered with distilled water. TMT + aminoguanidine-treated groups were administered with aminoguanidine (10 mg/kg or 100 mg/kg b.w., i.p.) for 3 days prior to TMT injection. The rats were sacrificed 2 days after TMT administration. In the TMT-treated group, a number of cell losses were seen in CA1, CA3 and the dentate gyrus. In the TMT + aminoguanidine-treated group, neuronal death was seen in CA1 and CA3, but reduced in the dentate gyrus compared to the TMT-treated group. Western blot analysis showed that cleaved caspase-3 expression was increased in the TMT-treated group compared to the control group. However, the expression significantly declined in the TMT + aminoguanidine-treated group. The present findings suggest that inducible nitric oxide synthase is involved in neuronal death induced by TMT.
